Terpyridine Eu(III) complexes combined with boric acid-functionalized carbon dots for on-site visual ratiometric fluorescence detection of anthrax spore biomarkers.

The development of reliable methodologies for the identification of potential biomarkers holds paramount significance for the early detection of serious diseases and conditions. In this work, we have designed a new terpyridine Eu(III) complexes with boric acid functional carbon dot-based fluorescent probe Eu-Tpy/R-CDs for the ratiometric of detection of dipicolinic acid (DPA). Eu-Tpy/R-CDs nanoprobe were obtained by co-assembly and immobilization of green luminescent Rhodanine B carbon dots to boric acid matrix sites in tripyridine‑europium (III) complexes using the solvent method. The novel n

Doxycycline, levofloxacin, and moxifloxacin are superior to ciprofloxacin in treating anthrax meningitis in rabbits and NHP.

Efficient treatment of anthrax-related meningitis in patients poses a significant therapeutic challenge. Previously, we demonstrated in our … s ineffective, while doxycycline protected the infected rhesus macaques (n = 2) from the lethal CNS Bacillus anthracis infection. To test whether the low efficacy of Ciprofloxacin is an example of low efficacy of all fluoroquinolones or only this substance, we treated rabbits that were inoculated intracisterna magna (ICM) with levofloxacin or moxifloxacin. We found …

Livestock owners' anthrax prevention practices and its associated factors in Sekota Zuria district, Northeast Ethiopia.

BACKGROUND: In Ethiopia, the second most prioritized of the zoonotic diseases next to rabies is anthrax. About 50.6% of anthrax cases and 33.3% of deaths of livestock have been reported from Wag-Himra Zone, where appropriate anthrax prevention practices are not implemented by the owners of the animals. Thus, the aim of this study was to determine the extent of appropriate anthrax prevention practices of livestock owners and associated factors in Sekota Zuria district, northwest Ethiopia. RESULTS: Twenty-five percent (95% CI: 25.2, 26.1%) of the livestock owners implemented appropriate anthrax

Cost-effectiveness comparison of response strategies to a large-scale anthrax attack on the chicago metropolitan area: impact of timing and surge capacity.

Rapid public health response to a large-scale anthrax attack would reduce overall morbidity and mortality. However, there is uncertainty about the optimal cost-effective response strategy based on timing of intervention, public health resources, and critical care facilities. We conducted a decision analytic study to compare response strategies to a theoretical large-scale anthrax attack on the Chicago metropolitan area beginning either Day 2 or Day 5 after the attack. These strategies correspond to the policy options set forth by the Anthrax Modeling Working Group for population-wide responses

Anthrax in animals.

Anthrax is the archetype zoonosis; no other infectious disease affects such a wide range of species, including humans, although most susceptible are herbivorous mammals. Although the disease appears to have been recognized for centuries, it has yet to be established scientifically how animals contract it. While primarily a disease of warmer regions, it has long been spread to cooler zones through the trade of infected animals or contaminated animal products. Today it is still endemic in many countries of Africa and Asia and non-endemic countries must remain alert to the possibility of imports

Serious adverse events among participants in the Centers for Disease Control and Prevention's Anthrax Vaccine and Antimicrobial Availability Program for persons at risk for bioterrorism-related inhalational anthrax.

On 20 December 2001, the Centers for Disease Control and Prevention (CDC) initiated the Anthrax Vaccine and Antibiotic Availability Program (hereafter, the "Program") under an investigational new drug application with the US Food and Drug Administration. This Program provided options for additional preventive treatment for persons at risk for inhalation anthrax as a result of recent bioterrorism attacks who had concluded or were concluding a 60-day course of antimicrobial prophylaxis. Participants were offered an additional 40 days of antibiotic therapy (with ciprofloxacin, doxycycline, or amo

Anthrax control and research, with special reference to national programme development in Africa: memorandum from a WHO meeting.

