Morphology and molecular characterisation of two new species of Mymarothecioides (Monopisthocotyla: Dactylogyridae) parasitising the gills of anostomid fishes from Brazil.

Using an integrative taxonomic approach, we describe two new species of Monopisthocotyla, Mymarothecioides leporini n. sp. and Mymarothecioides paranaensis n. sp., parasitising the gills of anostomid fishes from the Jaguaribe River and Paraná River basins, Brazil. These descriptions represent the first record of species of Mymarothecioides in hosts of the family Anostomidae. Mymarothecioides leporini n. sp. is distinguished by a male copulatory organ (MCO) with a spoon-shaped distal portion and a ventral bar with an ornamented anteromedial process. Mymarothecioides paranaensis n. sp. differs f

A New Species of Creptotrematina (Trematoda: Allocreadiidae) from the Red Minor Tetra, Hyphessobrycon eques (Steindachner, 1882) (Characidae) from Brazil, with Comments on the Genetic Divergence of C. Aguirrepequenoi Jiménez-Guzmán, 1973 across a Wide Geographical Range in Middle America.

BACKGROUND: Allocreadiids are relatively small digeneans that appear to be restricted to freshwater systems distributed across the world. Allocreadiids are highly diverse in the Americas, particularly in the Neotropical biogeographical region. Their taxonomic history has been rather controversial, with several taxonomic reassessments and the description of new genera and species. METHODS: We sampled Creptotrematina specimens from a characid collected in the Pardo River, Paranapanema River basin in Brazil, and specimens of C. aguirrepequenoi, from Astyanax spp. in several localities between no

The evolution of TNF signaling in platyhelminths suggests the cooptation of TNF receptor in the host-parasite interplay.

BACKGROUND: The TNF signaling pathway is involved in the regulation of many cellular processes (such as apoptosis and cell proliferation). Previous reports indicated the effect of human TNF-α on metabolism, physiology, gene expression and protein phosphorylation of the human parasite Schistosoma mansoni and suggested that its TNF receptor was responsible for this response. The lack of an endogenous TNF ligand reinforced the idea of the use of an exogenous ligand, but also opens the possibility that the receptor actually binds a non-canonical ligand, as observed for NGFRs. METHODS: To obtain a

Parasitic Ulcerous Caseous Gastroesophagitis Associated with Rameshwarotrema uterocrescens Rao, 1975 (Digenea: Pronocephalidae) in a Juvenile Green Turtle ( Chelonia mydas , Linnaeus 1758 [Testudines: Cheloniidae]): A Case Report.

Here we report a case of ulcerative caseous gastroesophagitis associated with Rameshwarotrema uterocrescens, Rao, 1975 (Digenea: Pronocephalidae), in a juvenile green turtle ( Chelonia mydas ) from southern Brazil. Similar pathologies have been reported only in adult green turtles from Costa Rica. This paper presents the second report of parasitic esophagitis due to R. uterocrescens and the first occurrence in juvenile green turtles along coastal Brazil.

Immune reactivity in early life stages of sea-cage cultured Pacific bluefin tuna naturally infected with blood flukes from genus Cardicola (Trematoda: Aporocotylidae).

Pacific bluefin tuna (PBT), Thunnus orientalis, due to its high average price on the market is an economically valuable fish species. Infections by blood flukes from the genus Cardicola (Trematoda: Aporocotylidae) represent a growing concern for the cage culture of bluefin tuna in Japan, Australia and Southern Europe. The accumulation of numerous Cardicola eggs in the fish gills causes severe pathology that has been linked to mortality in PBT juveniles up to one year old. The only effective treatment used to mitigate the infection is the oral administration of the antihelminthic drug praziquan

Cross-fertilization as a reproductive strategy in a tissue flukes Didymosulcus katsuwonicola (Platyhelmintes: Didymozoidae) inferred by genetic analysis.

