Attenuation of perfluoropolymer fume pulmonary toxicity: effect of filters, combustion method, and aerosol age.

Thermal decomposition products of some perfluorinated polymers are toxic to experimental animals in small-scale combustion toxicity tests; the toxicity is dependent upon the heating procedure, combustion temperature, and other experimental conditions. In the current studies we investigated the time course of fume generation and exposure on pulmonary effects in rats following a 30-min exposure to perfluoropolymer decomposition products (i.e., fume concentration = 0.2 mg/m3 of tetrafluoroethylene/hexafluoropropylene copolymer (FEP)) pyrolyzed with either static or dynamic airflows. In the first

Oxidative defluorination of 1,1,1,2-tetrafluoroethane by rat liver microsomes.

1,1,1,2-Tetrafluoroethane (R-134a) is a non-ozone-depleting alternative to dichlorodifluoromethane for use as an air-conditioning refrigerant and as a propellant in anti-asthmatic and other pharmaceutical preparations. Hepatic microsomes, supplemented with NADPH, catalyzed the release of F- from R-134a; metabolite production was positively correlated with both duration of incubation and gas phase [R-134a]. Defluorination of R-134a was inhibited by CO, lack of NADPH, or heat denaturation of microsomes. Release of F- from R- 134a biotransformation as shown by the near-total lack of dehalogenatio

Ex vivo autoradiography of the human gastrointestinal tract: a new approach to cell kinetic studies of surgically removed tumor-bearing organs.

Surgically obtained portions of the human gastrointestinal tract were perfused with oxygenated perfluorochemical blood substitute, Fluosol-DA (FDA), containing tritiated thymidine. This procedure of ex vivo autoradiography was performed at the RI Center by perfusion of the isolated organ with FDA from a reservoir 190 cm above the organ through a cannula inserted into the main artery. Microautoradiographic sections revealed that the distributions of labeled nuclei in the normal epithelium of the esophagus, stomach, jejunum, colon and rectum coincided well with those reported previously. However

Metal fume fever and polymer fume fever.

INTRODUCTION: Inhalational exposure to metal-containing fumes generated by welding and related processes may result in the development of the clinical syndrome known as "metal fume fever." Polymer fume fever is a separate and distinct but related disorder that has been associated with inhalational exposure to specific fluorinated polymer products, such as polytetrafluoroethylene or Teflon(®). We undertook a review of the peer-reviewed medical literature as it relates to these two disease entities in order to describe their epidemiology, pathophysiology, clinical presentation, diagnosis, treatm

PFOS increases cellular uric acid content and the abundance of URAT1 and GLUT9 in HK-2 cells.

Perfluorooctane sulfonate (PFOS) is a persistent environmental pollutant associated with hyperuricemia. How PFOS affects urate-related responses in human proximal-tubule cells remains incompletely understood. We examined cellular uric acid content and URAT1/GLUT9 protein abundance in PFOS-treated HK-2 cells, with benzbromarone as a pharmacological comparator. Network analysis, molecular docking, molecular dynamics simulations and public mouse kidney single-cell RNA sequencing provided complementary candidate and cell-type information. PFOS increased cellular uric acid content under exogenous-u

Exposure to PFOA and GenX elicits cell typespecific impacts on p53 and TGF-β signaling pathways.

Per- and polyfluoroalkyl substances (PFAS) are a class of synthetic chemicals extensively used as plastic additives. Their environmental persistence and potential for bioaccumulation have raised significant toxicological concerns. This study evaluates the cytotoxic and molecular effects of Perfluorooctanoic acid (PFOA) and its replacement compound, Perfluoro(2-methyl-3-oxohexanoic) acid (GenX), in human-derived skin (A375), liver (HepG2), kidney (SN12C), and colon (SW620) cell lines. The experimental design assessed cell viability, gene expression, and perturbations in key cellular stress path

Female reproductive toxicity of chronic exposure to perfluorohexane sulfonate (PFHxS) under the confounding factor of dietary protein deficiency.

Per- and polyfluoroalkyl substances (PFHxS) is a perfluoroalkyl substance (PFAS). PFASs are ubiquitous and persistent, due to their bioaccumulative tendency, posing long-term health-related concerns. Human exposure to PFHxS is mainly via contact and ingestion. Diet and environmental exposure to toxicants significantly affect fertility outcomes. Nutrition is also a confounding factor in the toxicity of drugs and chemicals. In this study, we investigated the effect of chronic exposure to PFHxS under low protein diet. A low-protein diet combined with exposure to PFHxS for 90 days via drinking wat

Associations of prenatal PFAS exposure with offspring neurobehavioral outcomes by maternal folate supplementation and cord blood 5-MTHF levels: a prospective birth cohort study.

While prenatal exposure to per- and polyfluoroalkyl substances (PFAS) has been associated with adverse neurodevelopment, epidemiological evidence on the modifying role of maternal folate remains limited. This study included 1267 mother-child pairs from the Shanghai Birth Cohort. We measured ten PFAS in maternal plasma and 5-methyltetrahydrofolate (5-MTHF) in cord blood red blood cells (RBCs). Data on maternal folic acid supplementation during early and late pregnancy was collected using questionnaires. Offspring neurodevelopment was assessed at ages 2 and 4 years using the Bayley Scales of Inf

Radial Hadamard-encoded 19F-MRI.

