Juvenile hormone affects the splicing of Culex quinquefasciatus early trypsin messenger RNA.

The full length of Culex quiquefasciatus early trypsin has been cloned and sequenced and a three-dimensional (3D) model of the enzyme was built showing that the enzyme has the canonical trypsin's active pocket containing H78, D123, S129, and D128. The biosynthesis of juvenile hormone (JH) III by the corpora allata (CA) in female Cx. quiquefasciatus is sugar-dependent. Females that were maintained on water after emergence synthesize very little JH III, JH III bisepoxide, and methyl farnesoate (MF) (3.8, 1.1, and 0.8 fmol/4 hr/CA, respectively). One hour after sugar feeding, the synthesis of JH

Cloning and functional expression of intracellular loop variants of the honey bee (Apis mellifera) RDL GABA receptor.

The insect GABA receptor, RDL (resistance to dieldrin), plays central roles in neuronal signalling and is the target of several classes of insecticides. To study the GABA receptor from an important pollinator species, we cloned Rdl cDNA from the honey bee, Apis mellifera. Three Rdl variants were identified, arising from differential use of splice acceptor sites in the large intracellular loop between transmembrane regions 3 and 4. These variants were renamed from previously, as Amel_RDLvar1, Amel_RDLvar2 and Amel_RDLvar3. When expressed in Xenopus laevis oocytes, the three variants showed no d

BBA52 facilitates Borrelia burgdorferi transmission from feeding ticks to murine hosts.

Borrelia burgdorferi, the pathogen of Lyme borreliosis, persists in nature through a tick-rodent transmission cycle. A selective assessment of the microbial transcriptome, limited to gene-encoding putative membrane proteins, reveals that bba52 transcription in vivo is strictly confined to the vector-specific portion of the microbial life cycle, with the highest levels of expression noted in feeding ticks and with swift down-regulation noted in mice. bba52 deletion did not affect murine disease as assessed by the genesis of arthritis and carditis or long-term persistence of pathogens in mice or

Distal-less and homothorax regulate multiple targets to pattern the Drosophila antenna.

The Drosophila antenna is a highly derived appendage required for a variety of sensory functions including olfaction and audition. To investigate how this complex structure is patterned, we examine the specific functions of genes required for antenna development. The nuclear factors, Homothorax, Distal-less and Spineless, are each required for particular aspects of antennal fate. Coexpression of Homothorax, necessary for nuclear localization of its ubiquitously expressed partner Extradenticle, with Distal-less is required to establish antenna fate. Here we test which antenna patterning genes a

A JA-OsMYB72-OsTAM1 module promotes (R)-β-tyrosine-mediated lignin defense against brown planthopper in rice.

Brown planthopper (Nilaparvata lugens, BPH) is a major pest that severely limits rice production. Although plant defense against BPH involves complex metabolic and structural responses, the role of non-protein amino acids (NPAAs) in this process remains poorly understood. Here, we demonstrate that (R)-β-tyrosine functions as a key defense metabolite that enhances rice resistance to BPH. Exogenous application and genetic manipulation across different rice backgrounds revealed that (R)-β-tyrosine does not directly affect insect survival but instead acts through host-mediated mechanisms. Nutritional adequacy and novel food products.

Adequate nutrition is an essential requirement for health, growth, prevention of nutritional diseases and normal physiological functions throughout life. Although there has been significant progress in agriculture, food systems, nutrition science, food insecurity and dietary insufficiency continue to be a significant public health challenge globally. This might be due to factors including limited dietary diversity, socioeconomic inequalities, infectious diseases, climate change and poor governance. This chapter thoroughly explains the concept of nutritional adequacy, its definition, evolution

What Shapes RNA Interference Responsiveness in Heteroptera (Insecta: Hemiptera)? An Evidence-Constrained Multilevel Framework for Experimental Design and Optimisation.

RNA interference (RNAi) is widely used in insect functional genomics and has potential for species-selective pest management, but its performance in Heteroptera varies among species, delivery routes, targets, and endpoints. This review evaluates processes that may constrain double-stranded RNA (dsRNA) delivery, processing, and phenotypic expression using a claim-level evidence hierarchy. Productive RNAi has been demonstrated in heteropteran lineages, particularly after injection, whereas oral and topical outcomes are more variable. Responses after oral, plant-mediated, and carrier-assisted del

Tenebrio molitor-Derived Enzyme Systems Enable Solvent-Reduced Recovery of Intracellular Polyhydroxyalkanoates.

Polyhydroxyalkanoates (PHAs) are intracellular microbial polyesters whose commercial deployment is strongly influenced by downstream processing costs and environmental burden. Here, we demonstrate a nearly solvent-free, mild aqueous strategy for the recovery of PHAs directly from wet bacterial biomass using crude enzyme systems derived from Tenebrio molitor. Under optimized conditions (0.6 wt% crude protein, pH 7.6, 40°C), near-quantitative (≥ 95%) recovery of PHB and PHB/HV and up to 60% recovery of mcl-PHA were achieved without prior biomass drying. Proteomic analysis identified abundant dig

Influenza Virus-Like Particles Displaying Leishmania donovani Promastigote Surface Antigen Induce Cross-Protective Immunity.

Virus-like particles expressing the Promastigote surface antigen (PSA) of Leishmania donovani have shown great potential as a vaccine candidate against L. donovani infection. However, their potential as a cross-protective leishmaniasis vaccine candidate has remained unexplored. In this study, we generated the L. donovani PSA-expressing influenza virus-like particle vaccines and evaluated their cross-protective potential against Leishmania amazonensis infection in mice. Virus-like particles expressing the L. donovani PSA on influenza M1 protein (LdPSA-VLP) were self-assembled in insect cells us

The multifaceted role of antimicrobial peptides in neurodegeneration: Insights from Drosophila and beyond.