Mitochondrial DNA locus cytochrome oxidase I was used to asses intraspecific genetic diversity of a didymozoid species Didymosulcus katsuwonicola. Adult forms of this species live encapsulated in pairs in the gills of the reared Atlantic bluefin tuna (Thunnus thynnus). The life cycle of this food-borne parasites and its migration in the host tissues after releasing from the digestive tract to the definitive site in the gills are unknown. Our goal was to assess whether two encysted didymozoids share the same haplotype, indicative of a common maternal origin, as well as the extent of cross- in r

A new species of Intromugil (Digenea: Haploporidae) and redescription of Intromugil mugilicolus.

Intromugil alachuaensis n. sp. is described based on specimens collected from the flathead grey mullet ( Mugil cephalus ) from the Santa Fe River in Florida. The new species is the fourth recognized species in the genus and the second from North America, with the other 2 being confined to South America. Intromugil mugilicolus from Louisiana and Mississippi is redescribed based on the holotype and newly collected material that was not flattened prior to fixation. Two generic features not previously reported are apparent in the new material from I. mugilicolus and I. alachuaensis n. sp.: an arme

Cercarial dermatitis in the Netherlands caused by Trichobilharzia spp.

Outbreaks of cercarial dermatitis which occurred in recreational lakes in the Netherlands were studied and a method for direct rapid detection of the parasite Trichobilharzia in water samples was developed. A standardized questionnaire with questions on health complaints and exposure was distributed to individuals who developed symptoms of cercarial dermatitis after visiting fresh water lakes. Snails from the suspected lakes were examined for the presence of Trichobilharzia by microscopy and PCR. Water samples were concentrated by filtration and examined by PCR. Water quality was tested accord

First next-generation sequencing data for Haploporidae (Digenea: Haploporata): characterization of complete mitochondrial genome and ribosomal operon for Parasaccocoelium mugili Zhukov, 1971.

The first data on a whole mitochondrial genome of Haploporidae, Parasaccocoelium mugili (Digenea: Haploporata: Haploporidae) was generated using the next-generation sequencing (NGS) approach. We sequenced the complete mitochondrial DNA (mtDNA) and ribosomal operon of Parasaccocoelium mugili, intestine parasite of mullet fish. The mtDNA of P. mugili contained 14,021 bp, including 12 protein-coding genes, two ribosomal genes, 22 tRNA genes, and non-coding region. The ribosomal operon of P. mugili was 8308 bp in length, including 18S rRNA gene (1981 bp), ITS1 rDNA (955 bp), 5.8S rRNA gene (157 bp

Worm expulsion of Gymnophalloides seoi from C57BL/6 mice: role of metacercarial exosomes in upregulating TLR2 and MUC2 expression in intestinal tissues.

Gymnophalloides seoi worms were rapidly expelled from C57BL/6 mice within days 3-6 post-infection probably due to operation of mucosal innate immunity. To understand better the mucosal immunity related to worm expulsion from the host, we isolated exosomes of G. seoi metacercariae and investigated their role in induction of mRNA and protein expression of several Toll-like receptors and mucin-related factors in vitro. G. seoi-secreted exosomes were collected using differential ultracentrifugation, and cellular internalization of the exosomes into HT-29 intestinal epithelial cells was visualized

Development of a multiplex quantitative PCR assay for detection and quantification of DNA from Fasciola hepatica and the intermediate snail host, Austropeplea tomentosa, in water samples.

Liver fluke (Fasciola hepatica) infection is an increasing threat to livestock production resulting in serious economic losses to the beef, dairy and sheep industries in Australia and globally. Triclabendazole (TCBZ) is the main drug used to control liver fluke infections in Australia and the widespread emergence of TCBZ resistance in cattle and sheep threatens liver fluke control. Alternative control measures to lower exposure of livestock to fluke infection would be useful to help preserve the usefulness of current chemical flukicides. Environmental DNA (eDNA) sampling methodology and associ

Morphological and molecular characterisation of three Indian Neascus-type metacercariae (Digenea: Diplostomidae).

Diplostomid digeneans are important parasites whose larval stages infect freshwater snails and fish as first and second intermediate hosts respectively. Diplostomid digeneans as adults are parasitic in many fish-eating birds and mammals. Our understanding of the species diversity of diplostomid digeneans remains incomplete, especially in the Indian sub-continent. Here, we describe three Neascus-type metacercariae (N. hanumanthai, N. gussevi, and N. xenentodoni) from freshwater fish specimens of Channa punctata (Bloch 1793), Trichogaster fasciata (Bloch and Schneider 1801) and Xenentodon cancil

Isorchis cannoni n. sp. (Digenea: Atractotrematidae) from Great Barrier Reef rabbitfishes and the molecular elucidation of its life cycle.