The prevalence of anthrax in both animal and human populations has been increasing in Africa. It was therefore appropriate for this WHO meeting to be convened in an endemic area of the Western Province of Zambia in 1992. The participants reviewed anthrax epidemiology and control in some African countries, elaborated national anthrax control and research programmes in Africa, discussed international cooperation and work plans, and elaborated recommendations for anthrax control in Africa. The discussions centred on anthrax surveillance and reporting systems, diagnosis, vaccine production and imm

Anthrax protective antigen forms oligomers during intoxication of mammalian cells.

The protective antigen component (PA) of anthrax toxin binds to receptors on target cells and conveys the toxin's edema factor (EF) and lethal factor (LF) components into the cytoplasm. PA (83 kDa) is processed by a cellular protease, yielding a 63-kDa fragment (PA63), which binds EF and/or LF. When exposed to acidic pH, PA63 inserts into membranes and forms ion-conductive channels. By electron microscopy, a significant fraction of purified PA63 was found to be in the form of a multi-subunit ring-shaped oligomer (outer diameter, 10.4 nm). The rings are heptameric, as judged by inspection and b

Antibodies to anthrax toxin in humans and guinea pigs and their relevance to protective immunity.

A forerunning study on the relationship between antibodies to the protective antigen (PA) and lethal factor (LF) components of anthrax toxin and protective immunity has been expanded and extended to include the third toxin component, the edema factor (EF). It was found that protection against the "vaccine resistant" Ames strain was possible in the absence of detectable anti-LF and anti-EF antibodies. Evidence is given that PA may be the essential anthrax-derived antigen for protection, but that equally essential is that it be presented to the host's immune system in such a manner as to provide

Accurately assembling nanopore sequencing data of highly pathogenic bacteria.

BACKGROUND: Bacterial genome exploration and outbreak analysis rely heavily on robust whole-genome sequencing and bioinformatics analysis. Widely-used genomic methods, such as genotyping and detection of genetic markers demand high sequencing accuracy and precise genome assembly for reliable results. METHODS: To assess the utility of nanopore sequencing for genotyping highly pathogenic bacteria with low mutation rates, we sequenced six reference strains using Oxford Nanopore Technologies (ONT) R10.4.1 chemistry and Illumina and evaluated different assembly strategies. The publicly available R

Trimethyl Chitosan Nanoparticles Encapsulated Protective Antigen Protects the Mice Against Anthrax.

Anthrax is an era old deadly disease against which there are only two currently available licensed vaccines named anthrax vaccine adsorbed and precipitated (AVP). Though they can provide a protective immunity, their multiple side-effects owing to their ill-defined composition and presence of toxic proteins (LF and EF) of Bacillus anthracis, the causative organism of anthrax, in the vaccine formulation makes their widespread use objectionable. Hence, an anthrax vaccine that contains well-defined and controlled components would be highly desirable. In this context, we have evaluated the potentia…

Cutaneous Anthrax.

CASE PRESENTATION: A 60-year-old male from the country of Jordan presented to the emergency department with swelling, pain, black lesions on his fingertips and thumb, and a red streak up his arm. The patient had been trimming sheep wool and goat skin two weeks prior to onset of symptoms and cut his left thumb with trimming shears. DISCUSSION: Cutaneous anthrax is caused by Bacillus anthracis, a Gram-positive, spore-forming rod found naturally in the soil. Populations at greatest risk include those who consume undercooked meat with contaminated spores or who live and work in rural/agricultural…

Anthrax: A narrative review.

Bacillus anthracis is a zoonotic bacterium, majorly responsible for causing human anthrax and the possibility of the outbreak spreading globally. Herbivorous animals serve as the inherent reservoir for the disease, whereas all endothermic species are vulnerable. Humans contract the disease inadvertently by contact with diseased animals or animal products or through the consumption or handling of infected flesh. There is no such reported data indicating the transmission of anthrax from human to human, which further does not guarantee the bacterium's mutations and new transmission route. Neverth…
