Memory performance in polyvalent MDMA (ecstasy) users who continue or discontinue MDMA use.

BACKGROUND: The popular dance drug ecstasy (3,4-methylenedioxymethamphetamine=MDMA) is a serotonergic neurotoxin in animal studies. Several cross-sectional investigations reported low memory and learning performance in ecstasy users, particularly in those reporting heavy patterns of drug use. Since, serotonin has a recognized role in memory processes, these findings were mostly interpreted as evidence for ecstasy-related neurotoxicity in humans. However, studies with user populations and controls suffer from many inherent methodological problems. Moreover, longitudinal data on memory performan

Cannabinoid-induced alterations in brain disposition of drugs of abuse.

Marijuana contains a complex mixture of compounds including tetrahydrocannabinol (THC), the major psychoactive constituent, and cannabidiol (CBD), a nonpsychoactive constituent. We have shown previously that CBD pretreatment of mice increases brain levels of THC and have now further characterized this effect and determined whether the brain pharmacokinetics of other drugs are also affected. CBD pretreatment of mice (30-60 min) increased brain levels of THC nearly 3-fold, whereas CBD co-administration did not. Because marijuana is often consumed with other drugs, the influence of cannabinoids o

Acute psychological and physiological effects of MDMA ("Ecstasy") after haloperidol pretreatment in healthy humans.

3,4-Methylenedioxymethamphetamine (MDMA, "Ecstasy") releases serotonin and dopamine. The role for dopamine in mediating the effects of MDMA has not yet been examined in humans. We investigated the effect of pretreatment with the dopamine D(2) antagonist haloperidol (1.4 mg i.v.) on psychological and physiological responses to MDMA (1.5 mg/kg p.o.) in 14 healthy volunteers using a double-blind placebo-controlled within-subject design. Subjective peak effects were rated using standardised scales. The physiological effects measured were blood pressure, heart rate and body temperature. Side effect

Testing for drugs in hair. Critical review of chromatographic procedures since 1992.

Up to now, more than 50 pharmaceuticals or drugs of abuse have been reported to be detectable in hair after oral or parenteral administration. The present paper reviews the literature devoted to drug testing in hair that has been published since 1992. Procedures for the detection of opiates, cocaine, amphetamines and cannabis in hair are described in detail. In particular, the papers on benzodiazepines show an increasing number of procedures using negative chemical ionisation with GC-MS and diode array detection with HPLC in hair analysis. For the most important benzodiazepines, diazepam and f

Effects of expectancies on subjective responses to oral delta9-tetrahydrocannabinol.

The effects of expectancies on subjective responses to oral delta9-tetrahydrocannabinol (delta9-THC) were examined. Thirty-five regular marijuana users were assigned to one of two groups: one group was told that they may receive a cannabinoid or placebo and a second group was told that they may receive a drug from one of several classes of drugs (e.g., stimulant, sedative, antiemetic) or placebo. Regardless of the group to which they were assigned, subjects received each of two oral doses of delta9-THC (7.5 and 15 mg) and placebo, one dose per session, for a total of three sessions. Measures o

Fatal poisoning by MDMA (ecstasy) and MDEA: a case report.

The first observation of lethal recreational use of MDMA (ecstasy) and MDEA in Italy is reported, together with extensive toxicological and histopathological documentation. Findings such as disseminated intravascular coagulation, rarely reported before, are colocated in the framework of the toxic syndrome for a better definition of criteria for forensic diagnosis.

Medical treatment of the adolescent drug abuser. An opportunity for rehabilitative intervention.

Illnesses related to both the pharmacologic properties of abused substances and their methods of administration often bring the teenager to medical attention and may provide sufficient motivation for the adolescent to seek help beyond the acute problem. Successful treatment of an overdose reaction, an abstinence syndrome, or any other medical complication of drug abuse may give the physician a unique opportunity to begine further evalution for future care.

Risk of substance use disorders in the adult children of parents with severe alcohol use disorder: a nationwide cohort study.

BACKGROUND: Offspring of parents with alcohol use disorder (AUD) have elevated risk of substance use. However, few studies have comprehensively assessed risks associated with different substances. This study investigated the risk of substance use disorders (SUDs) in adult children with severe parental AUD over four decades, contributing information on the risk of each disorder, the roles of important risk factors, and the general versus substance-specific nature of SUD risk. METHODS: Swedish national register data were used to follow children with and without ≥ 1 parent with an inpatient diagn

[Issues of risky behaviours in university students].

AIM: Risk behaviour is typical of adolescence and young adulthood, but the consequences can persist for the rest of life - in all domains - … llicit substances - marijuana, sedatives/tranquilizers without a doctor's prescription, cocaine, or hallucinogens compared to the age-matched general population. In the area of risky sexual behaviours, a question was included       about sexual intercourse with casual acquaintances. It was experienced by one in four respondents. In 83.3%, this behaviour was inf …
