Identification and partial characterization of a novel serpin from Eudiplozoon nipponicum (Monogenea, Polyopisthocotylea).

BACKGROUND: Serpins are a superfamily of serine peptidase inhibitors that participate in the regulation of many physiological and cell peptidase-mediated processes in all organisms (e.g. in blood clotting, complement activation, fibrinolysis, inflammation, and programmed cell death). It was postulated that in the blood-feeding members of the monogenean family Diplozoidae, serpins could play an important role in the prevention of thrombus formation, activation of complement, inflammation in the host, and/or in the endogenous regulation of protein degradation. RESULTS: In silico analysis showed

HISTOPATHOLOGICAL AND IMMUNOHISTOCHEMICAL STUDIES IN EXPERIMENTAL HETEROPHYIASIS AND THE ROLE OF PRAZIQUANTEL AND AMINOGUANIDINE.

Histopathological diagnosis was used to understand the pathological events associated with Heterophyes heterophyes (H. heterophyes) infection. CD3 and CD79α antibodies had been used as markers for both T and. B lymphocytes respectively. Immunohistochemical techniques had several advantages as remarkable sensitivity and specificity. This study aims to evaluate the roles-of praziquantel (PZQ) and aminoguanidine (AG) treatment in H heterophyes infected dogs pathologically and immunohisto-chemically. Study design included experimental infection of dogs with encysted metacercariae of H heterophyes

A comprehensive survey of larval digenean trematodes and their snail hosts in central Alberta, Canada.

Digenean trematode distributions, compatibility profiles with their snail hosts, and complete life cycles remain mysteries in many parts of the world. Surveys of digenean biology and ecology provide further insight and perspective into just how incredibly diverse and important helminth parasites are in shaping local ecosystems. Past surveys have provided substantial characterizations of adult digeneans within their definitive hosts, and many now have contributed toward furthering our understanding of larval digeneans within their intermediate host communities. However, much information about t

Transcriptome analysis of the adult rumen fluke Paramphistomum cervi following next generation sequencing.

Rumen flukes are parasitic trematodes (Platyhelminthes: Digenea) of major socioeconomic importance in many countries. Key representatives, such as Paramphistomum cervi, can cause "Rumen fluke disease" or paramphistomosis and undermine economic animal productivity and welfare. P. cervi is primarily a problem in sheep, goat and buffalo production as a consequence of reduced weight gain and milk production, clinical disease or death. Recent technological advances in genomics and bioinformatics now provide unique opportunities for the identification and pre-validation of drug targets and vaccines

Two New Species of Homalometron (Digenea: Apocreadiidae) from Nearctic Freshwater Fundulids, Elucidation of the Life Cycle of H. cupuloris, and Molecular Phylogenetic Analysis of Some Congeners.

Two species of digeneans belonging in Homalometron are described from Nearctic freshwater fundulid fishes: Homalometron robisoni n. sp. is described from the Blackstripe Topminnow, Fundulus notatus, from Oklahoma and Homalometron frocioneae n. sp. is described from the Banded Killifish, Fundulus diaphanus, from New York. Homalometron robisoni n. sp. differs from all congeners by having vitelline follicles that extend into the forebody, a feature that necessitates altering the generic diagnosis for the genus. Homalometron frocioneae n. sp. may be distinguished from North and Middle American con

Confirmation of Pseudolepidapedon balistis in the Acanthocolpidae (Digenea) based on phylogenetic analysis of ribosomal DNA.

Psuedolepidapedon balistis Manter, 1940 is reported from the grey triggerfish, Balistis capriscus Gmelin, 1789, from off Destin, Florida, in the northern Gulf of Mexico. A fragment of ribosomal DNA comprising a short portion of the 3' end of 18S nuclear rDNA gene, internal transcribed spacer (ITS) region (=ITS1 + 5.8S rDNA gene + ITS2) and the 5' end of the 28S rDNA gene (including variable domains D1-D3) was sequenced for the species and deposited in GenBank. The fragment of 28S rDNA gene was aligned with similar fragments from 25 other digenean species, including 14 species of acanthocolpids

[Ecological bases of the combination of natural foci of Trematoda infections in the floodplain-river ecosystem of the Konda River. Communication 2. Host population-combined foci of Trematoda infections].

In the context of the present-day teaching of parasitocenoses and the proposition that the pathogen's population is the only compulsory and specific component of a natural focus, the author discloses the ecological bases of the combination of natural foci of opisthorchiasis and methorchiasis (M. bilis), methorchiasis (M. bilis) and methorchiasis (M. xanthosomus). These foci are host population-combined. While analyzing the combination of foci, it is expedient to consider them in pairs since this provides a way of identifying only the combination bases that are unique to these foci. The parasit

Presence of Trichobilharzia szidati in Lymnaea stagnalis and T. franki in Radix auricularia in northeastern France: molecular evidence.

A molecular approach was used to analyse a focus of cercarial dermatitis in northeastern France (Lake Der-Chantecoq), including both cercariae and snails,by sequencing the internal transcribed spacers (ITS1 for ocellate furcocercariae and ITS2 for snails). Lymnaea stagnalis were found infected with the furcocercariae of Trichobilharzia szidati, and T. franki furcocercariae were found in Radix auricularia. The record of these two visceral parasites of birds in northern France confirms strong host-parasite relationships. The use of these standardised markers will be of the highest significance f

Trematode diversity in southern Africa: metacercariae of the Clinostomidae and Cryptogonimidae in intermediate freshwater fish hosts.

