The effects of casein glycomacropeptide on general health status in children with PKU: A randomized crossover trial.

In PKU, it is suggested that casein glycomacropeptide based protein substitute (GMP) may have physiological advantage when satiety, oxidative stress, renal function and inflammation are considered. Its prebiotic properties may also help gastrointestinal (GI) tolerance. In children with PKU, a randomized/crossover trial comparing phenylalanine-free amino acids (AA) vs GMP as the single source of protein substitute for 12-weeks in each arm was conducted. There was a 4-week wash out period with AA in-between. At baseline and end of each intervention, blood and fecal samples were taken to monitor

Evaluation of vaginal microbiome equilibrium states identifies microbial parameters linked to resilience after menses and antibiotic therapy.

The vaginal microbiome (VMB) is a complex microbial community that is closely tied to reproductive health. Optimal VMB communities have compositions that are commonly defined by the dominance of certain Lactobacillus spp. and can remain stable over time or transition to non-optimal states dominated by anaerobic bacteria and associated with bacterial vaginosis (BV). The ability to remain stable or undergo transitions suggests a system with either single (mono-stable) or multiple (multi-stable) equilibrium states, though factors that contribute to stability have been difficult to determine due t

Dietary resistant starch alleviates Escherichia coli-induced bone loss in meat ducks by promoting short-chain fatty acid production and inhibiting Malt1/NF-κB inflammasome activation.

BACKGROUND: Escherichia coli (E. coli) infection in humans and animals usually comes with gut dysbiosis, which is potential culprit to skeletal health, it is still unclear to whether diet interfered gut microbiome changes can be a protective strategy to bone loss development. Here, the effects of resistant starch from raw potato starch (RPS), a type of prebiotic, on E. coli-induced bone loss and gut microbial composition in meat ducks were evaluated. RESULTS: The results showed that dietary 12% RPS treatment improved bone quality, depressed bone resorption, and attenuated the pro-inflammatory

Effect of a Lactobacillus fermentation product on postweaning heifer performance.

The objective of the experiment was to compare the effect of dietary inclusion of a prebiotic fermentation product of Lactobacillus acidophilus (LaP, RumaCell; 5 mL animal-1 d-1) or monensin on performance of replacement beef heifers. Heifers received a total mixed ration containing either LaP (n = 77) or monensin (MON; Rumensin; 200 mg animal-1 d-1; n = 79). Heifers were fed for 71 d in a GrowSafe unit, so individual feed intake could be measured. Heifers were weighed every 2 wk and feed efficiency calculated by residual feed intake (RFI). At the end of the RFI trial, heifers remained on thei

Nutrition and Metabolism: Foundations for Animal Growth, Development, Reproduction, and Health.

Consumption of high-quality animal protein plays an important role in improving human nutrition, growth, development, and health. With an exponential growth of the global population, demands for animal-sourced protein are expected to increase by 60% between 2021 and 2050. In addition to the production of food protein and fiber (wool), animals are useful models for biomedical research to prevent and treat human diseases and serve as bioreactors to produce therapeutic proteins. For a high efficiency to transform low-quality feedstuffs and forages into high-quality protein and highly bioavailable

Evolutionary Aspects of the Oxido-Reductive Network of Methylglyoxal.

In the chemoautotrophic theory for the origin of life, offered as an alternative to broth theory, the archaic reductive citric acid cycle operating without enzymes is in the center. The non-enzymatic (methyl)glyoxalase pathway has been suggested to be the anaplerotic route for the reductive citric acid cycle. In the recent years, much has been learned about methylglyoxal, but its importance in the metabolic machinery is still uncovered. If methylglyoxal had been essential participant of the early stage of evolution, then it is a legitimate question whether it might have played a role in the ea

Restraint stress induced gut dysmotility is diminished by a milk oligosaccharide (2'-fucosyllactose) in vitro.

BACKGROUND: Stress causes severe dysmotility in the mammalian gut. Almost all research done to date has concentrated on prevention of stress-induced altered gut motility but not on treatment. We had previously shown that intraluminal 2'FL could acutely moderate propulsive motility in isolated mouse colonic segments. Because 2'FL appeared to modulate enteric nervous system dependent motility, we wondered if the oligosaccharide could reverse the effects of prior restraint stress, ex vivo. We tested whether 2'FL could benefit the dysmotility of isolated jejunal and colonic segments from animals s

Galacto-oligosaccharides ameliorate polystyrene nanoplastic-induced anxiety- and depression-like behaviors via a gut-initiated serotonergic cascade.

