Comparing behavior following binge ethanol in adolescent and adult DBA/2 J mice.

The adolescent brain undergoes maturation in areas critically involved in reward, addiction, and memory. Adolescents consume alcohol more than any other drug, typically in a binge-like manner. While adults also binge on alcohol, the adolescent brain is more susceptible to ethanol-related damages due to its ongoing development, which may result in persistent behavioral and physical changes, including differences in myelination in the frontal cortex. Sex also impacts ethanol metabolism and addiction progression, suggesting females are more sensitive than males. This study addressed memory, socia

Allele-Specific Methylation of SPDEF: A Novel Moderator of Psychosocial Stress and Substance Abuse.

OBJECTIVE: Psychosocial stress is a key risk factor for substance abuse among adolescents. Recently, epigenetic processes such as DNA methylation have emerged as potential mechanisms that could mediate this relationship. The authors conducted a genome-wide methylation analysis to investigate whether differentially methylated regions are associated with psychosocial stress in an adolescent population. METHODS: A methylome-wide analysis of differentially methylated regions was used to examine a sample of 1,287 14-year-old adolescents (50.7% of them female) from the European IMAGEN study. The Il

Project Northland: comprehensive alcohol use prevention for young adolescents, their parents, schools, peers and communities.

OBJECTIVE: Project Northland is an ongoing prevention trial with the objective of reducing underage drinking and related problems. Phase I focused on early adolescence and this study describes the multiple interventions, highlighting its parent components. METHOD: A cohort design was used with sixth graders from 24 school districts (N = 2,35 1: 97% of the eligible population: 51.3% boys), randomly assigned to intervention or reference condition. Phase I ended in eighth grade (N = 1,901: 81% retention rate). Both demand and supply reduction guided the interventions. This study examined Project…

Age of alcohol use initiation and psychiatric symptoms among young adult trauma survivors.

Alcohol use disorder (AUD) evidences high rates of comorbidity with a range of psychiatric disorders, particularly within high-risk populations, such as individuals exposed to physical or sexual violence. Increasing efforts are focused on understanding the role of early alcohol use (e.g., during adolescence) on emotional and psychiatric functioning over time, as well as sex differences in these associations. The aim of the current study was to evaluate patterns of association between age of initiation of regular alcohol use and posttraumatic stress disorder (PTSD), anxiety, and depressive symp

Latent classes of substance use precursors predict positive alcohol and cannabis expectancies one year later in Black and Latino early adolescents in the ABCD study.

INTRODUCTION: Precursors of alcohol and cannabis use, such as friend (dis)approval, have rarely been examined in person-centered analyses that can inform tailored prevention, particularly among Black and Latino youth. This study aimed to identify person-centered patterns (classes) of precursors of alcohol and cannabis use in Black and Latino youth, examine correlates across classes, and determine whether the classes predicted positive anticipated effects (expectancies) of alcohol and cannabis one year later. METHODS: Latent class analyses used data from Black and Latino pre-adolescents in the

Alcohol Use Disorder and Parental Alcohol Use Disorder as Predictors of Reproductive Timing.

OBJECTIVE: Research suggests associations between adolescent alcohol use and early reproduction, but other findings show that alcohol use disorder (AUD) may actually predict delayed reproduction. However, most studies generally do not consider the effects of parental AUD, which is correlated with AUD and may influence reproductive timing. The present study addressed these gaps by testing whether the individual's own AUD and parental AUD interacted with sex to predict reproductive timing. METHOD: In a longitudinally followed community sample that oversampled familial alcohol disorder (n = 776)

Substance Use and School Characteristics in Lesbian, Gay, Bisexual, and Heterosexual High School Students.

BACKGROUND: Tobacco, alcohol, and illicit drug use among lesbian, gay, and bisexual (LGB) high school students as well as related environment and school-level risk and protective factors were examined. METHODS: Data was acquired from the 2015 CDC's Youth Behavior Risk Survey for Kentucky (N = 2577). Prevalence of substance use was calculated for all high school respondents by reported sexual orientation. Multivariate analyses estimated the relationship between school and environmental-level factors (eg, having an adult to talk to) and tobacco, alcohol, and illicit drug use via logistic regres

Impact of adolescent alcohol use across the lifespan: Long-lasting tolerance to high-dose alcohol coupled with potentiated spatial memory impairments to moderate-dose alcohol.

Understanding how alcohol exposure during adolescence affects aging is a critical but understudied area. In the present study, male rats were exposed to either alcohol or saline during adolescence, then tested every 4 months following either an ethanol or saline challenge; animals were tested until postnatal day (PD) 532. It was found that long-lasting tolerance to high-dose ethanol exists through the test period, as measured by loss of righting reflex, while tolerance to lower doses of ethanol is not found. In addition, alcohol exposure during adolescence facilitated spatial memory impairment

Screening, brief Intervention, and referral to treatment: Persistence of effects in adolescents at risk.

