Bioprinting Soft 3D Models of Hematopoiesis using Natural Silk Fibroin-Based Bioink Efficiently Supports Platelet Differentiation.

Hematopoietic stem and progenitor cells (HSPCs) continuously generate platelets throughout one's life. Inherited Platelet Disorders affect ≈ 3 million individuals worldwide and are characterized by defects in platelet formation or function. A critical challenge in the identification of these diseases lies in the absence of models that facilitate the study of hematopoiesis ex vivo. Here, a silk fibroin-based bioink is developed and designed for 3D bioprinting. This bioink replicates a soft and biomimetic environment, enabling the controlled differentiation of HSPCs into platelets. The formulati

Gold-Enhanced Brachytherapy by a Nanoparticle-Releasing Hydrogel and 3D-Printed Subcutaneous Radioactive Implant Approach.

Brachytherapy (BT) is a widely used clinical procedure for localized cervical cancer treatment. In addition, gold nanoparticles (AuNPs) have been demonstrated as powerful radiosensitizers in BT procedures. Prior to irradiation by a BT device, their delivery to tumors can enhance the radiation effect by generating low-energy photons and electrons, leading to reactive oxygen species (ROS) production, lethal to cells. No efficient delivery system has been proposed until now for AuNP topical delivery to localized cervical cancer in the context of BT. This article reports an original approach devel

Early wound healing of the hard-palate mucosal harvest site using artificial dermis fixation by a transparent plate.

BACKGROUND: There are currently no guidelines for the postoperative wound management of the hard-palate donor site in cases involving mucosal harvesting. This study describes our experiences with the use of an artificial dermis for early epithelialization and transparent plate fixation in cases involving hard-palate mucosal harvesting. METHODS: A transparent palatal plate was custom-fabricated using a thermoplastic resin board. After mucosal harvesting, an alginic acid-containing wound dressing (Sorbsan) was applied to the donor site, which was then covered with the plate. After confirming he

A Safe, Fibrosis-Mitigating, and Scalable Encapsulation Device Supports Long-Term Function of Insulin-Producing Cells.

Encapsulation and transplantation of insulin-producing cells offer a promising curative treatment for type 1 diabetes (T1D) without immunosuppression. However, biomaterials used to encapsulate cells often elicit foreign body responses, leading to cellular overgrowth and deposition of fibrotic tissue, which in turn diminishes mass transfer to and from transplanted cells. Meanwhile, the encapsulation device must be safe, scalable, and ideally retrievable to meet clinical requirements. Here, a durable and safe nanofibrous device coated with a thin and uniform, fibrosis-mitigating, zwitterionicall

Imprinted hydrogels with LbL coating for dual drug release from soft contact lenses materials.

A combined strategy to control the release of two drugs, one anti-inflammatory (diclofenac sodium, DCF) and one antibiotic (moxifloxacin hydrochloride, MXF), from a soft contact lens (SCL) material, was assessed. The material was a silicone-based hydrogel, which was modified by molecular imprinting with MXF and coated by the layer-by-layer (LbL) method using natural polyelectrolytes: alginate (ALG), poly-l-lysine (PLL) and hyaluronate (HA), crosslinked with 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC). Imprinting was used to increase the amount of MXF loaded and to sustai

Immobilizing Microcystis aeruginosa and powdered activated carbon for the anaerobic digestate effluent treatment.

The environment pollution caused by livestock anaerobic digestate effluent (ADE) is becoming increasingly severe recently. In this study, immobilized technology, embedding Microcystis aeruginosa (MA) and powdered activated carbon (PAC) with sodium alginate (SA), was employed to investigate the removal performance of nitrogen (N), phosphorus (P) and dissolved organic matter (DOM) in the treatment of ADE solution. Initially, orthogonal experiment was carried out to achieve the optimal conditions of the beads fabrication with the concentration of imbedding agents (PAC-SA) of 5% (w/w) and the rati

In Situ Gel Formulation for Enhanced Ocular Delivery of Nepafenac.

Nepafenac is a water-insoluble nonsteroidal antiinflammatory drug that is available as an ophthalmic suspension (Nevanac®). Suspensions are undesirable for 2 reasons: they tend to cause foreign body sensation and lacrimation, which could limit residence time and drug bioavailability. This decreases the amount of time the drug has to reach the site of action, the cornea. Previously, we improved the solubility and ocular permeability of nepafenac by complexing the drug with hydroxypropyl-β-cyclodextrin. In this study, we used the complex to formulate an ion-activated in situ gel system using sod

Xylitol production from non-detoxified and non-sterile lignocellulosic hydrolysate using low-cost industrial media components.

Immobilized Candida tropicalis cells in freeze dried calcium alginate beads were used for production of xylitol from lignocellulosic waste like corn cob hydrolysate without any detoxification and sterilization of media. Media components for xylitol fermentation were screened by statistical methods. Urea, KH2PO4 and initial pH were identified as significant variables by Plackett-Burman (PB) design. Significant medium components were optimized by response surface methodology (RSM). Predicted xylitol yield by RSM model and experimental yield was 0.87 and 0.79 g/g, respectively. Optimized conditio

Synthesis of irregular graphene oxide tubes using green chemistry and their potential use as reinforcement materials for biomedical applications.

