Influence of the Manufacturing Method on the Adhesion of Candida albicans and Streptococcus mutans to Oral Splint Resins.

Microbial adhesion to oral splints may lead to oral diseases such as candidiasis, periodontitis or caries. The present in vitro study aimed to assess the effect of novel computer-aided design/computer-aided manufacturing (CAD/CAM) and conventional manufacturing on Candida albicans and Streptococcus mutans adhesion to oral splint resins. Standardized specimens of four 3D-printed, two milled, one thermoformed and one pressed splint resin were assessed for surface roughness by widefield confocal microscopy and for surface free energy by contact angle measurements. Specimens were incubated with C.…

Specific strains of Streptococcus mutans, a pathogen of dental caries, in the tonsils, are associated with IgA nephropathy.

Streptococcus mutans is known to be a major causative agent of dental caries, and strains expressing the cell surface collagen-binding Cnm protein contribute to the development of several systemic diseases. A relationship between tonsillar immunity and glomerulonephritis has been recognized in IgA nephropathy (IgAN), and specific pathogens may have effects on tonsillar immunity (mucosal immunity). Here, we present findings showing a relationship between the presence of Cnm-positive S. mutans strains in the tonsils of IgAN patients and IgAN condition/pathogenesis. Analyses of tonsillar specimen…

Isolation and Characterisation of Probiotics for Antagonising the Cariogenic Bacterium Streptococcus mutans and Preventing Biofilm Formation.

PURPOSE: Consumption of refined foods and beverages high in sugar make the teeth susceptible to the formation of biofilm and may lead to dental caries. The aim of the present study was to determine the ability of selected probiotics to inhibit growth and biofilm formation by the cariogenic bacterium Streptococcus mutans in vitro. MATERIALS AND METHODS: Strains of latic acid bacteria (LAB) (n = 120) from the Bioresources Collection and Research Center (BCRC), saliva of healthy adults and infant stool were screened. The antimicrobial activity of LAB in vitro was evaluated by agar spot culture a…

Assessment of Streptococcus mutans in healthy versus gingivitis and chronic periodontitis: A clinico-microbiological study.

BACKGROUND: Dental caries and periodontal disease are most common oral diseases. Streptococcus mutans are considered to be the major pathogens in initiation of dental caries. Evidence shows that periodontal disease and caries share a number of contributory factors. Thus in view of these findings it would be worthwhile to examine whether Streptococcus mutans persist within the saliva and subgingival environment of the periodontitis patients and to determine whether there is any association between Streptococcus mutans colonization, pH of saliva and sub-gingival plaque pH in periodontal diseases…

Contribution of the Collagen-Binding Proteins of Streptococcus mutans to Bacterial Colonization of Inflamed Dental Pulp.

Streptococcus mutans is a major pathogen of dental caries. Collagen-binding proteins (CBPs) (approximately 120 kDa), termed Cnm and Cbm, are regarded as important cell surface antigens related to the adherence of S. mutans to collagenous tissue. Furthermore, CBP-positive S. mutans strains are associated with various systemic diseases involving bacteremia, such as infective endocarditis. Endodontic infection is considered to be an important cause of bacteremia, but little is known regarding the presence of S. mutans in dental pulp tissue. In the present study, the distribution and virulence of …

Contribution of the interaction of Streptococcus mutans serotype k strains with fibrinogen to the pathogenicity of infective endocarditis.

Streptococcus mutans, a pathogen responsible for dental caries, is occasionally isolated from the blood of patients with bacteremia and infective endocarditis (IE). Our previous study demonstrated that serotype k-specific bacterial DNA is frequently detected in S. mutans-positive heart valve specimens extirpated from IE patients. However, the reason for this frequent detection remains unknown. In the present study, we analyzed the virulence of IE from S. mutans strains, focusing on the characterization of serotype k strains, most of which are positive for the 120-kDa cell surface collagen-bind…

Effect of an essential oil-containing antiseptic mouthrinse on plaque and salivary Streptococcus mutans levels.

