Molecular features of myosin F adapted for driving actin flows in Toxoplasma gondii.

Toxoplasma gondii is a single-celled apicomplexan parasite that relies on a highly polarized endomembrane system for its invasion of, and survival within, host cells. Recent advances in imaging technologies have revealed that vesicle transport and the organization of organelles within the endomembrane pathway require a highly dynamic actin cytoskeleton. These dynamics in turn rely on the activity of myosin F (MyoF), a molecular motor unique to alveolates. The defining characteristic of this molecular motor is a WD40 β-propeller domain, exclusively found in this class of myosin. To understand t

α-Synuclein Radiotracer Development and In Vivo Imaging: Recent Advancements and New Perspectives.

α-Synucleinopathies including idiopathic Parkinson's disease, dementia with Lewy bodies and multiple systems atrophy share overlapping symptoms and pathological hallmarks. Selective neurodegeneration and Lewy pathology are the main hallmarks of α-synucleinopathies. Currently, there is no imaging biomarker suitable for a definitive early diagnosis of α-synucleinopathies. Although dopaminergic deficits detected with single-photon emission computed tomography (SPECT) and positron emission tomography (PET) radiotracers can support clinical diagnosis by confirming the presence of dopaminergic neuro

Differentiating Alzheimer's Aβ Isoforms Coaggregated in Cerebrospinal Fluid via Single-Particle Imaging.

Amyloid polymorphism can reflect Alzheimer's disease (AD) stages. This paper demonstrates that amyloid β (Aβ) peptides, primarily Aβ-40 and Aβ-42 (implicated in AD pathology), present in cerebrospinal fluid (CSF), can be differentiated, and their morphology studied in detail using fluorescence-based super-resolution and atomic force microscopy (AFM). An inhibitory effect of Aβ-40 on Aβ-42 protein aggregation, marked by Aβ-40 oligomers colocalizing along the Aβ-42 fibril backbone, was resolved at the single-particle level. Molecular dynamics simulations revealed that coaggregation is modulated

Clinical Applications of Theranostics.

Radiotheranostics integrates molecular imaging and targeted radionuclide therapy to provide personalized cancer treatment. By using radiolabeled agents that bind tumor-specific targets, clinicians can both visualize and treat malignancies, guided by the principle of "treat what you see." Diagnostic tools such as positron emission tomography (PET) and single-photon emission computed tomography (SPECT) confirm target presence, while therapeutic radioisotopes deliver cytotoxic radiation to tumor cells, enabling real-time monitoring and individualized treatment. These agents pair a targeting molecu

Single-cell RNA sequencing reveals chemokine self-feeding of myeloma cells promotes extramedullary metastasis.

In this study, we aimed to determine the mechanisms underlying the initial extramedullary translocation of myeloma cells from bone marrow into peripheral blood. We found that clonal circulating plasma cells (cPCs) are more frequently detected by flow cytometry in extramedullary plasmacytoma (EMP) patients and worsen their prognosis. It is technically much easier to collect single cPCs using FACS than it is to perform EMP biopsy. Therefore, combining EMP imaging with cPC detection may be a promising strategy for prognostic stratification. Here, using single-cell transcriptome analysis, we found

Dengue Virus Capsid-Protein Dynamics in Live Infected Cells Studied by Pair Correlation Analysis.

It has become increasingly evident that unveiling the mechanisms of virus entry, assembly, and virion release is fundamental for identifying means for preventing viral spread and controlling viral disease. Due to virus mobility and structural and/or functional heterogeneity among viral particles, high spatiotemporal resolution single-virus/single-particle techniques are required to capture the behavior of viral particles inside infected cells. In this chapter, we present fluorescence imaging analysis methods for studying the mobility of fluorescently labeled dengue virus (DENV) proteins in live

Emerging Nanoscale Platforms for Precision Glycan Analysis: Molecular Fingerprinting, Nanopore Sensing, and Spatial Readouts.

