Developmental riboflavin deficiency results in structural and functional changes in the neural retina and RPE.

The retina, a metabolically active tissue, relies on adequate flavin levels for optimal function. Our previous research demonstrated that ablation of RTBDN, a retina-specific riboflavin binding protein, plays a pivotal role in maintaining flavin levels, leading to progressive retinal degeneration. This raises the fundamental question of how riboflavin deficiency impacts retinal structure and function. We have previously evaluated an adult diet-induced model of riboflavin deficiency and showed that lack of flavins resulted in severe functional and structural deficits in the neural retina and re…

Antioxidant enzyme levels in the erythrocytes of riboflavin-deficient and Trichinella spiralis-infected rats.

The erythrocyte antioxidant enzyme levels of catalase, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) of riboflavin-deficient and Trichinella spiralis-infected rats were investigated. The rats were deprived of riboflavin at the 8th week of the experiment. At that time, the erythrocyte glutathione reductase activity coefficient (EGR AC), as an indicator of riboflavin status, was > or = 1.30 in rats fed a riboflavin-deficient diet and T. spiralis-infected rats fed a riboflavin-deficient diet showed no biochemical sign of riboflavin deficiency. At the 12th week of the experiment, …

Correlation of H2O2 production and liver catalase during riboflavin deficiency and repletion in mammals.

A substantial decrease in liver peroxisomal catalase was found during riboflavin deficiency in rats. This decrease is greater than that found among other hemoproteins and seems to follow decrease in flavin-dependent peroxisomal oxidases. This is not due to a general depression of peroxisomal enzymes, since Cu-dependent urate oxidase activity was not changed. Furthermore, the level of catalase activity as well as flavin-dependent oxidases was restored by riboflavin repletion. These results suggest that hydrogen peroxide, the substrate for catalase produced by several flavoprotein oxidases, indu…

Feeding responses of riboflavin-deficient rats to energy dilution, cold exposure and glucoprivation.

Feeding in response to various stimuli, was investigated in riboflavin-deficient rats. The animals failed to increase their food intake when fed energy-diluted diets with cellulose contents of either 15 or 30%. Furthermore, they lost reactivity to the hyperphagic action of either insulin of 2-deoxy-D-glucose regardless of a perfect response in blood glucose. Cold exposure (10 degrees), however, stimulated energy intake of the deficient rats to the same extent as that of the riboflavin-supplemented normal rats. These findings indicate that there was an operative thermostatic, but not a glucosta

Riboflavin deficiency in man: effects on haemoglobin and reduced glutathione in erythrocytes of different ages.

1. Erythrocytes (RBC) from control and marginally riboflavin-deficient subjects were fractionated into nine fractions using a discrete density gradient. 2. Glutathione reductase (NAD(P)H: glutathione oxidoreductase; EC 1.6.4.2) activity and aspartate aminotransferase (EC 2.6.1.1) activity (with and without the appropriate co-enzymes) reduced glutathione, methaemoglobin, sulphaemoglobin and oxyhaemoglobin and susceptibility to peroxide were measured in RBC in the different fractions. 3. Glutathione reductase and aspartate aminotransferase activities and concentrations of reduced glutathione and

L-gulonolactone oxidase activity and vitamin C status in riboflavin-deficient rats.

The effect of riboflavin deficiency on the activity of L-gulonolactone oxidase [L-gulono-gamma-lactone: oxygen 2-oxidoreductase, EC 1.1.3.8] and on vitamin C status was studied. A marked decrease in the specific activity of L-gulonolactone oxidase was observed in the liver microsomes isolated from riboflavin-deficient rats: the specific activity was approx. one-third of that in the microsomes isolated from control rats. The L-ascorbic acid content in the liver of the riboflavin-deficient rats was approx. one-half of that in the liver of the control rats. It seems that the rate of production of…

Effect of riboflavin deficiency on the 32P turnover in the macromolecular phosphate compounds of the rat lens.

In order to clarify the role of riboflavin in phosphorylation in the lens, the authors have attempted to investigate the 32P turnover in the macromolecule fraction in the lens supernatant from riboflavin deficient rats. 32P turnover in the macromolecular phosphate compounds was reduced remarkably. The macromolecular phosphate compounds in the lens supernatant which was separated by Sephadex G-50 was identified as RNA. These findings reveal that there are close relationships between reduced RNA turnover in the lens and riboflavin deficiency. This concept may contribute greatly to clarifying the…

Maternal riboflavin deficiency causes embryonic defects by activating ER stress-induced hepatocyte apoptosis pathway.

Riboflavin deficiency (RD) induces liver damage, abnormal embryonic development, and high mortality. We hypothesized that the phenotype could be rescued by inhibiting ER stress. The objectives of the present study were to investigate the underlying molecular mechanisms of RD-induced embryonic defects using in vitro and in vivo models. Primary duck embryonic hepatocytes were treated with an ER stress inhibitor (4-PBA) or transfected with CHOP siRNA, and cultured in RD medium and riboflavin-sufficient (CON) medium for 8 days. Laying ducks (n = 20 cages/diet, 1 bird/cage) were fed an RD diet or C…

Misdiagnosis of Total Parental Nutrition-Related Riboflavin Deficiency: Three Case Reports of Diagnostic Error.

Total parental nutrition (TPN) is a critical component of neonatal intensive care. Supply shortages leading to deficiencies in TPN constituents can have devastating consequences for critically ill patients in the neonatal intensive care unit (NICU), who may be initially misdiagnosed as potential inborn errors of metabolism. Here, we present three cases of patients with prolonged TPN dependence due to intra-abdominal pathology who presented with signs and symptoms concerning for metabolic disorders and who were ultimately determined to be a result of vitamin deficiencies in the TPN after unnece

ETFDH mutations as a major cause of riboflavin-responsive multiple acyl-CoA dehydrogenation deficiency.

Multiple acyl-CoA dehydrogenation deficiency (MADD) is a disorder of fatty acid, amino acid and choline metabolism that can result from defects in two flavoproteins, electron transfer flavoprotein (ETF) or ETF: ubiquinone oxidoreductase (ETF:QO). Some patients respond to pharmacological doses of riboflavin. It is unknown whether these patients have defects in the flavoproteins themselves or defects in the formation of the cofactor, FAD, from riboflavin. We report 15 patients from 11 pedigrees. All the index cases presented with encephalopathy or muscle weakness or a combination of these sympto

Early paranodal myelin swellings (tomacula) in an avian riboflavin deficiency model of demyelinating neuropathy.

INTRODUCTION: Disruption of the complex architectural and molecular organization of the paranodal region of myelinated peripheral nerve fiber may initiate the evolving time dependent process of segmental demyelination. In support of this notion was the finding of focal paranodal myelin swellings (tomacula) due to redundant folding of myelin sheaths, early in the time course of an avian riboflavin deficiency model of demyelinating neuropathy. METHODS: Newborn broiler meat chickens were maintained either on a routine diet containing 5.0 mg/kg riboflavin (control group) or a riboflavin-deficient…
