Integrative omics and network pharmacology analysis of Glossocardia bosvallia L. phytocompounds for non-small cell lung carcinoma: Multi-target mechanism and molecular insights.

Non-small cell lung cancer (NSCLC) remains a leading cause of cancer-related mortality worldwide, with limited therapeutic efficacy due to tumor heterogeneity in conventional treatments. In the present study, an integrative, network pharmacology approach was employed to elucidate the multi-target mechanism of action of phytochemicals derived from Glossocardia bosvallia against NSCLC. Among 38 phytocompounds identified, 31 compounds that satisfied pharmacokinetic properties were selected for subsequent analysis. Ligand-based target prediction identified 429 potential protein targets, which are

Decoding tumor immune microenvironment heterogeneity by single-cell and spatial multi-omics: From immunotherapy resistance to translational biomarkers.

Immune checkpoint blockade has transformed cancer therapy, yet primary and acquired resistance remain major clinical challenges. Increasing evidence indicates that immunotherapy resistance cannot be fully explained by tumor-intrinsic alterations or conventional biomarkers such as PD-L1 expression, tumor mutational burden, or microsatellite instability. Instead, therapeutic response is shaped by the tumor immune microenvironment (TIME) as a heterogeneous, spatially organized, and dynamically evolving ecosystem. Single-cell omics has revealed diverse immune and stromal cell states, including pro

Enzyme-Metabolite Network Analysis of Endometrial Cancer-Derived Extracellular Vesicles Through Integrated Proteomics and Metabolomics.

Endometrial cancer (EC) is the most common gynecological malignancy in high-income countries. Extracellular vesicles (EVs) are key mediators of intercellular communication and metabolic reprogramming, but their molecular cargo in EC remains poorly characterized. EVs were isolated from four EC cell lines representing Type I and Type II subtypes (AN3CA, ISHIKAWA, HEC1A, and KLE). Untargeted metabolomics was performed by HILIC-LC-MS/MS, proteomics by data-independent acquisition (DIA) mass spectrometry, and multi-omics integration using MetaboAnalyst and OmicsNet. Metabolomic profiling identified

Multi-omics analysis revealed heterogeneity of AARS1 and AARS2 in pan-cancer and identified AARS1 as a potential prognostic biomarker for urologic neoplasms.

Lactate, the primary byproduct of glycolysis, has been established as a critical barometer of tumor microenvironment homeostasis and is implicated in tumor progression. Alanyl-tRNA synthetases AARS1 and AARS2 (AARS1/2) have been identified as sensors of intracellular L-lactate and as contributors to cancer development. Nonetheless, investigations into the different roles of AARS1 and AARS2 across various cancer types remain lacking. In this study, we preliminarily explored the correlations between AARS1/2 and the tumor microenvironment in pan-cancer. We first examined the expression patterns o

Multi-omic spatially resolved analysis of the neuroinflammatory response to intracortical microelectrode arrays.

Intracortical microelectrode arrays (MEAs) are devices implanted into the brain's cortex with the ability to record or stimulate neuronal activity. Unfortunately, MEAs tend to fail over chronic time points, limiting their clinical utility. Chronic failure has largely been attributed to the brain's neuroinflammatory response. Until recently, most of what was understood about the neuroinflammatory response to MEAs was learned through immunohistochemical analysis of small numbers of proteins. More recently, gene expression studies have sequenced thousands of mRNA molecules that contribute to neur

Multi-omics analysis reveals fluorinated liquid crystal monomers BDPrB/EDPrB exposure disrupts the steroid biosynthesis function via inhibition of CYP17A1.

Fluorinated liquid crystal monomers (F-LCMs) are emerging environmental contaminants that have been detected in human urine, breast milk, and serum. However, their potential impacts on reproductive health remain unclear. In this study, we investigated the reproductive toxicity of two major F-LCMs-BDPrB and EDPrB-using a 70-day exposure model in male ICR mice. Multi-omics analyses of testicular tissue and serum showed that both compounds reduced testosterone and increased progesterone levels. Further mechanistic studies revealed that BDPrB and EDPrB inhibit CYP17A1 enzyme activity by directly b

Integrative multi-omics identifies coordinated alterations in the gut microbiome and in plasma and aqueous humor metabolomes in high myopic cataract.

Growing evidence suggests that chronic inflammation contributes to the development of myopia, but the biological link between systemic processes and ocular changes remains poorly understood. In this study, we explored the gut-retina axis using a multi-omics approach in patients with high myopic cataract (HMC) compared with age-related cataract (ARC) controls. Fecal 16S rDNA sequencing revealed distinct microbial signatures in HMC, notably the enrichment of Lactobacillales and depletion of Roseburia, taxa associated with systemic inflammatory status. Metabolomic profiling of plasma and aqueous

Integrated multi-omics analysis dissects di-isobutyl phthalate exposure-induced male fertility impairment.

Di-isobutyl phthalate (DiBP), a member of the phthalate esters, is frequently used in manufacturing consumer and industrial products as plasticizer to improve durability and flexibility. Despite much research, little is known about the direct mechanisms by which DiBP harms the male reproductive system. In the present study, a male ICR mice model was developed to investigate the reproductive effect and mechanism of DiBP exposure, followed by transcriptomics, non-targeted metabolome, and 16S rDNA sequencing accordingly. The results showed that DiBP exposure induced male reproductive malfunction,

[Advances in animal models and multi-omics technologies for cleft palate research].

Non-syndromic cleft palate only is one of the most common congenital craniofacial malformations, arising from complex interactions between genetic and environmental factors. In recent years, animal models have been extensively utilized in cleft palate research, especially in conjunction with multi-omics technologies such as single-cell RNA sequencing, transcriptomics, proteomics, and epigenomics. These approaches have revealed multidimensional molecular mechanisms underlying cleft palate formation. Animal models provide valuable data for investigating gene regulation and intercellular communic

Redefining Malignant Hyperthermia: Multi-Omics Insights into a Complex Anesthetic Disorder.

BACKGROUND: Malignant hyperthermia (MH) is a rare but serious pharmacogenetic disorder triggered by specific anesthetic agents, leading to a rapid and often fatal hypermetabolic response. While its genetic roots - primarily involving RYR1 and CACNA1S mutations - are well documented, many susceptible individuals remain undiagnosed until they are exposed to the triggering anesthetic. Despite dantrolene being an instrumental drug in combatting MH mortality, global access remains inconsistent, and morbidity rates remains high. Current diagnostic tools are invasive and limited to specialized center