BACKGROUND: Clinical studies in which antimicrobial mouthrinses were shown to have significant antiplaque activity most frequently have used gingivitis as the clinically relevant endpoint. However, there is evidence to suggest that mouthrinses containing active agents effective against Streptococcus mutans, such as chlorhexidine, may also have a role in inhibiting dental caries. This clinical study was conducted to determine the effect of 2x-daily rinsing with an essential oil-containing antiseptic mouthrinse (Listerine Antiseptic) on levels of recoverable S. mutans and total streptococci in s…

Identification and characterization of a surface protein-releasing activity in Streptococcus mutans and other pathogenic streptococci.

Surface proteins of Streptococcus mutans have been reported to be released into the culture filtrate at concentrations that vary with the growth conditions. The reason for this is not clear. The present study attempts to investigate the mechanism of the protein release. The results showed that whole cells and raffinose-stabilized protoplasts of S. mutans NG8, when incubated in buffers, were capable of releasing their surface proteins in a pH-dependent manner with optimal release at pH 5 to 6. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis revealed that the released proteins…

Acetoin production by wild-type strains and a lactate dehydrogenase-deficient mutant of Streptococcus mutans.

Eleven different laboratory strains of Streptococcus mutans representing the various serogroups were found to produce an average of 6.0 +/- 4.8 mM acetoin when grown in glucose-containing medium under aerobic conditions. None of the strains produced detectable acetoin when grown anaerobically. A lactate dehydrogenase-deficient mutant produced acetoin both aerobically and anaerobically and in substantially greater amounts than the wild-type strains did. Substitution of mannitol for glucose resulted in decreased acetoin production by wild-type strains and the lactate dehydrogenase-deficient muta…

Dissecting the essential role of luxS-relevant small RNAs in biofilm formation of Streptococcus mutans.

Streptococcus mutans is considered the key contributor to human dental caries. The LuxS/AI-2 quorum‑sensing (QS) system in S. mutans plays a crucial role in the development of cariogenic oral biofilms. Many QS systems rely on regulation carried out by small RNAs (sRNAs) to achieve optimal performance. However, the identities and functions of sRNAs in S. mutans QS system are not well elucidated. Here, we identify two novel sRNAs named SmqsR2 and SmqsR4 (Streptococcus mutans quorum-sensing-related small RNA 2 and 4), which respond to luxS expression and support biofilm formation by mediating exo…

New insights into iron uptake in Streptococcus mutans: evidence for a role of siderophore-like molecules.

Streptococcus mutans, a gram-positive coccus commonly found in the human oral cavity, is the primary causative agent of dental caries as well as infective endocarditis. Bacteria produce potent iron chelators called siderophores to absorb iron. Because, there are few studies on siderophore-mediated iron transport in S. mutans, the current study investigates the presence of such a mechanism in S. mutans GS-5. Deferration of culture medium and different concentrations of 2, 2'-Bipyridyl has been used to simulate iron-restricted conditions. Iron restriction alters the colony morphology and slows b…

Serotype and distribution of adhesion genes in Streptococcus mutans isolated from people with Parkinson's disease.

The delicate balance of the oral cavity is disrupted by changes in hygiene and diet, leading to caries. The virulence of Streptococcus mutans, a key caries pathogen, is partly explained by its rhamnose-glucose polysaccharide (RGP) and its sucrose-dependent and -independent adhesion mechanisms. Given their role in comorbidities, this study investigates the diversity of S. mutans through analysis of RGP and sucrose-independent adhesion proteins (SpaP, WapA, Cnm, Cbm) in Parkinson's disease patients. In the PARKIDENT clinical trial, strains were isolated from saliva samples of Parkinson's patient…

Anti-cariogenic activity of mutanocyclin, a secondary metabolite of Streptococcus mutans, in mono- and multispecies biofilms.

UNLABELLED: Streptococcus mutans (S. mutans), a principal cariogenic pathogen responsible for dental caries, alters the oral biofilm ecology through the production of various metabolites. One such metabolite, mutanocyclin (MUC), a recently identified secondary metabolite, may have implications for both caries development and oral microbiome interactions. This study aimed to investigate the properties of MUC and its effects on S. mutans, as well as other oral commensal streptococci in mono- and multispecies biofilms. Our findings revealed that MUC significantly inhibited S. mutans planktonic gr…
