Betulin alleviates on myocardial inflammation in diabetes mice via regulating Siti1/NLRP3/NF-κB pathway.

The aim of this study is to study the effect of betulin (BE) on myocardial injury in diabetic mice. Insulin-related indexes and inflammation-related cytokines are detected by commercial kits. The mechanism of BE on diabetic myocardial injury was studied by modern molecular biology techniques. BE significantly improved glocose tolerance, reduced lipid accumulation and reduced the content of inflammatory cytokines in diabetic mice. Furthermore, BE regulated Siti1/NLRP3/NF-κB signaling pathway in db/db mice and H9C2 cells. Siti1 inhibitor (EX-57) counteracted those changes. BE significantly prote

A novel semisynthetic inhibitor of the FRB domain of mammalian target of rapamycin blocks proliferation and triggers apoptosis in chemoresistant prostate cancer cells.

The mammalian target of rapamycin (mTOR) is a key regulator of cell growth and its uncontrolled activation is a hallmark of cancer. Moreover, mTOR activation has been implicated in the resistance of cancer cells to many anticancer drugs, rendering this pathway a promising pharmacotherapeutic target. Here we explored the capability of a semisynthetic compound to intercept mTOR signaling. We synthesized and chemically characterized a novel, semisynthetic triterpenoid derivative, 3-cinnamoyl-11-keto-β-boswellic acid (C-KβBA). Its pharmacodynamic effects on mTOR and several other signaling pathway

Astragaloside IV improves homocysteine-induced acute phase endothelial dysfunction via antioxidation.

Astragaloside IV, the major active component extracted from Astragalus membranaceus, exerts multipotent activities under pathophysiological conditions. Hyperhomocysteinemia, an independent risk factor for cardiovascular disease, induces oxidative stress leading to endothelial dysfunction. We investigated the effect of astragaloside IV on acute phase endothelial dysfunction induced by homocysteine. In a concentration-dependent manner, endothelial dysfunction was induced by homocysteine. In organ bath experiment using rat aortic rings, treatment with astragaloside IV resulted in an improvement o

Inhibition of allergen-induced eosinophil recruitment by natural tetranortriterpenoids is mediated by the suppression of IL-5, CCL11/eotaxin and NFkappaB activation.

The present study reports the anti-allergic activity of a group of six different tetranortriterpenoids (TNTP) isolated from the seeds of Carapa guianensis Aublet: 6a-acetoxygedunin, 7-deacetoxy-7-oxogedunin, andirobin, methyl angolensate, 6a-acetoxyepoxyazadiradione and gedunin. Oral pretreatment with TNTP significantly inhibited total leukocyte and eosinophil accumulation in C57BL/10 mice pleural cavities 24 h after the intrathoracic (i.t.) injection of ovalbumin (OVA), but had no effect on CD4, CD8 or gammadelta T lymphocyte accumulation. Pleural washes recovered from 6 h OVA-stimulated mice

Antiproliferative effects of pachymic acid on thyroid cancer cells and characterization of its metabolites.

Thyroid cancer (THCA) is the most prevalent endocrine malignancy, underscoring the need for novel therapies. Pachymic acid (PA), a key bioactive triterpenoid from P. cocos, possesses documented anticancer activity, yet its role in thyroid cancer is poorly understood. In this study, we evaluated PA's effects on THCA in vitro and in vivo. Our results revealed that PA potently suppressed THCA cell proliferation, migration and tumor growth. Metabolite profiling identified seven metabolites and their formation pathways. Network pharmacology predicted 147 therapeutic targets, primarily enriched in P
