Immune cell subsets as therapeutic targets in liver fibrosis: from mechanisms to translational applications.

Liver fibrosis represents a chronic hepatic pathological condition characterized by excessive deposition of extracellular matrix (ECM) components and aberrant connective tissue proliferation. In recent years, the global prevalence of liver fibrosis has shown a concerning upward trend. However, therapeutic options remain limited, with only few pharmacological interventions currently approved by the Food and Drug Administration (FDA) for clinical management. Therefore, elucidating the molecular mechanisms underlying fibrogenesis is crucial for the development of targeted therapeutic strategies t

A translational approach to improve therapeutics in atopic dermatitis and beyond.

Atopic dermatitis (AD) and alopecia areata are highly prevalent inflammatory skin/hair conditions. While previously not fully understood and limited in treatment options, AD is currently undergoing a therapeutic revolution. Our increased understanding of the underlying immunologic and barrier dysregulations and disease heterogeneity across its spectrum is facilitating hypothesis-driven therapeutic development. Early transcriptomic analyses in AD skin and blood have identified disease-specific biomarkers and uncovered immune and barrier abnormalities that may contribute to disease pathogenesis.

Bilirubin: translational perspectives.

Bilirubin, an old tetrapyrrolic compound that had occurred on Earth early on, is the final product of the catabolic heme pathway in the intravascular bed. Data from recent decades revealed its enormous bioactivity in a human body, including antioxidant, anti-inflammatory, immunosuppressive, antiproliferative, and even cell signaling activities that translate into beneficial effects of mildly elevated serum bilirubin concentrations resulting in prevention or amelioration of progression of many diseases of civilization. Furthermore, recent advances in bilirubin research have changed our understa

Nonhuman Primate Models in Translational Surgery.

BACKGROUND: Nonhuman primates (NHPs) play a unique role in translational science by bridging the gap between basic and clinical investigations and are often seen as a last step before clinical application. They are widely utilized in biomedical research due to their anatomical and physiological similarities to humans. Examples of commonly used species include the genera Macaca (macaques), Papio (baboons), Aotus (owl monkeys), Callithrix (marmosets), Saimiri (squirrel monkeys), and Chlorocebus (vervet monkeys). SUMMARY: NHP models have played an instrumental role in the development of surgical

Navigating the Critical Translational Questions for Implementing FLASH in the Clinic.

The "FLASH effect" is an increased therapeutic index, that is, reduced normal tissue toxicity for a given degree of anti-cancer efficacy, produced by ultra-rapid irradiation delivered on time scales orders of magnitude shorter than currently conventional in the clinic for the same doses. This phenomenon has been observed in numerous preclinical in vivo tumor and normal tissue models. While the underlying biological mechanism(s) remain to be elucidated, a path to clinical implementation of FLASH can be paved by addressing several critical translational questions. Technological questions pertine

Translational horizons in stem cell therapy for osteonecrosis of the femoral head: a journey from basic research to clinical practice through bibliometric insights.

BACKGROUND: Osteonecrosis of the femoral head (ONFH) significantly impacts young and middle-aged adults, with steroid use implicated in many cases. Traditional treatments have limited efficacy, prompting a shift towards innovative approaches, such as stem cell therapy, offering less invasive regenerative solutions. METHODS: Using bibliometric analysis from 1997 to 2023, we identified 392 articles on stem cell therapy for ONFH from the Web of Science Core Collection and analysed them using VOSviewer and CiteSpace to identify key trends and research directions. RESULTS: From 1997 to 2023, stem

Creation of an Electronic Resource Repository for Differences of Sex Development (DSD): Collaboration Between Advocates and Clinicians in the DSD-Translational Research Network.

INTRODUCTION: People with differences of sex development (DSD) and their families need education about these conditions while receiving emotional and peer support to participate in shared decision-making, reduce social isolation, and optimize physical and psychosocial outcomes. Barriers to education and support include limited knowledge and awareness by healthcare providers, tension among patient and medical communities, varied quality of educational resources, and the sensitive nature of DSD. We aimed to create an electronic repository of vetted quality online resources about DSD. METHODS: T

Challenges in translational research: MDMA in the laboratory versus therapeutic settings.

Despite substantial progress in the use of mind-altering drugs to treat psychiatric disorders, the psychological processes through which these drugs change mood or behavior are poorly understood. Controlled laboratory studies with well-defined psychological constructs are valuable to understand how these drugs manifest their therapeutic benefit. However, there are substantial methodological differences between clinical studies investigating therapeutic outcome and laboratory studies investigating the processes that might underlie the therapeutic effects. Here, we examine some of these differen

Translational Preclinical Pharmacologic Disease Models for Ophthalmic Drug Development.

Preclinical models of human diseases are critical to our understanding of disease etiology, pathology, and progression and enable the development of effective treatments. An ideal model of human disease should capture anatomical features and pathophysiological mechanisms, mimic the progression pattern, and should be amenable to evaluating translational endpoints and treatment approaches. Preclinical animal models have been developed for a variety of human ophthalmological diseases to mirror disease mechanisms, location of the affected region in the eye and severity. These models offer clues to

Neural circuitry and mechanisms of waiting impulsivity: relevance to addiction.

Impatience-the failure to wait or tolerate delayed rewards (e.g. food, drug and monetary incentives)-is a common behavioural tendency in humans. However, when rigidly and rapidly expressed with limited regard for future, often negative consequences, impatient or impulsive actions underlie and confer susceptibility for such diverse brain disorders as drug addiction, attention-deficit hyperactivity disorder (ADHD) and major depressive disorder. Consequently, 'waiting' impulsivity has emerged as a candidate endophenotype to inform translational research on underlying neurobiological mechanisms an

Metastasis and cachexia: alongside in clinics, but not so in animal models.

Cancer cachexia is a paraneoplastic syndrome characterized by lean mass wasting (with or without fat mass decrease), culminating in involuntary weight loss, which is the key clinical observation nowadays. There is a notable lack of studies involving animal models to mimic the clinical reality, which are mostly patients with cachexia and metastatic disease. This mismatch between the clinical reality and animal models could at least partly contribute to the poor translation observed in the field. In this paper, we retrieved and compared animal models used for cachexia research from 2017 and 10 y

Advances in the clinical translation of nanotechnology.

The use of novel materials in the nano-scale size range for applications in devices, drugs and diagnostic agents comes with a number of new opportunities, and also serious challenges to human applications. The larger size of particulate-based agents, as compared to traditional drugs, allows for the significant advantages of multivalency and multi-functionality. However, the human use of nanomaterials requires a thorough understanding of the biocompatibility of the synthetic molecules and their complex pharmacology. Possible toxicities created by the unusual properties of the nanoparticles are