Antimicrobial peptides (AMPs) are tiny proteins essential for innate immunity in various taxa, including mammals and insects. They provide defence against a wide range of pathogens, including bacteria, viruses, fungi, and parasites. Apart from their antimicrobial properties, new studies have revealed the roles of AMPs in brain ageing, neurodegeneration, and neuroinflammation. With an emphasis on their dysregulation in glial and neuronal tissues and their role in neuroinflammation, mitochondrial dysfunction, and neuronal loss, we reviewed the new function of AMPs beyond their antimicrobial acti

An Evolutionary Framework Exploiting Virologs and Their Host Origins to Inform Poxvirus Protein Functions.

Poxviruses represent evolutionary successful infectious agents. As a family, poxviruses can infect a wide variety of species including humans, fish, and insects. While many other viruses are species-specific, an individual poxvirus species is often capable of infecting diverse hosts and cell types. For example, the prototypical poxvirus, vaccinia, is well known to infect numerous human cell types but can also infect cells from divergent hosts like frog neurons. Notably, poxvirus infections result in both detrimental human and animal diseases. The most infamous disease linked to a poxvirus is s

Chitosan-melanin complex microsphere: A potential colonic delivery system for protein drugs.

The characteristics and performance of chitosan-based colon delivery systems are significantly influenced by the method of preparation. Insect chitosan-melanin complex (CMC) may offer superior attributes over traditional shrimp and crab chitosan (CS) for colon-targeted administration. This study used dung beetle CMC as the carrier matrix and comprehensively examined the impact of various crosslinking techniques on the colonic drug delivery efficacy of microspheres, encompassing drug loading, swelling, drug release behavior, adhesion, enzymatic degradation, and absorption enhancement. Identification and functional analysis of CG3526 in spermatogenesis of Drosophila melanogaster.

Spermatogenesis is a critical part of reproduction in insects; however, its molecular mechanism is still largely unknown. In this study, we identified a testis-specific gene CG3526 in Drosophila melanogaster. Bioinformatics analysis showed that CG3526 contains a zinc binding domain and 2 C2 H2 type zinc fingers, and it is clustered to the vertebrate really interesting new gene (RING) family E3 ubiquitin-protein ligases. When CG3526 was knocked down by RNA interference (RNAi), the testis became much smaller in size, and the apical tip exhibited a sharp and thin end instead of the blunt and roun

Functional Analysis of Pheromone Biosynthesis Activating Neuropeptide Receptor Isoforms in Maruca vitrata.

Insect sex pheromones are volatile chemicals that induce mating behavior between conspecific individuals. In moths, sex pheromone biosynthesis is initiated when pheromone biosynthesis-activating neuropeptide (PBAN) synthesized in the suboesophageal ganglion binds to its receptor on the epithelial cell membrane of the pheromone gland. To investigate the function of PBAN receptor (PBANR), we identified two PBANR isoforms, MviPBANR-B and MviPBANR-C, in the pheromone glands of Maruca vitrata. These two genes belong to G protein-coupled receptors (GPCRs) and have differences in the C-terminus but s

A novel asexual blood-stage malaria vaccine candidate: PfRipr5 formulated with human-use adjuvants induces potent growth inhibitory antibodies.

PfRipr is a highly conserved asexual-blood stage malaria vaccine candidate against Plasmodium falciparum. PfRipr5, a protein fragment of PfRipr inducing the most potent inhibitory antibodies, is a promising candidate for the development of next-generation malaria vaccines, requiring validation of its potential when formulated with adjuvants already approved for human use. In this study, PfRipr5 antigen was efficiently produced in a tank bioreactor using insect High Five cells and the baculovirus expression vector system; purified PfRipr5 was thermally stable in its monomeric form, had high pur

Functional analysis of odorant-binding proteins for the parasitic host location to implicate convergent evolution between the grain aphid and its parasitoid Aphidius gifuensis.

(E)-β-farnesene (EBF) is a typical and ecologically important infochemical in tri-trophic level interactions among plant-aphid-natural enemies. However, the molecular mechanisms by which parasitoids recognize and utilize EBF are unclear. In this study, we functionally characterized 8 AgifOBPs in Aphidifus gifuensis, one dominant endo-parasitoid of wheat aphid as well as peach aphid in China. Among which, AgifOBP6 was the only OBP upregulated by various doses of EBF, and it showed a strong binding affinity to EBF in vitro. The lack of homology between AgifOBP6 and EBF-binding proteins from aphi

Impact of disabled circadian clock on yellow fever mosquito Aedes aegypti fitness and behaviors.

Like other insects, Aedes aegypti displays strong daily patterns in host seeking and mating. Much of these behaviors are believed to be under the control of a circadian clock, an endogenous timekeeping mechanism relying on transcriptional/translational negative feedback loops that drive rhythmic physiology and behavior. To examine the connection between the circadian clock and various Ae. aegypti behaviors, we knocked out the core clock gene cycle using CRISPR/Cas9. We found that the rhythmic pattern and intensity of mRNA expression of seven circadian genes, including AeCyc-/-, were altered ac

Case Report: Fatal Neurotoxicity Following Resmethrin Poisoning in a Child.

Resmethrin, a type I pyrethroid insecticide, can activate sodium channels, causing neurotoxicity in both mammals and insects. Possible routes of poisoning include inhalation, dermal contact and ingestion. There are no specific symptoms for resmethrin poisoning. Until now, no antidote has been available for resmethrin. Resmethrin poisoning is rarely reported in children. Here, we report a fatal case of resmethrin poisoning that might have been caused by accidental ingestion by a 26-month-old child. He presented with neurotoxic symptoms that included vomiting, recurrent seizures, and coma. The c