Glycans present two intrinsic properties that make nanoscale analysis a necessity rather than a refinement: a microheterogeneity that arises from non-templated biosynthesis, and a structural information density that surpasses that of nucleic acids and proteins by orders of magnitude. Both demand readout at the level of the individual molecule, on scales from the chemical bond to the residue to the spatial register. Five emerging platforms now meet these requirements along complementary dimensions. Tip-enhanced Raman spectroscopy reads single glycosidic linkages through vibrational fingerprints

Sample preparation strategies for high-throughput mass spectrometry imaging of primary tumor organoids.

Patient-derived 3D organoids show great promise for understanding patient heterogeneity and chemotherapy response in human-derived tissue. The combination of organoid culture techniques with mass spectrometry imaging provides a label-free methodology for characterizing drug penetration, patient-specific response, and drug biotransformation. However, current methods used to grow tumor organoids employ extracellular matrices that can produce small molecule background signal during mass spectrometry imaging analysis. Here, we develop a method to isolate 3D human tumor organoids out of a Matrigel

Early Steps of Individual Multireceptor Viral Interactions Dissected by High-Density, Multicolor Quantum Dot Mapping in Living Cells.

Viral capture and entry to target cells are the first crucial steps that ultimately lead to viral infection. Understanding these events is essential toward the design and development of suitable antiviral drugs and/or vaccines. Viral capture involves dynamic interactions of the virus with specific receptors in the plasma membrane of the target cells. In the last years, single virus tracking has emerged as a powerful approach to assess real time dynamics of viral processes in living cells and their engagement with specific cellular components. However, direct visualization of the early steps of

Molecular imaging: an overview and clinical applications.

Molecular imaging is a new medical discipline that integrates cell biology, molecular biology and diagnostic imaging. Clinical applications of molecular imaging include the use of nuclear medicine, magnetic resonance imaging (MRI) and ultrasound (US). The nuclear medicine applications utilize devices such as single photon emission computerized tomography (SPECT) and positron emission tomography (PET). Molecular imaging has two basic applications. The first is diagnostic imaging, which is used to determine the location and extent of targeted molecules specific to the disease being assessed. The

Whole-Cell Lysosome SMLM Imaging as Indicators for Functional Diagnostics with a Low-Phototoxic Spontaneously Blinking Probe.

Lysosomal morphology and pH dynamics are key indicators of lysosomal function, making long-term single-molecule localization microscopy (SMLM) imaging a promising tool for functional diagnostics. However, phototoxicity often compromises imaging reliability. Here, we develop Aze-HMSiR, a spontaneously blinking silicon rhodamine probe with near-infrared excitation, enabling low-phototoxicity, long-term (50 min) SMLM imaging of lysosomal morphology and pH dynamics. This probe provides super-resolution insights into lysosomal distribution, size, and lumen pH, essential for understanding their phys

Dopamine Self-Polymerization-Assisted Oriented Antibody Immobilization on a Transistor for Alzheimer's Disease Diagnosis.

Antibody transistor biosensors show promise for early Alzheimer's disease (AD) diagnosis due to their single molecule detection capability. However, the random attachment of a payload to antigen-binding fragments (Fab) during biosensor preparation can disrupt target recognition, leading to poor consistency and repeatability. Here, we present a rapid and simple antibody-oriented immobilization method through a dopamine self-polymerization (DASP) process. Experimental and molecular dynamics simulation validation confirms the performance and mechanism of antibody orientation modification, attribu

Lecanemab preferentially binds to smaller aggregates present at early Alzheimer's disease.

INTRODUCTION: The monoclonal antibodies Aducanumab, Lecanemab, Gantenerumab, and Donanemab were developed for the treatment of Alzheimer's disease (AD). METHODS: We used single-molecule detection and super-resolution imaging to characterize the binding of these antibodies to diffusible amyloid beta (Aβ) aggregates generated in-vitro and harvested from human brains. RESULTS: Lecanemab showed the best performance in terms of binding to the small-diffusible Aβ aggregates, affinity, aggregate coating, and the ability to bind to post-translationally modified species, providing an explanation for

A functional RNA-origami as direct thrombin inhibitor with fast-acting and specific single-molecule reversal agents in vivo model.

