Protease inhibitors: a panacea?

With the increasing evidence of protease involvement in several diseases, novel strategies for drug development involve the use of protease inhibitors (PIs). The local balance between protease inhibitors and proteases is an important determinant of the occurrence and progression of a particular disease. Hence, enzymes and their cognate inhibitors are finding their applications as diagnostic and prognostic markers. PIs are widely implicated for their use in host defense against infection, tissue repair and matrix production, blood coagulation, cancer, and they are, therefore, the current focus

Influence of helminth infections on childhood nutritional status in lowland Bolivia.

Infectious disease, such as diarrheal disease, respiratory infections, and parasitic infections, are an important source of nutritional and energetic stress in many populations. Inspired by the research and methodological innovations of A. Roberto Frisancho, this work considers the impact of childhood environment and local disease ecology on child health and nutritional patterns among an indigenous group in lowland Bolivia. Specifically, we examine the association between soil-transmitted helminth infection, especially hookworm species, and anthropometric markers of short- and long-term nutrit

Coinfections with Schistosoma haematobium, Necator americanus, and Entamoeba histolytica/Entamoeba dispar in children: chemokine and cytokine responses and changes after antiparasite treatment.

The effect of polyparasite infections on cytokine and chemokine responses as well as the effect of antiparasite treatment was studied in children without parasite infection (the G0 group), in children singly infected with Schistosoma haematobium (the G1 group), and in children multiply infected with S. haematobium/Schistosoma mansoni, Entamoeba histolytica/Entamoeba dispar, and Necator americanus (the G3+ group). Linear regression analysis disclosed a significant risk for coinfection with hookworm and Schistosoma species. Polyparasite infections detected in 23% of children before treatment wer

The burden of polyparasitism among primary schoolchildren in rural and farming areas in Zimbabwe.

A cross-sectional study was conducted in Zimbabwe among 1303 primary schoolchildren from a rural (53.3%) and a commercial farming area (46.7%) to determine the prevalence of co-infection by helminths and Plasmodium falciparum. Urine was examined on three successive days using the filtration method. Two stool specimens were processed using the Kato-Katz method and a third specimen was processed using the sedimentation method. Plasmodium falciparum was diagnosed from thick blood films. The prevalence of Schistosoma haematobium in the rural and farming areas was 66.8% and 52.3%, respectively, and

Plasmodium falciparum and helminth coinfection in a semi urban population of pregnant women in Uganda.

BACKGROUND: Helminth infections and malaria are widespread in the tropics. Recent studies suggest helminth infections may increase susceptibility to Plasmodium falciparum infection. If confirmed, this increased susceptibility could be particularly important during pregnancy-induced immunosuppression. OBJECTIVE: To evaluate the geographical distribution of P. falciparum-helminth coinfection and the associations between P. falciparum infection and infection with various parasite species in pregnant women in Entebbe, Uganda. METHODS: A cross-sectional study was conducted at baseline during a tr

Intestinal parasite infections and fecal steroid levels in wild chimpanzees.

Immune-endocrine interactions have been evaluated much less frequently in nonhuman primates, and this may be due, in part, to logistical and ethical concerns regarding trapping and sampling of endangered species, especially apes. Using noninvasive fecal collection methods, the present study evaluates possible relationships between fecal steroid levels and gastrointestinal parasite infections in the Ngogo chimpanzee community in Kibale National Park, Uganda. Because both testosterone and cortisol exhibit immunosuppressive effects in vitro and in other animal models, it was hypothesized that bot

[Intestinal parasites in dogs and cats with gastrointestinal symptoms in Santiago, Chile].

BACKGROUND: There is an increasing importance of pet-transmitted infections, some of those are considered emerging infections. AIM: To determine the species and frequency of intestinal parasites in pets with diarrhea (hemorrhagic gastroenteritis excluded). MATERIAL AND METHODS: A descriptive retrospective study. Fecal samples from 972 dogs and 230 cats consulting in two veterinary practices in Santiago, between 1996 and 2003, were studied using Burrows' technique. RESULTS: Protozoa were found in 64.8% of dogs and in 66.5% of cats; helminthes in 24% of dogs and 45.2% of cats. The species fou

Perforation of Meckel's diverticulum by roundworm.