OBJECTIVES: Developing a 19F imaging method to acquire images of the molecular inflammation tracer perfluorooctyl bromide (PFOB) without chemical shift artifacts. MATERIALS AND METHODS: PFOB is a molecular tracer that can be used to track the response of myeloid cells. However, imaging of PFOB with 19F-MRI is challenging due to its complex spectrum which leads to unwanted chemical shift artifacts. Spectral HE allows for separate reconstructions of each peak of the PFOB spectrum, which was combined into a single image after resonance shift correction. In this work, a Hadamard-encoded (HE) radi

Bisphenol AF Induced Neurodevelopmental Toxicity of Human Neural Progenitor Cells via Nrf2/HO-1 Pathway.

Bisphenol AF (BPAF) is widely utilized as an analog of bisphenol A (BPA) in the plastics industry. However, there is limited evidence on its neurodevelopmental toxicity. Existing studies suggest that BPAF has greater accumulation in vivo than other bisphenol analogs, and could pass through the placental barrier and the blood-brain barrier. In this study, we used the human neural progenitor cells line ReNcell CX, which was derived from 14-week human cortical brain tissue, as an in vitro model to investigate the neurodevelopmental toxicity effects of BPAF and BPA on ReNcell CX cells, and explore

Maternal, fetal, and neonatal toxicity and potency estimates of perfluorohexane sulfonate (PFHxS) from oral maternal exposure in the Sprague-Dawley rat.

Perfluorohexane sulfonate (PFHxS) is a legacy, long-chain per- and polyfluoroalkyl substance (PFAS) frequently detected in human serum and environmental media across the globe. Previously published developmental studies in rats indicated a general lack of effects in common endpoints like maternal and offspring body and liver weights at doses up to 50 mg/kg/d. To facilitate mixture-based studies of PFAS co-exposure, individual dose response parameters are necessary for model predictions. To address these data needs we exposed timed-pregnant Sprague-Dawley rat dams to PFHxS (3-125 mg/kg/d) via o

Bile acids and apical sodium-dependent bile acid transporter impact per- and polyfluoroalkyl substances absorption: Evidence from in vivo and cross-sectional studies.

Per- and polyfluoroalkyl substances (PFAS) are synthetic chemicals with worldwide contamination and human exposure. Epidemiological studies have linked high PFAS levels to various diseases, highlighting their potential health risks. However, current studies often overlook the possibility that diseases may influence PFAS levels in patients, potentially overestimating or underestimating their true public health impacts. Here, we found that a high-fat diet, which increases bile acids secretion, decreased the area under the curve (AUC) of nearly all target PFAS in rats, with about 50 % of these de

Paternal and maternal exposures to per- and polyfluoroalkyl substances (PFAS) and birth outcomes: a multi-country cohort study.

BACKGROUND: Maternal prenatal exposures to per- and polyfluoroalkyl substances (PFAS) have been linked to adverse birth outcomes. However, few investigations have considered paternal PFAS exposure. We estimated the parent-specific associations of prenatal PFAS exposures with adverse birth outcomes. METHODS: This study included 498 couples from the INUENDO cohort recruited at antenatal care visits in Greenland, Poland, and Ukraine during 2002-2004. We measured five major types of PFAS in parental serum during pregnancy. We analyzed three birth outcomes ascertained from medical records, includi

Differential hepatic activation of mouse and human peroxisome proliferator-activated receptor-α by perfluorohexane sulfonate.

Exposure of perfluorohexane sulfonate (PFHxS) is associated with hepatomegaly and accumulation of lipids that may be mediated by nuclear receptors like peroxisome proliferator-activated receptor-α (PPARα), constitutive androstane receptor (CAR), or pregnane X receptor (PXR). This study tested the hypotheses that: (i) PFHxS causes changes in liver by activating PPARα, CAR, or PXR, and (ii) there is a species difference in PPARα activity by PFHxS. Wild-type, Ppara-null, and PPARA-humanized mice were fed either a control diet, or one containing 2.2 mg PFHxS/kg diet or 25.8 mg PFHxS/kg diet for ei

Exposure to per- and polyfluoroalkyl substances during fetal development and risk of testicular germ cell cancer in adulthood.

BACKGROUND: Testicular germ cell cancer (TGCC) originates during fetal life. Fetal exposure to environmental chemicals may contribute to its development, but epidemiological data are lacking. We investigated per- and polyfluoroalkyl substances (PFAS), which can act as endocrine disruptors during fetal development, and TGCC risk in adulthood. METHODS: We conducted a nested case-control study of 549 mother-male offspring pairs (103 TGCC cases, 446 matched controls). The source population included over 100,000 pregnant women with biobanked serum samples collected during 1985-1994, a period befor

BACKGROUND: Prenatal exposure to per- and polyfluoroalkyl substances (PFASs) influences neurodevelopment. Thyroid homeostasis disruption is thought to be a possible underlying mechanism. However, current epidemiological evidence remains inconclusive. OBJECTIVES: This study aimed to explore the effects of prenatal PFAS exposure on the intelligence quotient (IQ) of school-aged children and assess the potential mediating role of fetal thyroid function. METHODS: The study included 327 7-year-old children from the Sheyang Mini Birth Cohort Study (SMBCS). Cord serum samples were analyzed for 12 PF
