Nerve Growth Factor: A Multifaceted Regulator of Pain, Inflammation, and Tissue Remodeling in Bone and Joint Disorders.

As an essential member of the neurotrophic factor family, nerve growth factor (NGF) plays multifaceted regulatory roles in the pathological processes of bone and joint diseases through its dual signaling mechanisms involving high-affinity receptor TrkA and low-affinity receptor p75NTR. This review systematically examines the profound impact of NGF on bone and joint diseases in multiple aspects, such as bone remodeling, pain perception, and angiogenesis. First, NGF promotes mechanical stress-induced bone formation through the Wnt/β-catenin signaling pathway and regulates osteogenic differentiat

Larotrectinib in Adjuvant and First-Line Treatment for Metastatic NTRK Fusion Colon Cancer.

We report an 83-year-old woman with severe concomitant pathology who underwent surgery for colon cancer, with recurrent bleeding from the tumor, and subcompensated intestinal stenosis. After right-sided hemicolectomy with regional lymph node dissection, the disease was staged as pIIIC, and a translocation of the NTRK1 (Neurotrophic Tyrosine Receptor Kinase) gene was detected. The patient required adjuvant treatment, given the high risk of disease progression, but due to her severe concomitant pathology, chemotherapy was contraindicated. Adjuvant therapy with larotrectinib was recommended given

In-depth analysis of active compounds targeting tropomyosin-related kinase A via constructed lipid raft @capillary monolith affinity chromatography.

In order to enrich the selection of biological ligands, realize the miniaturization analysis, and broaden the application of monolith materials for active ingredients screening and separating, we sough to construct a lipid raft @capillary monolith microcolumn affinity chromatography model. Single factor experiments and various characterization methods, including scanning electron microscopy (SEM) and thermogravimetric analysis, were employed to investigate the polymerization of the monolith column under different material ratios to determine optimal preparation conditions. Subsequently, the li

A rare case of congenital insensitivity to pain with anhidrosis.

A 22-month-old girl of consanguineous parents was admitted with a high-grade fever. She was found to have insensitivity to painful stimuli and an absence of perspiration. She also displayed self-mutilating behaviour and was insensitive to cold/hot water on her body. On examination, there was loss of the tip of the tongue, missing teeth, generalised xerosis, and several ulcers at sites of minor trauma. She also had dysplastic nails and digital ulcers. Sensory examination demonstrated a complete lack of awareness of pain and temperature, vibration and fine touch were intact and lacrimation was n

The Alternative TrkAIII Splice Variant, a Targetable Oncogenic Participant in Human Cutaneous Malignant Melanoma.

Post-therapeutic relapse, poor survival rates and increasing incidence justify the search for novel therapeutic targets and strategies in cutaneous malignant melanoma (CMM). Within this context, a potential oncogenic role for TrkA in CMM is suggested by reports of NTRK1 amplification, enhanced TrkA expression and intracellular TrkA activation associated with poor prognosis. TrkA, however, exhibits tumour-suppressing properties in melanoma cell lines and has recently been reported not to be associated with CMM progression. To better understand these contradictions, we present the first analysis

Characterization of on-target adverse events caused by TRK inhibitor therapy.

BACKGROUND: The tropomyosin receptor kinase (TRK) pathway controls appetite, balance, and pain sensitivity. While these functions are reflected in the on-target adverse events (AEs) observed with TRK inhibition, these AEs remain under-recognized, and pain upon drug withdrawal has not previously been reported. As TRK inhibitors are approved by multiple regulatory agencies for TRK or ROS1 fusion-positive cancers, characterizing these AEs and corresponding management strategies is crucial. PATIENTS AND METHODS: Patients with advanced or unresectable solid tumors treated with a TRK inhibitor were

Phenotypic heterogeneity of intellectual disability in patients with congenital insensitivity to pain with anhidrosis: A case report and literature review.

Congenital insensitivity to pain with anhidrosis (CIPA) is a rare autosomal recessive heterogeneous disorder mainly caused by mutations in the neurotrophic tyrosine receptor kinase 1 gene ( NTRK1) and characterized by insensitivity to noxious stimuli, anhidrosis, and intellectual disability. We herein report the first north Han Chinese patient with CIPA who exhibited classic phenotypic features and severe intellectual disability caused by a homozygous c.851-33T>A mutation of NTRK1, resulting in aberrant splicing and an open reading frame shift. We reviewed the literature and performed in silic

Mimicking Nature, Modulating Vision: Peptidomimetic Neurotrophin Agonists and Emerging Regenerative Strategies in Ocular Disease.

Therapeutic strategies for ocular diseases are undergoing a transformative shift from symptom management to regenerative and disease-modifying approaches. This review highlights the development of neurotrophin receptor agonists-including recombinant nerve growth factor (NGF) (cenegermin), peptidomimetics (e.g., REC-0559, tavilermide), and synthetic microneurotrophins (BNN27, ENT-A010)-that target tropomyosin receptor kinases (TrkA/TrkB) and the p75 neurotrophin receptor (p75NTR) pathways to promote neuronal survival, synaptic plasticity, and tissue repair in neurotrophic keratitis, dry eye dis

Identification of Novel Positive Allosteric Modulators of Neurotrophin Receptors for the Treatment of Cognitive Dysfunction.

