Large-scale epidemiology of opisthorchiasis in 21 provinces in Thailand based on diagnosis by fecal egg examination and urine antigen assay and analysis of risk factors for infection.

INTRODUCTION: Infection with the carcinogenic fish-borne trematode Opisthorchis viverrini, known as opisthorchiasis, is a major cause of biliary cancer (cholangiocarcinoma). Despite decades of disease prevention and control in Thailand, the parasite remains endemic. Here we apply a novel antigen assay for mass screening of opisthorchiasis and compare the prevalence against the conventional examination and analyze risk factors associated with current O. viverrini infection. MATERIALS AND METHODS: We conducted a large-scale cross-sectional survey to assess transmission of O. viverrini in the No

Characterisation of a secretory serine protease inhibitor (SjB6) from Schistosoma japonicum.

BACKGROUND: Proteins belonging to the serine protease inhibitor (serpin) superfamily play essential physiological roles in many organisms. In pathogens, serpins are thought to have evolved specifically to limit host immune responses by interfering with the host immune-stimulatory signals. Serpins are less well characterised in parasitic helminths, although some are thought to be involved in mechanisms associated with host immune modulation. In this study, we cloned and partially characterised a secretory serpin from Schistosoma japonicum termed SjB6, these findings provide the basis for possib

Host-associated mitochondrial structure and broad geographic connectivity in Drepanocephalus associated with migratory cormorants.

Digenetic trematodes are important parasites of aquaculture systems, where transmission may be amplified by interactions among aquatic birds, fishes, and snail intermediate hosts. Species boundaries and transmission dynamics within the widespread echinostomatid genus Drepanocephalus remain poorly understood despite reports of pathogenicity in North American aquaculture facilities. Here, we characterize Drepanocephalus recovered from Double-crested Cormorants (Nannopterum auritus) in New York State and evaluate their evolutionary relationships using multilocus phylogenetic analyses and next-gen

Diversity of helminths parasitising North-East Atlantic and Antarctic seabirds.

Seabirds are largely used as indicators of Ocean health and are final hosts of several helminth parasites. However, the helminth fauna of seabirds is still poorly studied. Here, we quantified the diversity of gastrointestinal parasites in 52 individuals belonging to 10 seabird species with different habitat preferences and feeding strategies from the North-East Atlantic and Antarctica. Fresh carcasses were collected in Northern France and at Svarthamaren (Dronning Maud Land, Antarctica), helminth parasites were extracted from the gastrointestinal tract, and were identified by morphological ins

Subcutaneous Migration in Paragonimiasis as a Diagnostic Clue in an Unusual Presentation: A Case Report.

Paragonimiasis is a foodborne parasitic infection caused by trematodes of the genus Paragonimus, primarily P. westermani and P. miyazakii in Japan. Though once endemic, its incidence has declined significantly in recent decades. However, globalization, changes in dietary customs, and an increase in foreign-born residents have contributed to the emergence of new cases, particularly in urban settings. We present a case of a 25-year-old man from Cambodia, living in Japan for two years as a technical intern trainee, who developed a productive cough and pleuritic chest pain. He reported consuming r

Fasciola hepatica vaccine based on Kunitz-type molecule reduces adult worm fecundity in experimentally infected sheep.

Fasciolosis is a widespread and continuously expanding helminthiasis caused by the trematode Fasciola hepatica. Sheep and cattle are the primary definitive hosts of F. hepatica and are economically significant hosts for this pathogen worldwide. F. hepatica is not only a major threat to livestock but also an important neglected zoonosis. Reports of anthelmintic resistance in F. hepatica emphasize the urgent need for the development of an effective vaccine. Such a vaccine would reduce the impact and spread of the disease by decreasing the number of viable eggs, as well as reducing the adult worm

Molecular and biochemical characterizations of a Fasciola gigantica retinoid X receptor-α isoform A (FgRXRα-A).

Fascioliasis is a parasitic infection in animals and humans caused by the parasitic flatworm genus Fasciola, which has two major species, F. hepatica and F. gigantica. A major concern regarding this disease is drug resistance, which is increasingly reported worldwide. Hence, the discovery of a novel drug as well as drug targets is crucially required. Therefore, this study aims to characterize the novel drug target in the adult F. gigantica. In the beginning, we hypothesized that the parasite might interact with some host molecules when it lives inside the liver parenchyma or bile ducts, specif

Fasciola hepatica juveniles interact with the host fibrinolytic system as a potential early-stage invasion mechanism.

BACKGROUND: The trematode Fasciola hepatica is the most widespread causative agent of fasciolosis, a parasitic disease that mainly affects humans and ruminants worldwide. During F. hepatica infection, newly excysted juveniles (FhNEJ) emerge in the duodenum of the mammalian host and migrate towards their definitive location, the intra-hepatic biliary ducts. Understanding how F. hepatica traverses the intestinal wall and migrates towards the liver is pivotal for the development of more successful strategies against fasciolosis. The central enzyme of the mammalian fibrinolytic system is plasmin,

Parasitic diseases of equids in Iran (1931-2020): a literature review.