This observational longitudinal study examines substance use reduction following implementation of screening, brief intervention, and referral to treatment (SBIRT) in a specialized foster care clinic. Following SBIRT we expected to observe reductions in past 30-day use of alcohol, tobacco, and cannabis, sustained to 6 months post-intervention. Adolescents (10-20 years) received standardized screening via the CRAFFT2.1 + N. An interventionist trained in the brief negotiated interview delivered brief intervention. Enrolled participants completed surveys in REDCap that assessed past 30-day use of

Glucocorticoid Receptor (NR3C1) Gene Polymorphism Moderate Intervention Effects on the Developmental Trajectory of African-American Adolescent Alcohol Abuse.

Accumulative evidence from recent genotype × intervention studies suggests that individuals carrying susceptible genotypes benefit more from intervention and provides one avenue to identify subgroups that respond differentially to intervention. This study examined the moderation by glucocorticoid receptor (NR3C1) gene variants of intervention effects on the developmental trajectories of alcohol abuse through adolescence. Participants were randomized into Fast Track intervention and control groups self-reported past-year alcohol abuse annually from grade 7 through 2 years post-high school and p

Not just a label: Teens' perceptions of what elements of cannabis product packaging appeal to underage individuals.

BACKGROUND: As cannabis legalization expands, so does product marketing, raising concerns about its appeal to underage audiences. Although previous studies have focused on marketing violations or adult perceptions of cannabis products, few studies have examined how teens perceive cannabis packaging. METHODS: The current study identifies specific packaging elements in cannabis-infused products (i.e., food and drink edibles) that teens and young adults perceive will appeal to minors through a method that maps visual attention and interest. In 2023, we fielded an online survey of 149 teens (ages

Adolescent binge ethanol-induced loss of basal forebrain cholinergic neurons and neuroimmune activation are prevented by exercise and indomethacin.

Basal forebrain cholinergic neurons mature in adolescence coinciding with development of adult cognitive function. Preclinical studies using the rodent model of adolescent intermittent ethanol (AIE; 5.0 g/kg, i.g., 2-days on/2-days off from postnatal day [P]25 to P55) reveal persistent increases of brain neuroimmune genes that are associated with cognitive dysfunction. Adolescent intermittent ethanol exposure also reduces basal forebrain expression of choline acetyltransferase (ChAT), an enzyme critical for acetylcholine synthesis in cholinergic neurons similar to findings in the post-mortem h

Parents' perspectives and behaviors regarding their child's access to alcohol: Variation by race/ethnicity, socioeconomic status, and neighborhood.

BACKGROUND: Setting rules about alcohol use and minimizing its availability in the home are known effective parent-level strategies for reducing underage drinking risk. However, parents' restrictions and their perceptions of their child's alcohol access have rarely been considered in combination (e.g., determining if rule-setting consistently accompanies perceived easy access), despite the potential to inform targeted prevention. The current study identified patterns in six parent-reported indicators of their child's alcohol restrictions and access and characterized them with respect to race/e…

Effect of comorbid psychologic and somatic symptom trajectories on early onset substance use among U.S. youth in the ABCD study.

BACKGROUND: Adolescent substance use (SU) is often motivated by a desire to alleviate undesirable symptoms. To test the self-medication hypothesis, we examined associations between comorbid psychologic and somatic symptom trajectories across early adolescence and early onset SU. METHODS: Using Adolescent Brain Cognitive Development Study® data, we differentiated youth who reported no SU at baseline based on their comorbid anxiety, depression, pain, somatic and somnolence symptom trajectories. The outcome, early onset SU (by age 13-14 years) was derived from self-reported alcohol (≥full drink)

Progression from First Drink, First Intoxication, and Regular Drinking to Alcohol Use Disorder: A Comparison of African American and European American Youth.

BACKGROUND: Differences between African Americans (AAs) and European Americans (EAs) in the prevalence and age at onset of alcohol use and alcohol use disorder (AUD) have been documented, but distinctions in the timing of early stage transitions and contribution of various psychiatric and psychosocial risk factors to the progression from initiation to AUD have yet to be investigated. The current study characterized progression from alcohol use initiation-defined alternatively as first drink, first intoxication, and regular drinking onset-to AUD in AA and EA youth. METHODS: Psychiatric intervi

Dysregulation of neurotrophin expression in prefrontal cortex and nucleus basalis magnocellularis during and after adolescent intermittent ethanol exposure.

A preclinical model of human adolescent binge drinking, adolescent intermittent ethanol exposure (AIE) recreates the heavy binge withdrawal consummatory patterns of adolescents and has identified the loss of basal forebrain cholinergic neurons as a pathological hallmark of this model. Cholinergic neurons of the nucleus basalis magnocellularis (NbM) that innervate the prefrontal cortex (PFC) are particularly vulnerable to alcohol related neurodegeneration. Target derived neurotrophins (nerve growth factor [NGF] and brain-derived neurotrophic factor [BDNF]) regulate cholinergic phenotype express