As emerging environmental contaminants, micro- and nanoplastics (MNPs) raise neurotoxic concerns. However, the mechanisms underlying their induction of emotional disorders remain poorly understood. In particular, intervention-oriented and functionally validated strategies for prevention and control are still lacking. To address this, we conducted a 28-day repeated oral exposure study in mice using a gradient of polystyrene nanoplastics (PS-NPs; 2, 10, and 50 mg/kg/day) spanning environmentally relevant doses, systematically assessing impacts along the microbiota-gut-brain axis. Our results sho

Effects of colloidal delivery systems for curcumin-Brassica rapa L. polysaccharide mixture encapsulation on physicochemical properties, stability, and gut microbiota modulation.

The therapeutic potential of curcumin is severely constrained by its poor physicochemical stability and low oral bioavailability. Co-formulation with Brassica rapa L. polysaccharide offers a promising strategy to enhance its functionality; however, the impact of different colloidal delivery systems on the encapsulation performance and subsequent biological fate of this mixture remains unclear. In this study, we systematically compared three spray-dried delivery platforms, including liposomes (LP-CP), sodium caseinate nanoparticles (SC-CP), and β-cyclodextrin inclusion complexes (CYC-CP), for e

Bletilla striata oligosaccharides alleviate high-fat diet-induced metabolic associated fatty liver in mice through modulation of gut microbiota and host metabolism.

BACKGROUND AND AIM: Gut-liver axis dysfunction drives metabolic associated fatty liver disease (MAFLD), but effective therapeutic strategies remain limited. Bletilla striata oligosaccharides (BSO) have immunomodulatory potential, yet their role in MAFLD via the gut-liver axis is unclear. This study aimed to investigate whether and how BSO ameliorates MAFLD by modulating gut microbiota, intestinal barrier function, and hepatic inflammation. RESULTS: MAFLD was induced in mice by 8-week high-fat diet followed by 12-week BSO (150, 300, 600 mg/kg) or metformin treatment via oral gavage. Compared w

The interplay between probiotics and host autophagy: mechanisms of action and emerging insights.

Autophagy, a lysosome-dependent protein degradation mechanism, is a highly conserved catabolic process seen in all eukaryotes. This cell protection system, which is present in all tissues and functions at a basic level, can be up- or downregulated in response to various stresses. A disruption in the natural route of the autophagy process is frequently followed by an interruption in the inherent operation of the body's cells and organs. Probiotics are live bacteria that protect the host through various mechanisms. One of the processes through which probiotics exert their beneficial effects on v

Citrus tangerine pith extract alleviates hypoxia-induced ileum damage in mice by modulating intestinal microbiota.

Visitors to high altitude are susceptible to hypoxia-induced acute intestinal mucosal barrier injury and severe gastrointestinal disorders, which are life-threatening. Citrus tangerine pith extract (CTPE) is rich in pectin and flavonoids and has been proved to enhance intestinal health and improve gut dysbiosis. In this study, we aim to explore the protective effect of CTPE on ileum injury induced by intermittent hypobaric hypoxia in a mouse model. Balb/c mice were divided into blank normoxia (BN), blank hypobaric hypoxia (BH), hypobaric hypoxia plus CTPE (TH), and hypobaric hypoxia plus Rhodi

Metabolomics analysis of the serum metabolic signature of nonalcoholic fatty liver disease combined with prediabetes model rats after the intervention of Lycium barbarum polysaccharides combined with aerobic activity.

Metabolic disorders accompany nonalcoholic fatty liver disease (NAFLD), associated with prediabetes. Lycium barbarum polysaccharides (LBP) seem to be a potential prebiotic, and aerobic exercise has shown protective effects on NAFLD with prediabetes. However, their combined effects on NAFLD and prediabetes remain unclear. This study investigated the effects of LBP and aerobic exercise alone, and their combined effects on the metabolomics of serum, and explored the potential mechanisms utilizing a high-fat diet-induced rat model of NAFLD and prediabetes. It provided the metabolic basis for the p

Research Note: Xylooligosaccharide directly attenuates Salmonella Typhimurium colonization and its induction of impairments in intestinal barrier and growth performance of broilers.

Xylooligosaccharide (XOS) is known as a prebiotic, however, it is unknown whether XOS can directly protect against bacterial infection. This study aimed to investigate the direct inhibitory effects of XOS on Salmonella Typhimurium colonization and the inductive impairments in gut health and growth performance in broilers. We first probed the inhibitory effects of XOS on S. Typhimurium adhesion and its induction of intestinal epithelial cell (IPEC-J2) injuries. Afterward, 168 one-day-old yellow-feathered broilers were randomly divided into 3 groups (7 replicates/group): negative control (NC, re