Injectable anticoagulants are widely used in medical procedures to prevent unwanted blood clotting. However, many lack safe, effective reversal agents. Here, we present new data on a previously described RNA origami-based, direct thrombin inhibitor (HEX01). We describe a new, fast-acting, specific, single-molecule reversal agent (antidote) and present in vivo data for the first time, including efficacy, reversibility, preliminary safety, and initial biodistribution studies. HEX01 contains multiple thrombin-binding aptamers appended on an RNA origami. It exhibits excellent anticoagulation activ

Multicolor in vivo targeted imaging to guide real-time surgery of HER2-positive micrometastases in a two-tumor coincident model of ovarian cancer.

One of the primary goals of oncological molecular imaging is to accurately identify and characterize malignant tissues in vivo. Currently, molecular imaging relies on targeting a single molecule that while overexpressed in malignancy, is often also expressed at lower levels in normal tissue, resulting in reduced tumor to background ratios. One approach to increasing the specificity of molecular imaging in cancer is to use multiple probes each with distinct fluorescence to target several surface antigens simultaneously, in order to identify tissue expression profiles, rather than relying on the

In situ single-cell therapeutic response imaging facilitated by the TRIPODD fluorescence imaging platform.

Purpose: Small molecule drugs such as tyrosine kinase inhibitors (TKIs) targeting tumoral molecular dependencies have become standard of care for numerous cancer types. Notably, epidermal growth factor receptor (EGFR) TKIs (e.g., erlotinib, afatinib, osimertinib) are the current first-line treatment for non-small cell lung cancer (NSCLC) due to their improved therapeutic outcomes for EGFR mutated and overexpressing disease over traditional platinum-based chemotherapy. However, many NSCLC tumors develop resistance to EGFR TKI therapy causing disease progression. Currently, the relationship betw

Upconversion nanoparticle-assisted single-molecule assay for detecting circulating antigens of aggressive prostate cancer.

Sensitive and quantitative detection of molecular biomarkers is crucial for the early diagnosis of diseases like metabolic syndrome and cancer. Here we present a single-molecule sandwich immunoassay by imaging the number of single nanoparticles to diagnose aggressive prostate cancer. Our assay employed the photo-stable upconversion nanoparticles (UCNPs) as labels to detect the four types of circulating antigens in blood circulation, including glypican-1 (GPC-1), leptin, osteopontin (OPN), and vascular endothelial growth factor (VEGF), as their serum concentrations indicate aggressive prostate

A thiol-activated near-infrared fluorescent H2S donor for visualizing H2S release and its applications.

Hydrogen sulfide (H2S), a critical endogenous signaling molecule, is involved in diverse physiological processes, and H2S donors have emerged as promising therapeutic agents. While organic donors address the uncontrolled release limitations of inorganic counterparts, most thiol-activated systems depend exclusively on cysteine (Cys), risking cellular thiol imbalance. To overcome this limitation, we developed AD-SH, a novel benzopyran salts-based NIR fluorescent H2S donor responsive to multiple biothiols. AD-SH exhibits high selectivity and sensitivity toward all intracellular biothiols (e.g., c

Stress hormone rapidly tunes synaptic NMDA receptor through membrane dynamics and mineralocorticoid signalling.

Stress hormones, such as corticosteroids, modulate the transmission of hippocampal glutamatergic synapses and NMDA receptor (NMDAR)-dependent synaptic plasticity, favouring salient behavioural responses to the environment. The corticosterone-induced synaptic adaptations partly rely on changes in NMDAR signalling, although the cellular pathway underlying this effect remains elusive. Here, we demonstrate, using single molecule imaging and electrophysiological approaches in hippocampal neurons, that corticosterone specifically controls GluN2B-NMDAR surface dynamics and synaptic content through mi…