Parasitic infections can cause many respiratory, digestive and other diseases and contribute to some performance conditions in equids. However, knowledge on the biodiversity of parasites of equids in Iran is still limited. The present review covers all the information about parasitic diseases of horses, donkeys, mules and wild asses in Iran published as articles in Iranian and international journals, dissertations and congress papers from 1931 to July 2020. Parasites so far described in Iranian equids include species of 9 genera of the Protozoa (Trypanosoma, Giardia, Eimeria, Klossiella, Crypt

Functional diversity of secreted cestode Kunitz proteins: Inhibition of serine peptidases and blockade of cation channels.

We previously reported a multigene family of monodomain Kunitz proteins from Echinococcus granulosus (EgKU-1-EgKU-8), and provided evidence that some EgKUs are secreted by larval worms to the host interface. In addition, functional studies and homology modeling suggested that, similar to monodomain Kunitz families present in animal venoms, the E. granulosus family could include peptidase inhibitors as well as channel blockers. Using enzyme kinetics and whole-cell patch-clamp, we now demonstrate that the EgKUs are indeed functionally diverse. In fact, most of them behaved as high affinity inhib

Protection against Fasciola gigantica infection in mice by vaccination with recombinant juvenile-specific cathepsin L.

Fasciola gigantica cathepsin L1H (FgCatL1H) is one of the major cathepsin L released by juveniles of F. gigantica to aid in the invasion of host's tissues. Due to its high sequence similarity with other cathepsin L (CatL) isoforms of late stage F. gigantica, it was considered to be a good vaccine candidate that can block all CatL-mediated protease activities and affect juveniles as well as adult parasites. In this study, recombinant proFgCatL1H protein expressed in yeast, Pichia pastoris, system was mixed with Freund's adjuvants and used to subcutaneously immunize mice that were later challeng

Fascioliasis and other plant-borne trematode zoonoses.

Fascioliasis and other food-borne trematodiases are included in the list of important helminthiases with a great impact on human development. Six plant-borne trematode species have been found to affect humans: Fasciola hepatica, Fasciola gigantica and Fasciolopsis buski (Fasciolidae), Gastrodiscoides hominis (Gastrodiscidae), Watsonius watsoni and Fischoederius elongatus (Paramphistomidae). Whereas F. hepatica and F. gigantica are hepatic, the other four species are intestinal parasites. The fasciolids and the gastrodiscid cause important zoonoses distributed throughout many countries, while W

Revisiting the diversity of freshwater fish trematode metacercariae in cenotes (sinkholes) of Yucatán: molecular characterisation and comments on their phylogenetic interrelationships.

The limestone soil of the Yucatán Peninsula is highly permeable, allowing fluvial waters to filter into the aquifer, resulting in the absence of rivers on the surface and the formation of underground reservoirs that, on some occasions, open to the surface, forming the so-called cenotes (or sinkholes). Information about the freshwater fish parasite diversity of cenotes in the Yucatán Peninsula is scarce. Only 50 cenotes of the approximately 9,000 occurring in the peninsula have been sampled to assess the fish parasite fauna. Available information predates the molecular era and is based on morph

Complex species structure of Lecithaster salmonis (Digenea: Lecithasteridae), a fish parasite in the Arctic and Pacific Northwest.

Studying complexes of cryptic or pseudocryptic species opens new horizons for the understanding of speciation processes, an important yet vague issue for the digeneans. We investigated a hemiuroidean trematode Lecithaster salmonis across a wide geographic range including the northern European seas (White, Barents, and Pechora), East Siberian Sea, and the Pacific Northwest (Sea of Okhotsk and Sea of Japan). The goals were to explore the genetic diversity within L. salmonis through mitochondrial (cox1 and nad5 genes) and ribosomal (ITS1, ITS2, 28S rDNA) marker sequences, to study morphometry of

Gyrodactylidae from Fundulus heteroclitus (Fundulidae, Cyprinodontiformes) in South Carolina, USA, and insights on the historical ecology of the gyrodactylids of Fundulidae.

Mummichog (Fundulus heteroclitus) hosts a diverse array of parasites, including several species of Gyrodactylidae. We surveyed populations of mummichogs across three estuarine systems in South Carolina, USA, for gyrodactylids and studied their taxonomy, phylogenetics, and evolutionary history. Molecular analysis using concatenated sequences of 18S, ITS, and 5.8S rDNA fragments and morphology support the synonymization of Fundulotrema with Swingleus, as the former was shown paraphyletic. Five species of Gyrodactylidae were encountered. Gyrodactylus kritskyi n. sp. is described and four other sp

Relationships between liver and rumen fluke infections, milk somatic cells and polymorphisms in the Toll-like receptor 5 gene and vitamin D metabolism-related genes in Holstein dairy cows.

This study investigated polymorphisms in the genes CYP3A4, CYP2R1, and TLR5, and their associations with liver fluke (Fasciola hepatica) and rumen fluke (Calicophoron / Paramphistomum spp.) infections as well as with milk somatic cell count (SCC) as an indicator for mastitis in Holstein Friesian dairy cows. DNA sequencing of the genes' exons, 5'-, 3'-untranslated regions (UTR), introns, and flanking regions of 24 cows revealed 442 variants (388 SNPs and 54 InDels) including 116 previously unknown variants. We detected three novel non-synonymous variants leading to the derived amino acid exchan
