The biological effects of Latrodectus spider venom are similar in animals from different phyla, but these symptoms are caused by distinct phylum-specific neurotoxins (collectively called latrotoxins) with molecular masses ranging from 110 to 140 kDa. To date, the venom has been found to contain five insecticidal toxins, termed alpha, beta, gamma, delta and epsilon-latroinsectotoxins (LITs). There is also a vertebrate-specific neurotoxin, alpha-latrotoxin (alpha-LTX), and one toxin affecting crustaceans, alpha-latrocrustatoxin (alpha-LCT). These toxins stimulate massive release of neurotransmit

Soil microbes are a major food source for free-living soil nematodes. It is known that certain soil bacteria have evolved systems to combat predation. We identified the nematode-antagonistic Pseudomonas protegens strain 15G2 from screening of microbes. Through protein purification we identified a binary protein, designated Pp-ANP, which is responsible for the nematicidal activity. This binary protein inhibits Caenorhabditis elegans growth and development by arresting larvae at the L1 stage and killing older-staged worms. The two subunits, Pp-ANP1a and Pp-ANP2a, are active when reconstituted fr

Protein kinase C (PKC), which is involved in cell signaling pathways, comprises a family of serine/threonine kinases ubiquitously present in animals and its members are grouped on the basis of structural and activation characteristics into novel, classical, and atypical PKC forms. In this study, an atypical PKC of Macrobrachium rosenbergii, designated MraPKC, was successfully cloned, and its protein comprised structural domains similar to those of atypical PKC homologues, including the Phox and Bem1 (PB1) domain, a zinc finger phorbol-ester/DAG-type signature, protein kinase signatures, and a

The full-length human renal mineralocorticoid receptor (hMR) has been overproduced in Spodoptera frugiperda (Sf9) insect cells using baculovirus-mediated expression. The overproduced hMR binds aldosterone with high affinity (Kd = 1.36 nM) and has high affinity for cortisol, cortexolone, and progesterone. Immunoprecipitation and immunoblot analysis of the recombinant hMR with MR-specific antibodies reveal three major protein bands with molecular masses of 115, 119, and 125 kDa. hMR isoforms show maximal accumulation at 48 h post-infection with the recombinant baculovirus. Maximal aldosterone bi

BACKGROUND: Malaria is one of the most important public health problems in Southeast Asia, including Hainan Island, China. Vector control is the main malaria control measure, and insecticide resistance is a major concern for the effectiveness of chemical insecticide control programs. The objective of this study is to determine the resistance status of the main malaria vector species to pyrethroids and other insecticides recommended by the World Health Organization (WHO) for indoor residual sprays.
METHODS: The larvae and pupae of Anopheles mosquitoes were sampled from multiple sites in Hainan

A series of fluorine-containing pyrazole benzamide derivatives were designed and synthesized. Compound Z1 exhibited significantly higher activity against P. xylostella (LC50 = 0.203 mg/L) than that of ethiprole (LC50 = 2.88 mg/L). Compounds Z1, Z8, and Z20 showed over 13-fold higher activity against S. frugiperda (LC50 = 3.38, 2.72, and 2.18 mg/L) than ethiprole (LC50 = 53.6 mg/L). Furthermore, compounds Z1, Z4, and Z20 exhibited good activity against O. furnacalis (LC50 = 4.21, 3.20, and 3.30 mg/L, respectively), with over 7-fold higher potency than ethiprole (LC50 = 30.8 mg/L). The extensive application of meta-diamide insecticides has raised serious concerns regarding their toxicity to aquatic nontarget organisms, necessitating the development of safer alternatives. In this study, a combined in silico approach integrating a scaffold-hopping strategy and three-dimensional quantitative structure-activity relationship (3D-QSAR) models was employed to discover novel meta-diamide derivatives as GABA receptor-targeted insecticides with improved safety profiles. Reliable 3D-QSAR models were constructed based on meta-diamide derivatives developed through a scaffold-hopping

Protease inhibitors (PIs) are promising bioactive compounds for sustainable pest management because they interfere with the digestive processes of herbivorous insects. However, the physiological mechanisms underlying insect adaptation to natural and synthetic PIs remain poorly understood. Here, we investigated the midgut responses of Anticarsia gemmatalis Hübner (Lepidoptera: Erebidae) to the soybean Kunitz trypsin inhibitor (SKTI) and the synthetic tripeptide GORE-2 using integrated transcriptomic, biochemical, and histological approaches, complemented by an exploratory metatranscriptomic ana

Entomopathogenic fungi can manipulate host behavior to promote infection success, but the molecular effectors remain largely unknown. Here we show that Metarhizium anisopliae suppresses feeding in the migratory locust Locusta migratoria by downregulating an antenna-enriched odorant-binding protein gene, LmOBP2. Metabolomic profiling of infected hemolymph identified infection-associated increases in indole, taurine, DL-serine and dimethylglycine; injection or exposure to these metabolites reduced antennal LmOBP2 transcript abundance. Recombinant LmOBP2 bound key corn volatiles (e.g., leaf alcoh

During metamorphic development from larva to adult, insect brains exhibit cell proliferation; however, the molecular mechanisms governing this process remain poorly understood. Employing the lepidopteran insect Helicoverpa armigera (cotton bollworm), an agricultural pest, as a model system, this study demonstrates that the steroid hormone 20-hydroxyecdysone (20E) regulates neural cell proliferation during imaginal brain development by upregulating the expression of SOX12 and DELTA via nuclear receptor EcRA. SOX12 exhibits significantly elevated expression and is localized in the brain during m

