Phototoxicity of Ultraviolet-A against the Whitefly Bemisia tabaci and Its Compatibility with an Entomopathogenic Fungus and Whitefly Parasitoid.

Ultraviolet (UV) radiation significantly affects insect life and, as a result, has been widely used to control different invertebrate pests. The current results demonstrate that when Bemisia tabaci first instar nymphs are exposed to UV-A light for 12, 24, 48, and 72 h, their developmental and biological parameters are negatively affected by UV-A exposure; the effect increased with an increase in exposure time. We hypothesized that UV-A light is compatible with other biological control agents. Results showed that when the entomopathogenic fungus Cordyceps fumosorosea was applied to third instar

The Florey lecture, 1983. Biological control, as exemplified by smallpox eradication and myxomatosis.

Biological control is an important method of dealing with plant and insect pests. The control of rabbits by myxomatosis and the eradication of smallpox by vaccination are unusual examples of biological control, in that they involve a vertebrate and a viral pest respectively. Myxomatosis is a benign disease in Sylvilagus rabbits in South America which is transmitted mechanically by mosquitoes. In the European rabbit, Oryctolagus, which is a pest in Australia and England, the virus from Sylvilagus produces a generalized disease that is almost always lethal. Myxomatosis was deliberately introduce

Wild Australian niches as a source of novel lactic acid bacteria and malolactic fermentation candidates.

Malolactic fermentation (MLF) by lactic acid bacteria (LAB) is essential for most red wine production, yet commercial starter cultures are largely limited to Oenococcus oeni, a species constrained by slow growth, fastidious nutritional requirements, and poor co-inoculation competitiveness. To identify novel MLF candidates from diverse ecological niches, we assembled 714 wild LAB strains from seven Australian geographic regions and eight ecological sources, including insects, flowers, honey, and fermented foods, spanning 21 genera. Four hundred strains were screened in a microplate assay quanti

Development of an in situ binding (ISB) method to evaluate the binding of Cry1Ac toxin to the larval midgut epithelium of Helicoverpa armigera.

The extensive use of Bacillus thuringiensis (Bt) insecticidal proteins in pest control has led to the evolution of resistance in target insects. Reduced toxin binding to the midgut epithelial membrane is considered a major resistance mechanism. Current methods for characterizing toxin binding suffer from limitations in physiological relevance or practical applicability. Here, we developed an in situ binding (ISB) method to visualize and quantify the binding of Cry1Ac toxin to the midgut epithelium of Helicoverpa armigera larvae. By culturing dissected midguts in a cell culture medium containin

Characterizing the transmission dynamics of Trypanosoma cruzi in Triatoma sanguisuga collected from dog kennels in southern Texas.

BACKGROUND: Chagas disease, caused by the protozoan Trypanosoma cruzi, remains a significant public health issue in South America, with increasing concern over its potential transmission in the USA. Triatoma sanguisuga, a triatomine vector, is found in Southern states of the USA, including Texas, raising questions about the local transmission dynamics of T. cruzi. This study aims to characterize Trypanosoma cruzi infection in Triatoma sanguisuga collected from dog kennels in Bulverde and Spring Branch, Texas, with a focus on parasite prevalence and load, genotypic diversity, and blood-feeding

Characterization of temporal expression of immune genes in female locust challenged by fungal pathogen, Aspergillus sp.

INTRODUCTION: The innate immune system provides defense against invading pathogens in insects and mammals. METHODS: We conducted transcriptomic analyses of the locust Locusta migratoria under Aspergillus oryzae infection to clarify temporal variation in its molecular immune response. RESULTS: We found that fat body cells and hemocytes play different roles in the immune response of locusts to Aspergillus infection at different time points after inoculation, and melanization was the main process underlying the immune response of female locusts. Most pattern recognition receptors (PRR) genes we

Molecular immune responses of male locusts Locusta migratoria to an Aspergillus oryzae XJ1 infection.

