Genetic Rescue of X-Linked Retinoschisis Mouse (Rs1-/y) Retina Induces Quiescence of the Retinal Microglial Inflammatory State Following AAV8-RS1 Gene Transfer and Identifies Gene Networks Underlying Retinal Recovery.

To understand RS1 gene interaction networks in the X-linked retinoschisis (XLRS) mouse retina (Rs1-/y), we analyzed the transcriptome by RNA sequencing before and after in vivo expression of exogenous retinoschisin (RS1) gene delivered by AAV8. RS1 is a secreted cell adhesion protein that is critical for maintaining structural lamination and synaptic integrity of the neural retina. RS1 loss-of-function mutations cause XLRS disease in young boys and men, with splitting ("schisis") of retinal layers and synaptic dysfunction that cause progressive vision loss with age. Analysis of differential ge…

Retinal detachment associated with the splice site mutation c.53-1G>A in the RS1 gene: A case report and review of the literature.

Retinoschisin protein is mainly produced in the retina, with minimal expression in other tissues. It is located on the surface of the photoreceptor inner segments and bipolar cells. X-linked retinoschisis, caused by mutations in the RS1 gene, primarily presents as cavities in the retina and separation of the layers. We report a case of a 2-year-old male patient with notable clinical differences in both eyes. During follow-up, genetic testing confirmed an RS1 gene mutation causing retinal detachment and X-linked retinoschisis, with no family history of the disease. The clinical diagnosis of ret…

Tractional elevations of the retina in patients with diabetes.

Tractional retinoschisis and tractional retinal detachment are both complications of proliferative diabetic retinopathy. The two conditions are frequently confused because they are similar in diagnostic features. We determined the respective incidence of tractional retinoschisis and tractional retinal detachment in 200 eyes with tractional elevations of the retina in patients with diabetes. In 39 eyes, the diagnosis was unequivocally tractional retinoschisis because the retinal elevation maintained its concave contour despite the development of retinal holes. In 65 eyes, tractional retinal det…

Removal of internal limiting membrane for recurrent myopic traction maculopathy.

A 63-year-old man presented with a foveal detachment and retinoschisis associated with myopic traction maculopathy of his left eye with a refractive error of -12.0 dioptres. Both the retinoschisis and foveal detachment were initially successfully treated with vitreous surgery that was limited to the induction of a posterior vitreous detachment beyond the areas of retinoschisis over the posterior staphyloma. Three years later, the macula re-detached and further surgery was performed, which involved peeling the internal limiting membrane (ILM). Following the second vitreous surgery the macula re…

The Use of Ophthalmic Ultrasonography to Identify Retinal Injuries Associated With Abusive Head Trauma.

Abusive head trauma includes any nonaccidental injury inflicted to a child's head and body. It is often characterized by, but not limited to … hemorrhages that are too numerous to count, multilayered, and extending to the periphery. Traumatic retinoschisis, splitting of the retinal layers with or without blood accumulating in the intervening space, is exclusive for abusive head trauma in infants without a history of significant cerebral crush injury. Direct visualization of intraocular structures is d …
