Tenuous transcriptional threshold of human sex determination. II. SRY exploits water-mediated clamp at the edge of ambiguity.

Y-encoded transcription factor SRY initiates male differentiation in therian mammals. This factor contains a high-mobility-group (HMG) box, which mediates sequence-specific DNA binding with sharp DNA bending. A companion article in this issue described sex-reversal mutations at box position 72 (residue 127 in human SRY), invariant as Tyr among mammalian orthologs. Although not contacting DNA, the aromatic ring seals the domain's minor wing at a solvent-exposed junction with a basic tail. A seeming paradox was posed by the native-like biochemical properties of inherited Swyer variant Y72F: its

The mouse Sry locus harbors a cryptic exon that is essential for male sex determination.

The mammalian sex-determining gene Sry induces male development. Since its discovery 30 years ago, Sry has been believed to be a single-exon gene. Here, we identified a cryptic second exon of mouse Sry and a corresponding two-exon type Sry (Sry-T) transcript. XY mice lacking Sry-T were sex-reversed, and ectopic expression of Sry-T in XX mice induced male development. Sry-T messenger RNA is expressed similarly to that of canonical single-exon type Sry (Sry-S), but SRY-T protein is expressed predominantly because of the absence of a degron in the C terminus of SRY-S. Sry exon2 appears to have ev

Overexpression of Kdm6b induces testicular differentiation in a temperature-dependent sex determination system.

In reptiles, such as the red-eared slider turtle ( Trachemys scripta elegans), gonadal sex determination is highly dependent on the environmental temperature during embryonic stages. This complex process, which leads to differentiation into either testes or ovaries, is governed by the finely tuned expression of upstream genes, notably the testis-promoting gene Dmrt1 and the ovary-promoting gene Foxl2. Recent studies have identified epigenetic regulation as a crucial factor in testis development, with the H3K27me3 demethylase KDM6B being essential for Dmrt1 expression in T. s. elegans. However,

The good, the bad, and the ugly: Evolutionary and pathological aspects of gene dosage alterations.

Diploid organisms contain a maternal and a paternal genome complement that is thought to provide robustness and allow developmental progression despite genetic perturbations that occur in heterozygosity. However, changes affecting gene dosage from the chromosome down to the individual gene level possess a significant pathological potential and can lead to developmental disorders (DDs). This indicates that expression from a balanced gene complement is highly relevant for proper cellular and organismal function in eukaryotes. Paradoxically, gene and whole chromosome duplications are a principal

Activation of stress response axis as a key process in environment-induced sex plasticity in fish.

The determination of sex is an important hallmark in the life cycle of organisms, in which the fate of gonads and then the individual sex are defined. In gonochoristic teleost fish, this process is characterized by a high plasticity, considering that in spite of genotypic sex many environmental factors can cause shifts from one to another molecular pathway, resulting in organisms with mismatching genotypic and phenotypic sexes. Interestingly, in most instances, both female-to-male or male-to-female sex-reversed individuals develop functional gonads with normal gametogenesis and respective prog

Efficacy of a commercially available post-thaw bovine semen sexing kit in both single-ovulating and hyperstimulated cows.

Currently, there are few inexpensive, reliable, effective methods for commercially separating X- and Y-chromosome bearing fresh and frozen bovine sperm. The objective of these experiments was to determine the efficacy of a commercially available post-thaw bovine semen sexing kit, HeiferPlus (HP) which claims to alter the sex ratio in favor of female calves following artificial insemination. Three trials included the insemination of hyperstimulated cows with Control or HP-treated semen, non-surgical embryo collection on Day 7, and a combined PCR/dot blot assay to determine embryo sex. Development of the genitalia in Drosophila melanogaster.

The imaginal discs of Drosophila melanogaster are an excellent material with which to analyze how signaling pathways and Hox genes control growth and pattern formation. The study of one of these discs, the genital disc, offers, in addition, the possibility of integrating the sex determination pathway into this analysis. This disc, whose growth and shape are sexually dimorphic, gives rise to the genitalia and analia, the more posterior structures of the fruit fly. Male genitalia, which develop from the ninth abdominal segment, and female genitalia, which develop mostly from the eighth one, disp

amh alleles on XY sex chromosomes regulate male differentiation in amphidiploid Carassius auratus.

The amh (anti-Müllerian hormone) gene, residing on the sex chromosomes, has been hypothesized to be a potential master sex gene in amphidiploid Carassius auratus (crucian carp/goldfish). However, its role in male determination or differentiation remains unclear. Here, we identified the heteromorphic X and Y chromosomes and confirmed the localization of amh on the sex chromosome by fluorescence in situ hybridization in amphidiploid C. auratus. The transcriptional expressions of X-linked and Y-linked amh alleles showed no significant difference during the period of sex determination and differen

Hermaphroditism in Cannabis sativa L.: Impacts, Inducers, and Industry Implications.

Cannabis sativa L. is a predominantly dioecious species, but sex expression is highly plastic and can be modified by genetic, hormonal, developmental, and environmental factors. This plasticity has major implications for commercial production because hermaphroditic expression in female plants can cause unintended pollination, seed formation, reduced floral quality, and losses in cannabinoid yield. This review summarizes current understanding of sex determination and sex-expression instability in C. sativa, with emphasis on hermaphroditism and its agronomic significance. We examine the genomic

Multilevel transcriptional regulation of embryonic sex determination and dosage compensation by the X-signal element sex-1.