Micrometer length tubes of graphene oxide (GO) with irregular form were synthesised following facile and green metal complexation reactions. These materials were obtained by crosslinking of GO with calcium, zinc or strontium chlorides at three different temperatures (24, 34 and 55°C) using distilled water as solvent for the compounds and following a remarkably simple and low-cost synthetic method, which employs no hazardous substances and is conducted without consumption of thermal or sonic energy. These irregular continuous GO networks showed a very particular interconnected structure by Fiel

Transdermal anti-inflammatory activity of bilayer film containing olive compound hydroxytyrosol: physical assessment, in vivo dermal safety and efficacy study in Freund's adjuvant-induced arthritic rat model.

Previous studies have shown that hydroxytyrosol (HT) can be a potential alternative therapeutic agent for the treatment of rheumatoid arthritis (RA). However, HT is extensively metabolized following oral administration, which leads to formulating HT in a topical vehicle to prolong drug action as well as to provide a localized effect. Hidrox-6 is a freeze-dried powder derived from fresh olives and contains a high amount of HT (∼3%) and other polyphenols. Alginate bilayer films containing 5% and 10% Hidrox-6 were formulated. The films were characterized with respect to their physical, morphology

Nozzleless Fabrication of Oil-Core Biopolymeric Microcapsules by the Interfacial Gelation of Pickering Emulsion Templates.

Ionotropic gelation has been an attractive method for the fabrication of biopolymeric oil-core microcapsules due to its safe and mild processing conditions. However, the mandatory use of a nozzle system to form the microcapsules restricts the process scalability and the production of small microcapsules (<100 μm). We report, for the first time, a nozzleless and surfactant-free approach to fabricate oil-core biopolymeric microcapsules through ionotropic gelation at the interface of an O/W Pickering emulsion. This approach involves the self-assembly of calcium carbonate (CaCO3) nanoparticles at

Multifunctional medicated lyophilised wafer dressing for effective chronic wound healing.

Wafers combining weight ratios of Polyox with carrageenan (75/25) or sodium alginate (50/50) containing streptomycin and diclofenac were prepared to improve chronic wound healing. Gels were freeze-dried using a lyophilisation cycle incorporating an annealing step. Wafers were characterised for morphology, mechanical and in vitro functional (swelling, adhesion, drug release in the presence of simulated wound fluid) characteristics. Both blank (BLK) and drug-loaded (DL) wafers were soft, flexible, elegant in appearance and non-brittle in nature. Annealing helped to improve porous nature of wafer

An evaluation of leukaemia inhibitory factor as a potential therapeutic agent in the treatment of muscle disease.

The exogenous delivery of growth factors and cytokines is a potential therapeutic strategy to alleviate the degenerative effects of primary inherited myopathies such as Duchenne muscular dystrophy. The mdx mouse diaphragm is a model for examining the progressive degeneration of dystrophic muscle. We have delivered leukaemia inhibitory factor to the mdx diaphragm using slow release alginate gels. Previous studies have reported an improvement in the histology of mdx diaphragms after delivery of leukaemia inhibitory factor in a similar manner, but little attention has been paid to the mechanism b

Bedside Intraoral Scanning to Reduce Procedural Burden in Older Adults With Dementia: Nonrandomized Counterbalanced Crossover Feasibility Study.

BACKGROUND: Older adults living with dementia often have limited tolerance for intraoral procedures, which can restrict access to even basic oral health assessment in clinical care. Procedural burden-characterized by sustained intraoral manipulation, sensory overload, and limited opportunities for interruption-may represent a key barrier to equitable oral care. Intraoral scanning (IOS) enables stepwise, interruptible acquisition of oral data and may reduce this burden, but feasibility and tolerability in people living with dementia have not been systematically evaluated. OBJECTIVE: This study

Cold atmospheric plasma-modified multifunctional nanoemulsion co-delivering curcumin and resveratrol: Construction and application in tilapia preservation.

Oxidative degradation and microbial contamination cause quality deterioration of aquatic products under refrigeration, posing a major challenge to their preservation. In this study, zein-curcumin-resveratrol-sodium alginate nanoparticles (ZCR/SA NPs) were pretreated with atmospheric cold plasma (ACP) for 1-4 min. The ACP-modified NPs were further fabricated into antioxidant nanoemulsions (NEs), and the influences of ACP pretreatment on the physicochemical properties, encapsulation efficiency, and sustained-release performance of the resultant NEs were systematically explored. The ACP treatment

ROS-Triggered Disassembly of Nanoparticle-Networked Hydrogels Enables Dynamic Gating of Corneal Tight Junctions.

The blood-ocular barrier has always been a major challenge in the treatment of eye disease, such as fungal keratitis (FK). Herein, we report a reactive oxygen species (ROS)-triggered disassembly of nanoparticle-networked hydrogels enabling dynamic modulation of corneal tight junctions for enhanced drug bioavailability. The voriconazole (VCZ)-loaded fluorinated chitosan nanoparticles (FCS NPs) are incorporated as dynamic crosslinking nodes within a network formed by phenylboronic acid-modified alginate (SA-PBA) and poly vinyl alcohol (PVA), featuring a macroporous backbone interconnected by web
