Symbiotic cancer cell-stroma crosstalk in the acidic tumor microenvironment.

The acidic microenvironment (AME) is a defining metabolic feature of solid tumors that fundamentally reshapes tumor-stroma interactions. Tumor acidosis arises from multiple metabolic processes, including hypoxia-driven glycolysis in poorly vascularized regions, the Warburg Effect (WE) in well-perfused areas, and carbon dioxide accumulation from tumor respiration. As a sustained ecological stress distinct from physiological pH (∼7.4), extracellular acidosis imposes powerful selective pressures within the tumor ecosystem. In response, cancer and stromal cells must adapt through selection of pre-

NAT10-Mediated ac4C Modification of circANKRD12 Reprograms the Tumor Microenvironment.

Developing anticancer strategies that simultaneously target both tumor proliferation and the immunosuppressive microenvironment remains a major challenge. However, the role of chemical modifications in circular RNAs (circRNAs) in this process remains poorly understood. In this study, we identified circANKRD12 as a key substrate for N4-acetylcytidine (ac4C) modification, catalyzed by N-acetyltransferase 10 (NAT10) in multiple myeloma (MM). This ac4C modification promotes the translation of circANKRD12 into a novel 354-amino acid protein (circANKRD12_354aa). Functionally, circANKRD12_354aa inter

Tumor immune microenvironment in non-small cell lung cancer progression.

Non-small cell lung cancer (NSCLC), the most prevalent form of lung malignancy, remains a leading cause of cancer-related mortality despite advances in surgery, targeted therapy, and immunotherapy. A growing body of evidence implicates the tumor immune microenvironment (TIME) as a critical determinant of disease progression, immune evasion, and therapeutic resistance. This review synthesizes current insights into the roles of innate and adaptive immune populations-including tumor-associated macrophages, neutrophils, dendritic cells, mast cells, myeloid-derived suppressor cells, natural killer

Tumor Microenvironment-Responsive Nanoparticles for Pancreatic Cancer Imaging and Treatment.

Early diagnosis and effective treatment of pancreatic cancer remain major clinical challenges, one of the most aggressive and lethal tumors. The tumor microenvironment (TME) in pancreatic cancer exhibits unique features, including high interstitial fluid pressure (IFP), abnormal blood vessels, hypoxia, acidosis, and excess reactive oxygen species (ROS), all favoring tumor progression and immunosuppression, meanwhile offsetting diagnostic efficiency and treatment efficacy. Thus, the TME of pancreatic cancer is considered as a target of profound theranostic significance. Nanoparticles (NPs) have…

Ultrasound-activated Piezocatalytic nanomedicine for tumor microenvironment remodeling and immunotherapy.

The physical and immunosuppressive features of the tumor microenvironment limit drug delivery, immune-cell infiltration, and the response of many solid tumors to immunotherapy. Piezocatalytic nanomedicine uses ultrasound to activate piezoelectric nanomaterials, producing charge separation, local electrical effects, and reactive oxygen species within tumors. These effects can induce cellular stress and regulated tumor-cell death and, when associated with immunogenic cell death, promote the release or exposure of damage-associated molecular patterns that support dendritic-cell activation and sub…

Biomimetic nanoimmunotherapy boosts spatiotemporal PANoptosis and reshapes desmoplastic tumor microenvironment.

Dendritic cell (DC)-based vaccines for solid tumors face major challenges, including limited tumor-specific antigens and immunosuppressive stroma. Here, we present a therapeutic nanovaccine (UCNP@MOF@MI@FM [UMMF]) composed of a DC/tumor fused cytomembrane-coated UCNP@MOF nanoparticle, co-loaded with a MutT homolog 1 (MTH1) inhibitor and combined with tetrahydrobiopterin (BH4). The fused membrane facilitates dual targeting to tumors and lymph nodes while enabling broad-spectrum tumor antigen presentation. Upon near-infrared (NIR) irradiation, upconversion-triggered reactive oxygen species (ROS)

Enzyme-Activated Orthogonal Proteolysis Chimeras for Tumor Microenvironment-Responsive Immunomodulation.

Precise modulation of dynamic and complex tumor microenvironment (TME) to disrupt tumorigenesis and reshape intratumoral immune infiltration has emerged as promising approaches for enhanced cancer therapy. Among recent innovations, proteolysis-targeting chimeras (PROTACs) represent a burgeoning chemical knockdown technology capable of degrading oncogenic protein homeostasis and inducing dynamic alternations within carcinoma settings, offering potential for antitumor manipulation. However, achieving selectivity in PROTACs that respond to disease environmental stimulation and precisely perturb o…

Oncolytic virotherapy: Sparking durable anti-tumor immunity through microenvironment modulation.

