Transcription factor AP2 and its role in epidermal-specific gene expression.

The epidermis is a stratified squamous epithelium whose major differentiation-specific products are keratins. To elucidate factors controlli … identify this factor as transcription factor AP2, shown to be transcribed in cells of epidermal and neural crest lineages. Furthermore, we demonstrate functional AP2 binding sites upstream from three additional epidermal genes, suggesting that AP2 may be generally involved in epidermal gene regulation. Finally, although AP2 is necessary, it is not sufficient f …

Conformer-Resolved Optical Rotation Prediction From Three-Dimensional Quantum and Semiempirical Molecular Fields Using 3D Convolutional Neural Networks.

Optical rotation is highly sensitive to molecular conformation, making prediction expensive when conformational ensembles and electronic-response calculations are required. We evaluate three-dimensional convolutional neural networks trained on volumetric molecular descriptors for conformer-level optical rotation. A chemically structured set of substituted phenylethanol and phenylethylamine derivatives yielded 82,790 valid conformer-level sTD-DFT reference records. Three descriptors were compared on a common grid: HF/STO-3G electron density, B3LYP/cc-pVDZ electron density, and GFN2-xTB charge f

The SHEDs-derived apoptotic bodies for inflammatory regulation in spinal cord repair.

Neuroinflammation is a key therapeutic target for spinal cord injury (SCI). Apoptotic bodies (ABs) derived from mesenchymal stem cells may modulate early inflammation, promoting SCI repair. Stem cells from human exfoliated deciduous teeth (SHEDs), with neural crest origins, are promising for neurological therapies, but their ABs' role in SCI remains unclear. In this study, as exploratory research, we aimed to investigate the preliminary effects and mechanisms of SHEDs-derived ABs on the treatment of SCI. Herein, SHEDs-derived ABs enhanced functional recovery in SCI mice, improving BMS scores, …

Meis2 transcription factor regulates craniofacial osteogenesis via IGF signaling and cell adhesion.

Cranial neural crest cell (cNCC)-derived mesenchyme must precisely integrate intrinsic transcriptional programs with localized signaling cues to orchestrate craniofacial skeletogenesis. While the transcription factor Meis2 is an essential regulator of this process, the spatially defined cellular contexts and downstream effector pathways remain poorly characterized. Building upon previous single-cell transcriptomic evidence of Igf2 dysregulation, we employed spatial transcriptomics integrated with single-cell references to map the mesenchymal landscape in Meis2-deficient embryos during early os…

A Rare Case of Arytenoid Cartilage Neurofibroma Mimicking Laryngeal Carcinoma.

Neurofibroma is a benign tumor arising from neural crest-derived cells and is characterized by a nonencapsulated growth pattern that incorporates surrounding nerve fibers. Laryngeal involvement is rare, and arytenoid cartilage localization is exceptionally uncommon. We present a 46-year-old woman with hoarseness and cough in whom flexible laryngoscopy demonstrated mucosal swelling over the right arytenoid cartilage. Magnetic resonance imaging revealed a 15 × 10 mm lesion that was isointense on T1-weighted images and heterogeneously hyperintense on T2-weighted images, with no diffusion restrict…

SIX1 branchio-oto-renal syndrome variants have different effects on embryonic craniofacial gene expression and cartilage formation.

SIX1 variants underlying branchio-oto-renal syndrome occur in the SIX domain (SD) or homeodomain (HD). We tested whether different variants - V17E (SD), Y129C (HD) - cause distinct developmental phenotypes in Xenopus embryos with reduced Six1 in comparison to wild-type Six1 (Six1WT). In Six1 morphants, Six1WT restored neural crest and preplacodal gene expression; V17E restored foxd3 and irx1 better than Y129C, and Y129C restored sox11 better than V17E. In six1-null otic vesicles, Six1WT partially restored tbx1 and sobp, V17E was less effective and Y129C was least effective; all three restored …

Targeting the β2-adrenergic receptor suppresses melanoma progression by disrupting the PKA-SOX10-MAGEA1 oncogenic axis.