BACKGROUND: Many fungal pathogens infect and kill host insects successfully through the suppression of host immune systems, making them potential biological control agents. Aspergillus oryzae is a new fungal pathogen of locust Locusta migratoria, which is an important insect pest worldwide; consequently, information on the molecular mechanisms that it uses to suppress the host's immune system is limited. Here, transcriptomic analyses of male locusts during an A. oryzae infection were used to characterize the temporal variations in the host's molecular immune responses. RESULTS: Two key immune

From the fat body to the hemolymph: Profiling tick immune and storage proteins through transcriptomics and proteomics.

Ticks are blood-feeding arachnids that are known to transmit various pathogenic microorganisms to their hosts. During blood feeding, ticks activate their metabolism and immune system to efficiently utilise nutrients from the host's blood and complete the feeding process. In contrast to insects, in which the fat body is known to be a central organ that controls essential metabolic processes and immune defense mechanisms, the function of the fat body in tick physiology is still relatively unexplored. To fill this gap, we sought to uncover the repertoire of genes expressed in the fat body associa

Fast prediction in marmoset reach-to-grasp movements for dynamic prey.

Primates have evolved sophisticated, visually guided reaching behaviors for interacting with dynamic objects, such as insects, during foraging.1,2,3,4,5 Reaching control in dynamic natural conditions requires active prediction of the target's future position to compensate for visuo-motor processing delays and to enhance online movement adjustments.6,7,8,9,10,11,12 Past reaching research in non-human primates mainly focused on seated subjects engaged in repeated ballistic arm movements to either stationary targets or targets that instantaneously change position during the movement.13,14,15,16,1

A new antimicrobial peptide, Pentatomicin, from the stinkbug Plautia stali.

Antimicrobial peptides (AMPs) play crucial roles in the innate immunity of diverse organisms, which exhibit remarkable diversity in size, structural property and antimicrobial spectrum. Here, we describe a new AMP, named Pentatomicin, from the stinkbug Plautia stali (Hemiptera: Pentatomidae). Orthologous nucleotide sequences of Pentatomicin were present in stinkbugs and beetles but not in other insect groups. Notably, orthologous sequences were also detected from a horseshoe crab, cyanobacteria and proteobacteria, suggesting the possibility of inter-domain horizontal gene transfers of Pentatom

Development of fipronil resistance, fitness cost, cross-resistance to other insecticides, stability, and risk assessment in Oxycarenus hyalinipennis (Costa).

Pesticides are extensively used to control pests, diseases, and weeds in order to increase agricultural production. Usage of indiscriminate doses and persistent pesticides has not only caused resistance issues in insect pests but has also had deleterious effects on non-target organisms (beneficial insects, fish, and wildlife) and caused environmental contamination (soil, water, and air) through leaching, overflow, and insecticide spray drift. Exposure from eating food and drinking water contaminated to pesticide residues is also affecting human health. This study was conducted to obtain inform

The Effect of Radiation on the Gut Bacteriome of Aedes albopictus.

The sterile insect technique (SIT) has been developed as a component of area-wide integrated pest management approaches to control the populations of Aedes albopictus, a mosquito vector capable of transmission of dengue, Zika and chikungunya viruses. One of the key factors for the success of SIT is the requirement of high biological quality sterile males, which upon their release would be able to compete with wild males for matings with wild females in the field. In insects, gut bacteriome have played a catalytic role during evolution significantly affecting several aspects of their biology an

Metabolic signatures of triatomine vectors of Trypanosoma cruzi unveiled by metabolomics.

Chagas disease is a trypanosomiasis whose causative agent is the protozoan parasite Trypanosoma cruzi, which is transmitted to humans by hematophagous insects known as triatomines and affects a large proportion of South America. The digestive tract of the insect vectors in which T. cruzi develops constitutes a dynamic environment that affects the development of the parasite. Thus, we set out to investigate the chemical composition of the triatomine intestinal tract through a metabolomics approach. We performed Direct Infusion Fourier Transform Ion Cyclotron Resonance Mass Spectrometry on fecal

The fat body transcriptomes of the yellow fever mosquito Aedes aegypti, pre- and post- blood meal.