Megoura crassicauda, a sap-feeder on legumes, remains a persistent threat, yet current control depends largely on neonicotinoids that pose serious risks to beneficial pollinators. We herein describe a set of new pyrido[1,2-a]pyrimidinone derivatives bearing a carbamate moiety attached via a methylene spacer at the 2-position. Insecticidal bioassays identified H3 as the most potent analogue against M. crassicauda, comparable to the commercial standard TFM. Encouragingly, H3 exhibited remarkably low toxicity to honeybees, with acute oral and contact LD50 values of 24.0 and 35.1 μg a.i./bee, resp

For many insects, the ecdysteroid biosynthetic pathway that converts cholesterol to ecdysone includes multiple cytochrome P450 monooxygenases, a Rieske oxygenase, and a short-chain dehydrogenase. Collectively, the genes encoding these enzymes are referred to as Halloween genes. While the importance of these enzymes in oogenesis is well-documented in holometabolous insects, less is known about their oogenic role in hemipterans. To address this, we used RNA interference-mediated knockdown to determine the role that the Halloween transcripts neverland, spookiest, shroud, phantom, disembodied, sha

Hymenopteran venoms contain diverse proteins and peptides that shape envenomation, defense, predation, and allergic responses. Honeybee venom from Apis mellifera is well characterized, while molecular resources for the Asian giant hornet Vespa mandarinia remain less curated. We compared protein- and peptide-fraction LC-MS/MS identification datasets from V. mandarinia venom (VM-V) and A. mellifera venom (AM-V), supported by a de novo V. mandarinia transcriptome-derived database. Protein-level identification yielded 197 protein groups in VM-V PRO and 164 protein groups in AM-V PRO. AM-V containe

Wasp venoms possess complex compositions and diverse bioactivities, making them potential pharmacological sources. In this study, venoms from four wasp species (Vespa mandarinia, V. velutina, V. basalis, and Provespa barthelemyi) were collected by electrical stimulation and analyzed using liquid chromatography-tandem mass spectrometry. A total of 681 peptides were identified, nearly 90% of which had not been previously reported. Comparative analyses revealed pronounced species-specific signatures at both peptide and peptide family levels. Sequence-based analyses indicated that peptide release

Laccase2 is a multicopper oxidase that plays a central role in insect cuticle tanning and sclerotization, yet its molecular characteristics and biological functions remain poorly understood in the edible beetle superworm Zophobas morio. In this study, the laccase2 homolog, Zmlaccase2, was identified and functionally characterized in Z. morio. Sequence analysis revealed conserved multicopper oxidase domains and copper-binding motifs typical of insect laccase2 orthologs. Expression profiling showed that Zmlaccase2 is dynamically expressed from embryogenesis to adulthood, with expression peaks co

BACKGROUND: Achieving malaria eradication by 2050 will require the development of novel transmission-blocking strategies alongside existing and emerging control measures. Since the innate immune responses of Anopheles salivary glands determine its vectorial capacity, a detailed assessment of vector-parasite interactions could help identify novel targets that play key roles in the immune response against Plasmodium. In this study, six candidate immune-related genes from Anopheles stephensi salivary gland transcriptomic datasets were selected, and their expression changes were assessed following

The Plasmodium infection cycle in mosquitoes relies on numerous host factors in the vector midgut, which can be targeted with therapeutics. The mosquito prefoldin complex is needed to fold proteins and macromolecular complexes properly. Here we show that the conserved Anopheles mosquito prefoldin (PFDN)-chaperonin system is a potent transmission-blocking target for multiple Plasmodium species. Silencing any prefoldin subunit or its CCT/TRiC partner via RNA interference reduces Plasmodium falciparum oocyst loads in the mosquito midgut, as does co-feeding mosquitoes with PFDN6-specific antibody

Royal jelly (RJ), a honeybee secretion, contains nine distinct water-soluble proteins known as major RJ proteins (MRJPs). MRJPs are the primary constituents of RJ and have demonstrated significant potential as antiviral agents. MRJPs exhibit antiviral effects against various viruses, including HCV, HBV, HIV, and SARS-CoV-2. Previous research has indicated that MRJPs can interfere with viral replication by targeting specific stages of the viral life cycle, such as by inhibiting key enzymes, including RNA-dependent RNA polymerase and reverse transcriptase. They also block viral entry into host c

BACKGROUND: Gustatory receptors (GRs) are known to mediate responses to various chemical stimuli including sugars and bitter compounds and sex pheromones. Although numerous types of GRs have been identified across insect species, the physiological roles of most GRs remain largely unknown. The silkworm, Bombyx mori, is representative of the order Lepidoptera, which includes over 70% of agriculturally important pest species. A notable feature of GRs in B. mori is that most GRs are organized in clusters, making them highly suitable for large-scale genetic studies.
RESULTS: Here we developed a tr

Autophagy and lipolysis are key metabolic pathways involved in lipid mobilization and energy homeostasis. In Rhodnius prolixus, a major vector of Chagas disease, previous studies have shown that silencing autophagy-related genes (RpAtg8, RpAtg6) or the brummer lipase gene RpBmm alters lipid metabolism and impairs flight activity. Here, we investigate the combined roles of lipolysis and lipophagy by performing a dual RNAi-mediated silencing of RpAtg8 and RpBmm under both fed and starved conditions. In the fat body, dual silencing did not exacerbate triacylglycerol (TAG) accumulation beyond sing