Chronic stress and sympathetic signaling, mediated by the β2-adrenergic receptor (ADRB2), are implicated in cancer progression. In melanoma, a neural crest-derived malignancy, the interplay between neuroendocrine signals and tumor cell plasticity remains poorly understood. Elucidating how ADRB2 activation translates into pro-tumorigenic transcriptional programs is crucial for developing novel therapeutics. We combined genetic and pharmacological ADRB2 modulation in melanoma models with transcriptomics, mechanistic assays, and patient sample analysis. ADRB2 inhibition suppressed tumor growth by…

Loss of Sox10 prevents tumor initiation in vivo and induces luminal-to-basal reprogramming in Neu+ tumor cells.

The SRY-HMG-Box transcription factor SOX10 plays a critical role in neural crest development, but its function in epithelial tumorigenesis remains unclear. Here, we identify SOX10 as a key regulator of tumor-initiating activity in Neu-driven mammary cancers. Genetic ablation of Sox10 in the luminal compartment of MMTV-Neu (NIC) mice resulted in delayed but normal mammary gland development. Sox10 deletion resulted in a reduction in mammary progenitors and a complete loss of tumor initiation in Sox10-deficient luminal cells. CRISPR/Cas9-mediated Sox10 inactivation in Neu-transformed tumor cells …

Integrated Genetic Networks and Epigenetic Regulation inTooth Development and Maturation.

Tooth development or odontogenesis is a complex morphogenetic process that requires tightly regulated interactions between the oral epithelium and mesenchyme of neural crest origin. In this narrative review, we compile existing knowledge regarding gene regulatory networks and epigenetic factors throughout tooth development from initiation to eruption. Signaling between the epithelium and mesenchyme is mediated by four conserved pathways-Wnt/β-catenin, bone morphogenetic protein (BMP), fibroblast growth factor (FGF), and Sonic hedgehog (Shh)-which operate iteratively and interact through extens…

MYCN reprograms postnatal cells toward embryonic-like transcriptional states and drives tumorigenesis.

The embryonic-like features of neuroblastoma are commonly explained by a developmental arrest model, in which neural crest-derived progenitors undergo transformation after failing to complete normal sympathoadrenal differentiation. Whether such features can instead be actively imposed on postnatal cells during tumor development remains unclear. Here, we show that Mycn, but not c-Myc, induced adrenal tumors after activation in adult tyrosine hydroxylase-positive cells, accompanied by repression of differentiated programs and acquisition of embryonic cell-like features. Similarly, in adult hepat…

Melanotic Neuroectodermal Tumor of Infancy.

BACKGROUND: Melanotic neuroectodermal tumor of infancy (MNTI) is a rare, benign but locally aggressive neoplasm of neural crest origin, typically arising in the anterior maxilla of infants under 1 year of age. Clinically, it often mimics an eruption cyst, leading to potential diagnostic delay. METHODS: A 4-month-old male presented with a painless, rapidly enlarging swelling in the left anterior maxilla. Clinical, radiographic, and histopathologic evaluations were performed. Excisional biopsy under general anesthesia was chosen as definitive management. Preoperative urinary vanillylmandelic ac…

Cerebellopontine Angle Meningeal Melanocytoma: A Rare Case and Literature Review of Outcomes and Recurrences.

Melanocytes are neural crest derived cells. Intracranial meningeal melanocytomas (MM) are rare tumors that were first described in 1972. Authors report a rare case of a cerebellopontine (CP) angle MM. In addition, a literature review of CP angle MM is provided and compared with other cranial MMs in terms of recurrence and outcomes. A 45 year old male presented with tinnitus and occipital headache. He had mild sensorineural hearing loss. MRI showed an extra axial CP angle lesion that was hyperintense on T1 and hypointense on T2, with blooming on SWI sequences. A retrosigmoid craniotomy and near…
