Biological treatments for pediatric Netherton syndrome.

Netherton syndrome (NS) is a rare and potentially life-threatening genetic skin disease responsible for skin inflammation and scaling, hair abnormalities and severe allergic manifestations. NS is caused by loss-of-function variants in Serine Peptidase Inhibitor Kazal-Type 5 (SPINK5), encoding the serine protease inhibitor LEKTI. NS patients have a profound skin barrier defect caused by unopposed kallikrein-related proteases activity (KLKs). They develop severe skin inflammation with eczematous-like lesions and high serum IgE levels. Multiomics studies have revealed that the IL-17/IL-36 pathway…

Initial chemotherapy is effective, but most pa … ly response 3) post-NACT, associated with poor chemo-response and time to recurrence. CONCLUSIONS: Multiomics profiling of ovarian tumor samples pre- and post-NACT provides unique insight into chemo-induced changes to the tumor microenvironment. We identified a novel IL6/IER3 signaling axis that may drive chemoresistance and disease recurrence.

MCOF: A Multi-Channel Attention-Based Omics Fusion Framework for Cancer Subtyping and Candidate Biomarker Prioritization.

Multi-omics molecular subtyping is complicated by small cohorts, heterogeneous data layers, and sensitivity to data splitting and feature selection. We developed multi-channel attention-based omics fusion (MCOF), an end-to-end framework that combines omics-specific projection, one-dimensional convolution, and integrated gradients for candidate biomarker prioritization. MCOF was evaluated against six baselines using five repeated stratified holdouts across four multi-omics datasets. In the restricted four-class breast invasive carcinoma task, MCOF achieved the highest observed mean for all five

Stem cells and nanomaterials for cardiac regeneration: integrating multi-omics for precision therapeutics.

Cardiovascular diseases (CVDs) are one of the leading causes of mortality worldwide, with myocardial infarction (MI) resulting in irreversible loss of cardiomyocytes and progressive cardiac dysfunction. Clinical interventions primarily restore perfusion and alleviate symptoms yet fail to regenerate the damaged myocardium. Stem cell-based therapeutics have emerged as effective and promising regenerative strategies, largely mediated through paracrine signaling, immunomodulation, and cellular replacement. However, poor cell survival, inadequate engraftment, and functional immaturity of stem cell-

Multi-omics integration and machine learning identify an ACKR1+ endothelial cell-based lactate-immune signature associated with carotid plaque vulnerability and progression.

BACKGROUND: Early identification of vulnerable carotid plaques is crucial for stroke prevention, but the upstream metabolic processes linked to endothelial transition and immune recruitment remain poorly defined. We characterised lactate metabolism in ACKR1+ endothelial cells (ECs) in human carotid plaques and derived a machine-learning signature for discriminating plaque vulnerability. METHODS: Internal and external (GSE253903) single-cell datasets were integrated to identify endothelial subpopulations, and bulk transcriptomic (GSE163154) and in-house proteomic data were used to assess metab

Multi-omics identifies lipid accumulation in Myalgic Encephalomyelitis/Chronic Fatigue Syndrome cell lines: a case-control study.

BACKGROUND: In recent years, evidence has indicated a metabolic shift towards increased demand for lipids in various lymphoid cell populations from people with Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS). We previously screened the mitochondrial function and gene expression of B cell-derived lymphoblastoid cell lines (LCLs) generated from the blood of people with ME/CFS to characterise a model for hypothesis discovery and testing, observing elevated expression of gene products facilitating amino acid and fatty acid degradation for energy. METHOD: In this follow-up study we hav

Multi-omics study identifies diagnostic metabolic signatures of early Parkinson's disease associated with dysregulated glutathione and TCA cycle metabolism.

BACKGROUND: The development of non-invasive diagnostic tools for Parkinson's disease (PD) remains an unmet clinical need. Given the contribution by peripheral metabolism and environmental factors toward PD pathogenesis, comprehensive profiling of the plasma phenome could enable early disease detection. Accordingly, this study aimed to develop a non-invasive diagnostic panel for PD, to comprehensively characterize potential environmental triggers of PD pathogenesis, and to delineate the relationship between peripheral metabolism and disease progression. METHODS: We performed quantitative plasm

Chondroitin Sulfate Supplementation Is Associated with Body Weight Control in Ovariectomized Rats: A Multi-Omics Study of Gut Microbiota and Metabolite Profiles.

The prevalence of obesity among elderly women is increasing, particularly in postmenopausal populations. This exploratory study investigated the associations between chondroitin sulfate (CS) supplementation and body weight control in ovariectomized (OVX) rats, a preclinical model for postmenopausal physiology. CS supplementation was associated with significantly reduced body weight gain and preserved adipocyte morphology. CS-treated OVX rats exhibited decreased gut microbiota diversity, with reduced abundance of Photobacterium and Rhodococcus and increased abundance of Turicibacter. Metabolomi

Analyses of plasma multi-omic data across ancestries identify novel pathways implicated in Alzheimer's disease.