An 11-month-old girl child presented with distension of abdomen, constipation, vomiting and fever. X-ray of the abdomen suggested intestinal obstruction. Exploration revealed the distal ileum to be kinked around a band running from the tip of a congested Meckel's diverticulum to the lateral pelvic wall. A small perforation was noticed at the tip of the diverticulum and a live roundworm was found lying free in the peritoneal cavity. The diverticulum along with a segment of adjoining bowel was resected and end-to-end anastomosis done. The diverticulum had a wide lumen and showed no heterotopic t

Effect of inducible costimulator blockade on the pathological and protective immune responses induced by the gastrointestinal helminth Trichinella spiralis.

Infections with gastrointestinal helminths elicit potent Th2 responses, which ultimately result in their expulsion. However, during expulsion of Trichinella spiralis this Th2 response also induces a severe enteropathy characterized by villus atrophy and crypt hyperplasia. Inducible costimulator (ICOS), a homologue of CD28, interacts with B7-related protein 1, and has been shown to be important in T-B cell interactions and antibody class switching. Significantly, ICOS appears to be involved in the induction of both Th1 and Th2 responses, but may be of heightened importance in Th2 responses. Her

Aggregation of helminths: the role of feeding behavior of fish hosts.

Individual Arctic charr (Salvelinus alpinus) from Fjellfrøsvatn, northern Norway, could be categorized by their stomach contents as zooplanktivores or benthivores. Feeding specialization among these fish was evident from negative correlations between helminths transmitted by pelagic copepods (Diphyllobothrium dendriticum and D. ditremum) and those transmitted by the benthic amphipod Gammarus lacustris (Cystidicola farionis and Cyathocephalus truncatus). Occurrences of parasite species acquired from the same types of invertebrate were positively correlated in the fish. Strong relationships amon

Geo-helminth infections in a rural area of Sri Lanka.

School children carry the heaviest burden of morbidity due to intestinal helminth infection. The objective of this investigation was to study geo-helminth infections in 349 school children aged 6 to 13 years living in a rural area of Sri Lanka. Stool samples were examined by direct saline smear in an initial survey to determine the prevalence of intestinal parasitic infections and thereafter the children were followed up over a two year period with cross sectional surveys of stool samples being carried out at yearly intervals. Following collection of a stool sample, all the subjects were treat

Intestinal nematodes, Toxocara infection, and pyogenic liver abscess in children: a possible association.

Eosinophilia and intestinal helminths are frequent in children with pyogenic liver abscess (PLA) diagnosed in Vitória (Brazil). For these reasons we hypothesized that nematode infection with larvae migrating through the tissues (Ascaris, Strongyloides, Ancylostoma, Necator and Toxocara) may be a predisposing factor for PLA in children. We compared the prevalence of intestinal helminth and Toxocara infection between children with PLA and a control group of children with no evidence of PLA at the Children's Hospital N.S. da Glória in Vitória. From October 1996 to February 1998, 13 children with

Parasites in cultured and feral fish.

Parasites, causing little apparent damage in feral fish populations, may become causative agents of diseases of great importance in farmed fish, leading to pathological changes, decrease of fitness or reduction of the market value of fish. Despite considerable progress in fish parasitology in the last decades, major gaps still exist in the knowledge of taxonomy, biology, epizootiology and control of fish parasites, including such 'evergreens' as the ciliate Ichthyophthirius multifiliis, a causative agent of white spot disease, or proliferative kidney disease (PKD), one of the most economically

Immune responses in parasitic diseases. Part B: mechanisms.

A number of host defenses provide variable resistance against parasites. From the biological point of view, invading parasites must not eliminate the susceptible host population; therefore, antiparasite immunity plays an essential role in limiting the invasion and proliferation of parasites. Several differences exist between immune responses to parasites and immune responses to other microorganisms. Effector mechanisms include T lymphocyte-mediated inflammatory granuloma formation and encapsulation involving the deposition of fibrous tissue. In some instances, passive immunity can be transferr