The freshwater fish fauna of southern Africa is highly diverse; however, the magnitude of parasitic species they host is unevenly known. The region's documented adult trematode fish fauna is sparse, while the opposite is evident for intermediate trematode stages. Perceived difficulty in identification of underdeveloped stages lead to the exclusion of reporting metacercariae or lack either morphological or molecular data resulting in a depauperate comparative molecular data repository for species of the region and Africa as a whole. In an effort to address the morphological and molecular data v

Systematics and phylogeny of Chalcinotrematinae (Digenea: Haploporidae): Erection of a new genus and description of two new species from South America through integrated analyses.

The subfamily Chalcinotrematinae (Haploporidae) comprises an understudied group of digenean parasites infecting freshwater and brackish water fishes across the Americas, Africa, and Asia. Its systematics has rarely been investigated using molecular data, with DNA sequences available for only a few species representing two of the six recognised genera. We collected chalcinotrematine trematodes from freshwater fishes in the Jari River, Amazon Basin, Brazil, and in Misiones, Argentina. Specimens from Brazil were characterised through an integrative approach combining morphological examination wit

Central European parasitic flatworms of the Cyclocoelidae Stossich, 1902 (Trematoda: Plagiorchiida): molecular and comparative morphological analysis suggests the reclassification of Cyclocoelum obscurum (Leidy, 1887) into the Harrahium Witenberg, 1926.

Cyclocoelidae Stossich, 1902 are medium-sized to large digenean bird parasites. Although these parasites bear few visible autapomorphic signs, and their diagnostic characters are unstable in response to the pressure applied during preparation, the numerous hitherto suggested re-classifications within the family have not been supported by any molecular analysis. We analyse here cyclocoelids found during the extensive examination of central European birds performed from 1962 to 2016, provide comparative measurements, host spectra, prevalence and intensity, and provide and analyse sequences of fo

Mapping freshwater snails in north-western Angola: distribution, identity and molecular diversity of medically important taxa.

BACKGROUND: This study was designed to determine the distribution and identity of potential intermediate snail hosts of Schistosoma spp. in Bengo, Luanda, Kwanza Norte and Malanje Provinces in north-western Angola. This is an area where infection with Schistosoma haematobium, causing urogenital schistosomiasis, is common but little is yet known about transmission of the disease. Angola has had a varied past with regard to disease control and is revitalising efforts to combat neglected tropical diseases. METHODS: Snails were sampled from 60 water-contact points. Specimens of the genera Bulinus

Gastrointestinal trichostrongylosis can predispose ewes to clinical mastitis after experimental mammary infection.

Objective was to study, in an experimental model, the possible role of gastrointestinal nematode infection in predisposing ewes to mastitis during the lactation period. Twenty-four ewes (A or B [n=12]), free from nematode and trematode helminths, were used. Group A animals received 5000 third-stage larvae of a trichostrongylid helminth cocktail and group B ewes were unparasitised controls. Animals in group A developed gastrointestinal trichostrongylosis confirmed by >500epg in faecal samples; mean epg of group B ewes were <20 (P<0.001). Ewes were challenged by deposition of Mannheimia haemolyt

Gastrointestinal Parasites of Zoonotic Importance Detected in Bats in the Conservation Area of Semuliki National Park, Western Uganda.

Bat guano may contain zoonotic parasites that contaminate the environment and/or serve as a potential source of infection to humans and animals. Repeated bat-human exposure could be a risk factor for zoonosis. To date, knowledge on the status of bat gastrointestinal parasites (GIPs) in Uganda is limited. We conducted a cross-sectional study to investigate the prevalence of bat GIP species in communities contiguous to Semuliki National Park (SNP), Bundibugyo district in western Uganda. We purposively collected faecal samples of micro- (n = 242) and megabats (n = 242) from bat roosts in communit

A genome sequence for Biomphalaria pfeifferi, the major vector snail for the human-infecting parasite Schistosoma mansoni.

BACKGROUND: Biomphalaria pfeifferi is the world's most widely distributed and commonly implicated vector snail species for the causative agent of human intestinal schistosomiasis, Schistosoma mansoni. In efforts to control S. mansoni transmission, chemotherapy alone has proven insufficient. New approaches to snail control offer a way forward, and possible genetic manipulations of snail vectors will require new tools. Towards this end, we here offer a diverse set of genomic resources for the important African schistosome vector, B. pfeifferi. METHODOLOGY/PRINCIPAL FINDINGS: Based largely on Pa

Effects of temperature and viscosity on miracidial and cercarial movement of Schistosoma mansoni: ramifications for disease transmission.

Parasites with complex life cycles can be susceptible to temperature shifts associated with seasonal changes, especially as free-living larvae that depend on a fixed energy reserve to survive outside the host. The life cycle of Schistosoma, a trematode genus containing some species that cause human schistosomiasis, has free-living, aquatic miracidial and cercarial larval stages that swim using cilia or a forked tail, respectively. The small size of these swimmers (150-350 µm) dictates that their propulsion is dominated by viscous forces. Given that viscosity inhibits the swimming ability of sm

Fasciolopsis buski (Digenea: Fasciolidae) from China and India may represent distinct taxa based on mitochondrial and nuclear ribosomal DNA sequences.

BACKGROUND: Fasciolopsis buski is a zoonotic intestinal fluke infecting humans and pigs, but it has been seriously neglected. It is yet to know whether there is any genetic diversity among F. buski from different geographical locations, particularly in sequences of nuclear ribosomal DNA (rDNA) and mitochondrial (mt) DNA. Therefore, we determined the sequences of partial 18S, the complete internal transcribed spacer (ITS) rDNA and the complete mt genome of F. buski from China, compared the rDNA and mtDNA sequences with those of isolates from India and Vietnam, and assessed the phylogenetic rela