BACKGROUND: The fat body is the main organ of intermediary metabolism in insects and the principal source of hemolymph proteins. As part of our ongoing efforts to understand mosquito fat body physiology and to identify novel targets for insect control, we have conducted a transcriptome analysis of the fat body of Aedes aegypti before and in response to blood feeding. RESULTS: We created two fat body non-normalized EST libraries, one from mosquito fat bodies non-blood fed (NBF) and another from mosquitoes 24 hrs post-blood meal (PBM). 454 pyrosequencing of the non-normalized libraries resulted…

Susceptibility of Aedes aegypti to spinosad larvicide and space spray adulticides in Brazil.

BACKGROUND: Insecticides play a critical role in controlling insect vectors, particularly during epidemics. Effective chemical control relies on the robust monitoring of insecticide resistance to guide evidence-based decision-making in vector control strategies. OBJECTIVES: This study assessed the susceptibility of Aedes aegypti, the primary vector of dengue, Zika, and Chikungunya viruses, to various larvicides and adulticides deployed during Brazil's national campaigns from 2020 to 2023. METHODS: Mosquito collection was performed in 46 Brazilian municipalities using ovitraps. Eggs were tran

Endemic Kaposi's sarcoma: incidence and risk factors in the West Nile District of Uganda.

Out of 72 Kaposi's sarcoma patients diagnosed between 1951 and March 1976 at Kuluva Hospital, West Nile District, Uganda, 64 with known co-ordinates were plotted on a map. Sixty-two of these were noted to live at an altitude of 853 metres or more (greater than or equal to 2,800 feet). Twenty-four patients were thought to be still alive and visits were made to their homes. Space-time grouping of four cases was noted on two occasions, although analysis did not reveal statistically significant clustering. A case-control study employing an interview questionnaire with 32 variables was performed on

Induction of hyperpigmentation and heat shock protein 70 response to the toxicity of methomyl insecticide during the organ development of the Arabian toad, Bufo arabicus (Heyden,1827).

Methomyl (MET) is a carbamate insecticide which is used as a substitute for organophosphorus compounds to protect crops against insects. The present study aims to evaluate the cytoprotection response of pigment cells and heat shock protein 70 (HSP70) after exposure to MET during the tadpole developmental stages of the Arabian toad, Bufo arabicus. Three developmental larval stages of the toad were selected and divided into two groups; Control and MET-exposed (MET-EX) tadpoles (10ppm). MET-EX tadpoles showed an increased number of pigment cells in the liver, kidney, anterior eye chamber, and ski

Culex quinquefasciatus larval microbiomes vary with instar and exposure to common wastewater contaminants.

Like many insects, mosquitoes, rely on endosymbionts to grow and develop. These can be acquired from the environment. We used next generation 454 pyrosequencing to discern the whole-body microbiome of the mosquito species Culex quinquefasciatus in various larval stadia and following exposure to common pharmaceutical and personal care products (PPCPs) found in wastewater. PPCP treatments included environmentally-relevant concentrations; 1) a combination of common antibiotics, 2) a combination of mammalian hormones, 3) a mixture of the antibiotic and hormone treatments plus acetaminophen and caf

Genetic and evolutional mechanisms explain associated malformations--a 'G-E-M' concept.

During the process of evolution, segmented structures like heart and kidneys appeared earlier than vertebral column. Single piece notochord was transformed into segmented vertebral column. Ribs followed the segmented vertebral column but preceded the fore limbs in evolution. Segmentation is the underlying principle of the body plan even in annelids and arthropods. In these animals apart from vertebral column; segmentation is obvious in other structures like kidneys and heart. Somewhere on the temporal axis of evolution--vertebral column, heart and kidneys have evolved together; and have shared

Current prospects of green-metallic nanoparticles in mosquito control: A brief review.

Mosquitoes (Diptera: Culicidae) transmit many pathogens and parasites such as viruses, bacteria, protozoans and nematodes, which cause serious diseases such as malaria, dengue, yellow fever and chikungunya fever, encephalitis and filariasis. Traditionally, control of these mosquitoes has relied on the use of several chemical insecticides. These insecticides have been used to control various life stages of mosquitoes, but some of these insecticides are causing harm to the environment, human, plants and animals which in turn cause environmental pollution. Presently, nanoparticles are thought to