Oncolytic virotherapy (OVT) is a novel approach to cancer treatment that utilizes viruses to infect and destroy tumor cells selectively. Beyond direct oncolysis, OVT significantly reshapes the tumor microenvironment (TME), activating the adaptive immune system to generate robust and durable anti-tumor immunity. This review examines the diverse mechanisms by which OVT modulates the TME, including physical remodeling, alterations in cellular composition, and the induction of immunogenic cell death, which releases antigens and adjuvants that enhance immune activation. We also explore the synergis…

Communication between gut microbiota-derived metabolites and the tumor microenvironment.

The gut microbiota has been increasingly recognized as a critical player in maintaining human health and influencing disease development. The tumor microenvironment (TME) is pivotal in tumor development and progression, comprising immune cells, stromal elements, extracellular matrix components, and cytokines. Recent studies have highlighted the promising potential of gut microbiota-derived metabolites (e.g., short-chain fatty acids, bile acids, polyamines, and tryptophan derivatives) to reshape the TME in various ways, generating significant interest for the development of novel therapeutic st…

Advances in Tumor Microenvironment and Immunotherapeutic Strategies for Hepatocellular Carcinoma.

Hepatocellular carcinoma (HCC) is a highly aggressive malignancy, largely driven by an immunosuppressive tumor microenvironment (TME) that facilitates tumor growth, immune escape, and resistance to therapy. Although immunotherapy-particularly immune checkpoint inhibitors (ICIs)-has transformed the therapeutic landscape by restoring T cell-mediated anti-tumor responses, their clinical benefit as monotherapy remains suboptimal. This limitation is primarily attributed to immunosuppressive components within the TME, including tumor-associated macrophages, regulatory T cells (Tregs), and myeloid-de…

Unveiling the "Dark Matter" of platelet involvement in tumor microenvironment.

Platelets are well-known for their functions in blood clotting and vascular repair. However, in recent years, the regulatory role of platelets in the occurrence and development of malignant tumors has received significant attention. While extensive research has been conducted on the regulation of tumors by circulating platelets in peripheral blood, there is a lack of coherence and continuity among these studies. The tumor microenvironment encompasses the intricate network of cellular and acellular elements that surround and interact with tumor cells, creating a supportive ecosystem for their s…

Single-Cell View of Tumor Microenvironment Gradients in Pleural Mesothelioma.

Immunotherapies have shown great promise in pleural mesothelioma (PM), yet most patients still do not achieve significant clinical response, highlighting the importance of improving the understanding of the tumor microenvironment (TME). Here, we utilized high-throughput, single-cell RNA sequencing (scRNA-seq) to de novo identify 54 expression programs and construct a comprehensive cellular catalog of the PM TME. We found four cancer-intrinsic programs associated with poor disease outcome and a novel fetal-like, endothelial cell population that likely responds to VEGF signaling and promotes ang…

Natural products reverse cisplatin resistance in the hypoxic tumor microenvironment.

Cisplatin is one of the most commonly used drugs for cancer treatment. Despite much progress in improving patient outcomes, many patients are resistant to cisplatin-based treatments, leading to limited treatment efficacy and increased treatment failure. The fact that solid tumors suffer from hypoxia and an inadequate blood supply in the tumor microenvironment has been widely accepted for decades. Numerous studies have shown that a hypoxic microenvironment significantly reduces the sensitivity of tumor cells to cisplatin. Therefore, understanding how hypoxia empowers tumor cells with cisplatin …

Tumor Microenvironment Remodeling-Mediated Sequential Drug Delivery Potentiates Treatment Efficacy.

Toll-like receptor 7/8 agonists, such as imidazoquinolines (IMDQs), are promising for the de novo priming of antitumor immunity. However, their systemic administration is severely limited due to the off-target toxicity. Here, this work describes a sequential drug delivery strategy. The formulation is composed of two sequential modules: a tumor microenvironment remodeling nanocarrier (poly(l-glutamic acid)-graft-methoxy poly(ethylene glycol)/combretastatin A4, termed CA4-NPs) and an immunotherapy nanocarrier (apcitide peptide-decorated poly(l-glutamic acid)-graft-IMDQ-N3 conjugate, termed apcit…
