Severing enzymes nanodamage microtubules by extracting tubulin subunits. This extraction is accompanied by spontaneous repair with soluble tubulin. Here, we show that GTP-, and not GDP-tubulin, incorporates preferentially at nanodamage sites where it recruits end binding protein-1 (EB1). Using GTP hydrolysis-defective recombinant tubulin mutants we show that the tubulin GTPase is a timer for EB1 association at repair sites. INTRODUCTION: The immediate impact of exposure to severe wounds, dead bodies, and immediate threat to life has been understudied. Most studies focus on the acute stress disorder and/or post-traumatic stress disorder phases in order to assess rescue personnel's symptomatology, and tend to neglect the immediate exposure to elements of the disaster.

A relevant percentage of genetic diseases (~10%) are caused by premature termination codon (PTC) mutations, including cystic fibrosis (CF). CF patients carrying a nonsense mutation in the CFTR gene are insensitive to CFTR modulators and cannot benefit from the pharmacological therapy with Kaftrio®. The rescue of CFTR with PTCs faces two limiting steps: 1) the nonsense-mediated RNA decay (NMD) mechanism that, by degrading mRNAs carrying PTCs, strongly reduces CFTR transcript levels; 2) PTCs cause the arrest of protein synthesis, with the production of a truncated and nonfunctional CFTR protein.