INTRODUCTION: Few genetic studies on Alzheimer's disease (AD) have incorporated multiple ancestries and omic datasets to pinpoint actionable AD risk effectors for each ancestry. METHODS: Here, we first performed genetic colocalization between molecular phenotypes (proteomics and metabolomics) from two ancestral groups (European [EUR] and African [AFR]) and the two largest EUR AD genome-wide association studies. We next performed pathway enrichment analyses to identify biological mechanisms. RESULTS: We found 21 proteins and one metabolite colocalized with AD risk that were shared between the

Integrative multi-omics profiling reveals an association between lipid metabolism dysregulation and bortezomib-induced cerebral hemorrhage in zebrafish.

Bortezomib (BTZ), a first-generation proteasome inhibitor, serves as a cornerstone chemotherapeutic agent for Multiple Myeloma (MM), with its clinical application in oncology expanding rapidly in recent years. Using vascular-specific transgenic zebrafish models, we systematically evaluated BTZ-induced toxicity at 4-5 mg/L gradient concentrations and its underlying molecular mechanisms. We found 72-hour BTZ exposure triggered significant intracerebral hemorrhage in zebrafish larvae with concentration-dependent severity, accompanied by cerebrovascular disruption, tight junction gene downregulati

Unraveling COPD pathogenesis: a multi-omics approach to identify metabolites and genetic links.

Chronic obstructive pulmonary disease (COPD) is a complex respiratory disorder driven by genetic, environmental, and metabolic factors. This study aims to elucidate the causal role of metabolites in COPD pathogenesis and identify novel therapeutic targets through a multi-omics approach coupled with experimental validation. We performed two-sample Mendelian randomization (MR) on 1,400 metabolites using genetic data from European-ancestry cohorts. Causal candidates were refined using stringent conditional colocalization (SuSiE, PP4 > 0.8) to exclude pleiotropic confounders. Pathway enrichment an

Investigating the potential role of propionylcarnitine in milk pentadecanoic acid synthesis in Chinese holstein dairy cows using multi-omics analysis.

BACKGROUND: Pentadecanoic acid (C15:0) is an odd-chain fatty acid (OCFA) with significant health benefits, its content in milk is regulated by both ruminal microbial metabolism and mammary gland synthesis. To investigate the regulatory mechanisms of C15:0 synthesis, we conducted 16S rRNA sequencing and ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) of ruminal fluid of Chinese Holstein dairy cows with low (1.03 ± 0.01%, Low group) and high (1.39 ± 0.06%, High group) milk C15:0 content. Additionally, an in vitro study was performed using key ruminal metabolit

Integrated Multi-Omics Analysis of Coagulation Dysregulation and Immune Pathway Alterations in 10-Gene Pig-to-Monkey Liver Xenotransplantation: A Case Report.

Liver xenotransplantation has the potential to address the global shortage of donor organs; however, coagulation dysregulation remains a predominant barrier to long-term liver xenograft survival. In this study, orthotopic liver xenotransplantation was performed using a 10-gene-edited (GTKO/CMAHKO/β4GalNT2KO/hCD46/hCD55/hCD59/hTBM/hCD39/hEPCR/hCD47) porcine donor liver transplanted into a nonhuman primate recipient. Independent of graft function, which remained stable, an unexpected interruption of the oxygen supply on POD 3 triggered a terminal event, leading to death on POD 4. Despite technic

Yellow tea extract ameliorates dexamethasone-induced hepatic steatosis by modulating the gut-liver axis and reshaping microbial metabolites: a multi-omics insight.

Long-term glucocorticoid therapy, exemplified by dexamethasone (DEX), frequently induces hepatic steatosis, posing a significant clinical challenge. Yellow tea (YT), a lightly fermented tea, is rich in polyphenols and polysaccharides, yet its protective effects against DEX-induced liver injury remain underexplored. This study investigated the hepatoprotective mechanisms of a yellow tea water extract (YT) using a DEX-induced mouse model, integrated with transcriptomic, metagenomic, and metabolomic analyses. YT intervention (500 mg-1 kg-1 day-1 for 6 weeks) significantly attenuated DEX-induced h

From Genes to Proteins: The Indispensable Role of Proteogenomics in Advancing Clear Cell Renal Cell Carcinoma Research.

Clear cell renal cell carcinoma (ccRCC) is characterized by a complex molecular landscape driven by recurrent genetic alterations. While genomic and transcriptomic profiling have identified core drivers, they often fail to provide robust biomarkers due to the significant decoupling of mRNA and protein levels, as well as the critical role of post-translational modifications in tumor biology. This review synthesizes current evidence from landmark proteogenomic initiatives, such as the Clinical Proteomic Tumor Analysis Consortium (CPTAC), and independent multi-omic studies. It evaluates the integ