We describe Isorchis cannoni n. sp. from the rabbitfishes Siganus fuscescens (Houttuyn) and Siganus lineatus (Valenciennes) (Siganidae) collected off Heron Island, southern Great Barrier Reef, Australia and, using molecular data, demonstrate that 'Cercariae queenslandae II' of Cannon (1978) from the gastropod Clypeomorus batillariaeformis Habe & Kosuge (Cerithiidae) is the larval form of this new species. The cercariae of I. cannoni n. sp. develop in rediae, encyst in the environment after emergence, and are inferred to then be consumed by grazing rabbitfish. Additionally, we provide a new rep

Human infections with liver and minute intestinal flukes in Guangxi, China: analysis by DNA sequencing, ultrasonography, and immunoaffinity chromatography.

The prevalence of liver and intestinal fluke infections was determined by surveying inhabitants of Hengxuan, Fusui, and Shanglin villages which were known to be endemic for liver flukes in Guangxi, China in May 2010. A total of 718 people were examined for helminth eggs by the Kato-Katz thick smear technique, ultrasonography, immunoaffinity chromatography, and DNA sequencing. The overall egg positive rate was found to be 59.6% (28.0-70.6%) that included mixed infections with liver and intestinal flukes. Cases showing higher than 20,000 eggs per gram of feces (EPG) were detected between 1.3% an

Macroderoides texanus n. sp. (Digenea: Macroderoididae) from alligator gar, Atractosteus spatula in Texas.

Macroderoides texanus n. sp. is described based on 16 specimens collected from the intestine of the North American alligator gar, Atractosteus spatula. Of the five established species of Macroderoides, the new species is morphologically most similar to Macroderoides spiniferus and Macroderoides trilobatus. M. texanus n. sp. differs from M. spiniferus by having the ovary situated immediately posterior to the cirrus sac rather than at mid-way between the cirrus sac and anterior testis, the ventral sucker situated further posteriorly, and the vitelline fields extending somewhat posterior to poste

[Statistical models for spatial analysis in parasitology].

The simplest way to study the spatial pattern of a disease is the geographical representation of its cases (or some indicators of them) over a map. Maps based on raw data are generally "wrong" since they do not take into consideration for sampling errors. Indeed, the observed differences between areas (or points in the map) are not directly interpretable, as they derive from the composition of true, structural differences and of the noise deriving from the sampling process. This problem is well known in human epidemiology, and several solutions have been proposed to filter the signal from the

Detection of circulating Fasciola gigantica antigen in experimental and natural infections of sheep with fasciolosis.

Detection of circulating Fasciola gigantica antigen was performed in sera of sheep with experimental and natural F. gigantica infections using the direct enzyme-linked immunosorbent assay method. Sera from sheep with monoinfections of Schistosoma bovis, Dicrocoelium hospes and Paramphistomum microbothrium were included in the assay to ascertain specificity. Circulating F. gigantica antigen (CFA) was detected as early as 1 week after infection in the experimentally infected sheep. No detectable CFA was observed 2 weeks after chemotherapy. Positivity rates of 82.5%, 12.5%, 10% and 10% were found

Time-course and accumulation of triclabendazole and its metabolites in bile, liver tissues and flukes collected from treated sheep.

The flukicidal compound triclabendazole (TCBZ) has a complex metabolic pattern that includes the systemic presence of its sulphoxide (TCBZ.SO) and sulphone (TCBZ.SO2) metabolites, usually recovered from the bile of treated animals. The aim of the current work was to evaluate the time-course and pattern of in vivo accumulation of TCBZ/metabolites into adult Fasciola hepatica specimens recovered from infected sheep. Twelve (12) healthy Corriedale sheep were orally infected with one hundred (100) metacercariae of the TCBZ-susceptible Cullomptom isolate of F. hepatica. Sixteen weeks after infectio