The Caenorhabditis elegans nuclear hormone receptor sex-1 is an embryonic X-signal element that represses xol-1, the sex-switch gene that is the master regulator of sex determination and dosage compensation. Several prior studies on sex-1 function have suggested that sex-1 may have additional downstream roles beyond the regulation of xol-1 expression. In this study, we characterize some of these additional roles of sex-1 in regulating the dual processes of sex determination and dosage compensation during embryogenesis. Our study reveals that sex-1 acts on many of the downstream targets of xol-

Detection of non-invasive sexing of early chick embryos in intact eggs using laser speckle contrast imaging and deep neural networks.

The ability to image blood flow in early-stage avian embryos has significant applications in developmental biology, drug and vaccine testing, as well as determining sex differentiation. In this project, a recently developed laser speckle contrast imaging (LSCI) system was used to non-invasively image extraembryonic blood vessels and used these images to attempt early sex identification of chick embryos. Specifically, blood vessels images were captured from 1,251 living chicken embryos between day three and day four of incubation. Then, deep neural network (DNN) models were applied to evaluate

Integration of ATAC-seq and RNA-seq unravels chromatin accessibility associated with gonad development after sex reversal in GIFT tilapia.

The epigenetic mechanisms regulating gonadal development and maintenance in fish following hormone-induced sex reversal remain poorly understood. In order to address this knowledge gap, an integrated multi-omics analysis was conducted in GIFT tilapia. The present study compared three distinct groups: normal females (CF; genetically female, XX), normal males (CM; genetically male, XY), and pseudo-females (S; estrogen-treated genetically males, XY). The S group demonstrated intermediate growth traits and sex hormone levels that fell between those observed in the CF and CM groups. In comparison w

Chromosome-level de novo genome assembly and whole-genome resequencing provide insights into genome evolution and sex determination of sleepy cod Oxyeleotris lineolata (Steindachner 1867).

The sleepy cod (Oxyeleotris lineolata Steindachner 1867) is a valuable freshwater teleost with male-biased growth dimorphism, making sex control breeding vital for aquaculture. We generated a high-quality chromosome-level genome assembly (⁓985.07 Mb) using PacBio HiFi and Hi-C sequencing, with 99.31% sequences anchored to 23 chromosomes (contig N50: 14.73 Mb; scaffold N50: 42.32 Mb). BUSCO assessment showed 98.1% completeness, with 25,933 protein-coding genes and 42.21% repetitive elements annotated. Phylogenetic analysis indicated closest affinity to Gobiidae (Boleophthalmus pectinirostris, P

Gonadal transcriptome analysis of Cirrhina molitorella reveals sex-associated genes.

Cirrhina molitorella is an important economic fish species in southern China, and it holds a significant position among the highest-yielding fish species in this region. However, studies on the identification of sex-related genes and gonadal development in C. molitorella remain scarce, which has severely hindered the development of its aquaculture industry. In the present study, transcriptome sequencing data from the female and male gonads of C. molitorella were analyzed and compared for the first time, leading to the successful assembly of 64,954 unigenes. Through comparative transcriptomic a

Identification of genes associated with sex expression and sex determination in hemp (Cannabis sativa L.).

Dioecy in flowering plants has evolved independently many times, and thus the genetic mechanisms underlying sex determination are diverse. In hemp (Cannabis sativa), sex is controlled by a pair of sex chromosomes (XX for females and XY for males). In an attempt to understand the molecular mechanism responsible for sex expression in hemp plants, we carried out RNA sequencing of male and female plants at different developmental stages. Using a pipeline involving differential gene expression analysis and weighted gene co-expression network analysis, we identified genes important for male and fema

A conserved GRAS-domain transcriptional regulator links meristem indeterminacy to sex determination in Ceratopteris gametophytes.

Most land plants alternate between generations of sexual gametophytes and asexual sporophytes. Unlike seed plants, fern gametophytes are free living and grow independently of their sporophytes. In homosporous ferns such as Ceratopteris, gametophytes derived from genetically identical spores exhibit sexual dimorphism, developing as either males or hermaphrodites. Males lack meristems and promote cell differentiation into sperm-producing antheridia. In contrast, hermaphrodites initiate multicellular meristems that stay undifferentiated, sustain cell division and prothallus expansion, and drive t

A novel c.1759T>G variant in follicle-stimulating hormone-receptor gene is concordant with male determination in the flathead grey mullet (Mugil cephalus).

Various master key regulators (MKRs) that control a binary switch of sex determination (SD) have been found in fish; these provide an excellent model for the study of vertebrate genetic SD. The SD region in flathead grey mullet has been previously mapped to a 1 Mbp region harboring 27 genes, of which one is follicle-stimulating hormone receptor (fshr). Although this gene is involved in gonad differentiation and function, it has not been considered as an MKR of SD. We systematically investigated polymorphism in mullet fshr using DNA shotgun sequences, and compared them between males and females