Alzheimer's disease (AD) is the most common neurodegenerative disorder and results in severe neurodegeneration and progressive cognitive decline. Neurotrophins are growth factors involved in the development and survival of neurons, but also in underlying mechanisms for memory formation such as hippocampal long-term potentiation. Our aim was to identify small molecules with stimulatory effects on the signaling of two neurotrophins, the nerve growth factor (NGF) and the brain derived neurotrophic factor (BDNF). To identify molecules that could potentiate neurotrophin signaling, 25,000 molecules

The role of neurotrophins in axonal growth, guidance, and regeneration.

Neurotrophins are proteins that regulate neuronal survival, axonal growth, synaptic plasticity and neurotransmission. They are members of the neurotrophic factors family and include factors such as the nerve growth factor (NGF), the brain derived neurotrophic factor (BDNF), the neurotrophin-3 (NT-3), and the neurotrophin-4/5 (NT-4/5). These molecules bind to two types of receptors: i) tyrosine kinase receptors (TrkA, TrkB, TrkC) and ii) a common neurotrophin receptor (p75NTR). The two receptor types can either suppress or enhance each other's actions. Neurotrophins have a multifunctional role

Retinoic acid restores adult hippocampal neurogenesis and reverses spatial memory deficit in vitamin A deprived rats.

A dysfunction of retinoid hippocampal signaling pathway has been involved in the appearance of affective and cognitive disorders. However, the underlying neurobiological mechanisms remain unknown. Hippocampal granule neurons are generated throughout life and are involved in emotion and memory. Here, we investigated the effects of vitamin A deficiency (VAD) on neurogenesis and memory and the ability of retinoic acid (RA) treatment to prevent VAD-induced impairments. Adult retinoid-deficient rats were generated by a vitamin A-free diet from weaning in order to allow a normal development. The eff

Adolescent alcohol exposure alters age-related progression of behavioral and neurotrophic dysfunction in the TgF344-AD model in a sex-specific manner.

Alzheimer's Disease (AD) and heavy alcohol use are widely prevalent and lead to brain pathology. Both alcohol-related brain damage (ABRD) and AD result in cholinergic dysfunction, reductions in hippocampal neurogenesis, and the emergence of hippocampal-dependent cognitive impairments. It is still unknown how ARBD caused during a critical developmental timepoint, such as adolescence, interacts with AD-related pathologies to accelerate disease progression later in life. The current study utilized a longitudinal design to characterize behavioral and pathological changes in a transgenic rat model

Nerve growth factor receptor (NGFR): A key player in glioblastoma progression.

Glioblastoma (GBM) is a highly invasive brain cancer arising from astrocytes, and current treatments are often ineffective, limiting patient five-year survival to <5%, despite maximal safe resection, temozolomide (TMZ) chemotherapy, and radiotherapy. Alterations in receptor tyrosine kinase (RTK) signaling are a hallmark of GBM progression and invasion. Numerous clinical trials have explored commonly altered RTKs, but none have offered a viable target that effectively increases patient outcomes and survival. This review synthesizes available literature on the role of the nerve growth factor rec

Discovery of a Novel Phenyl Thiophene-3-carboxamide Derivative DZX19 as an Orally TRK Inhibitor with Potent Antitumor Effects.

Tropomyosin receptor kinase (TRK) is an important therapeutic target for tumors driven by NTRK gene fusions. However, the clinically approved TRK inhibitors, including Larotrectinib and Entrectinib, are limited by insufficient efficacy and resistance due to kinase mutations. Here, we report DZX19 (C02), a novel phenyl thiophene-3-carboxamide TRK inhibitor developed via a pharmacophore-guided scaffold-hopping approach combined with structure-based design, based on the key binding features of Entrectinib. DZX19 demonstrated stronger in vitro activity and an improved resistance profile against En

Analgesic effects of nerve growth factor-directed monoclonal antibody on diabetic neuralgia in an animal model.

Current treatment options for diabetic neuralgia are limited and unsatisfactory. Tanezumab, a monoclonal antibody that blocks nerve growth factor (NGF) signaling, has been shown to be effective in relieving the clinical symptoms of osteoarthritis pain, chronic low back pain, cancer pain induced by bone metastasis, and diabetic neuralgia. However, the clinical development of tanezumab has been terminated due to the risk of induction of rapidly progressive osteoarthritis (RPOA), and no other NGF antibodies have been examined for their ability to treat diabetic neuralgia in either animal models o

Nerve growth factor inhibitor with novel-binding domain demonstrates nanomolar efficacy in both cell-based and cell-free assay systems.

Nerve growth factor (NGF), a member of the neurotrophin family, is known to regulate the development and survival of a select population of neurons through the binding and activation of the TrkA receptor. Elevated levels of NGF have been associated with painful pathologies such as diabetic neuropathy and fibromyalgia. However, completely inhibiting the NGF signal could hold significant side effects, such as those observed in a genetic condition called congenital insensitivity to pain and anhidrosis (CIPA). Previous methods of screening for NGF-inhibitors used labeling techniques which have the
