Sprayable hydrogel loaded with engineered fusion cellular vesicles for cancer recurrence suppression.

BACKGROUND: Surgical resection often leaves residual lesions that lead to tumor recurrence and high mortality across various cancer types, a challenge that is particularly pronounced in oral squamous cell carcinoma. Herein, we developed a sprayable hydrogel loaded with fusion cellular vesicles (Fus-CVs) and characterized its structural and mechanical properties for the prevention of postoperative tumor recurrence. Fus-CVs co‑express PD‑1 to engage PD‑L1 on tumor cells and CD36 to competitively bind oxidized lipids in the tumor microenvironment, thereby effectively reversing T cell exhaustion a

3D Bioprinted Cellular Scaffolds Containing Monetite for the Treatment of Critical Bone Defects.

Repairing critical bone defects is a clinical challenge that urgently needs to be addressed. 3D bioprinting strategies using bioinks composed of living cells and hydrogel biomaterials can mimic natural tissues, offering a novel repair approach. In particular, the freeform reversible embedding of suspended hydrogels (FRESH) technique employs a support bath to stabilize mechanically weak hydrogels during printing while maintaining their biocompatibility. In this study, we fabricated a 3D bioprinter, and prepared a bioink composed of monetite, sodium alginate, and hydroxypropyl methylcellulose. T

Feralgine™ a New Approach for Iron Deficiency Anemia in Celiac Patients.

BACKGROUND: Celiac disease (CD) is an immunologically-mediated disorder characterized by duodenal mucosa villi atrophy. Iron absorption is usually reduced in celiac patients making every kind of oral iron treatment unhelpful because of malasorption. Feralgine™ is a new product that has been demonstrated to be more bioavailable. As such, the aim of our study was to evaluate the absorption of Feralgine™ in adult patients with CD. METHODS: Twenty-six adults affected by Iron Deficiency Anemia (IDA), of which 14 were also affected by CD and 12 were not affected by CD, were enrolled. An oral iron a

PAM/GO/gel/SA composite hydrogel conduit with bioactivity for repairing peripheral nerve injury.

The effective repair and functional recovery after peripheral nerve injury is still a hot topic in clinical research. The aim of this study was to construct a bioactive composite hydrogel conduit as a three-dimensional scaffold for the repair of rat sciatic nerve defects. The composite hydrogel conduit was fabricated by blending different volumes of polyacrylamide, graphene oxide, gelatin, and sodium alginate (PAM/GO/Gel/SA,PGGS) and injecting into a self-made mold with an inner diameter of 2 mm and an outer diameter of 6 mm. Then the conduit was further modified with YIGSR peptide. The proper

In vivo engineering of organs: the bone bioreactor.

Treatment of large defects requires the harvest of fresh living bone from the iliac crest. Harvest of this limited supply of bone is accompanied by extreme pain and morbidity. This has prompted the exploration of other alternatives to generate new bone using traditional principles of tissue engineering, wherein harvested cells are combined with porous scaffolds and stimulated with exogenous mitogens and morphogens in vitro and/or in vivo. We now show that large volumes of bone can be engineered in a predictable manner, without the need for cell transplantation and growth factor administration.

Casting the implant for reconstruction of pectus excavatum.

Fourteen patients with pectus excavatum underwent a total of 17 operations for the insertion of subcutaneous implants aimed at camouflaging their defects. A silicone prosthesis in one patient early in the series caused severe capsular formation. Although a block of Proplast may occasionally be used with success, the rational solution to the problem is to produce a custom made Silastic implant that adheres optimally to the defect in each individual case. This retrospective study shows that a subcutaneous implant clearly improves the appearance of the chest wall in most of the patients.

Modified rice protein-polysaccharide complexes as stabilizers for high internal phase emulsions in application of 3D printing and β-carotene delivery.

This research investigated how the phosphorylation followed by polysaccharide complexation regulated the stabilization of rice protein (RP)-based high internal phase emulsion (HIPE) and improved its 3D printability. The phosphorylation modification reduced particle size of RP and increased its solubility. Xanthan gum (XG), guar gum (GG) or sodium alginate (SA) further dose-dependently (0.05-0.2%) improved the rheological and printing performance of the HIPEs, and the effects followed XG > GG > SA. There was strong electrostatic repulsion between highly electronegative XG and phosphorylated RP

An injectable pH-responsive marine polysaccharide hydrogel (AE&LF@pOA) for sequential therapy of infected diabetic wounds.

Chronic diabetic wound healing remains challenging owing to impaired angiogenesis, persistent inflammation, and a high risk of infection. To address these limitations, we developed an injectable, pH-responsive hydrogel based on dynamic crosslinking between phenylboronic acid-grafted oxidized agarose (pOA) and alginate oligosaccharide-conjugated ε-polylysine (AE). This hydrogel exhibits antibacterial and antioxidant properties, enabling on-demand drug release triggered by the acidic microenvironment of chronic wounds. To enhance therapeutic efficacy, low-molecular-weight fucoidan (LF), a potent

Engineered gastroretentive amorphous ferulate matrix: a novel raft-forming paradigm for enhanced bioavailability.

This study aimed to enhance the solubility of ferulic acid using solid dispersion techniques and develop chewable tablets that neutralize stomach acid, form a protective gel layer, prevent gastric fluid reflux, and ensure prolonged retention in the stomach with controlled release of the active ingredient. Researchers developed solid dispersions of ferulic acid using Eudragit® E PO as a carrier, with a 1:2 w/w ratio, achieving the highest solubility (39.9 mg/mL). Chewable tablets were formulated by direct compression, incorporating sodium alginate as a gelling agent, calcium carbonate for calci

Bioinspired core-shell microparticle for dual-delivery of prebiotic and probiotic for the treatment of ulcerative colitis.

Lactiplantibacillus plantarum (LP) is a well-known probiotic strain that has a beneficial effect in preventing ulcerative colitis. However, delivering a sufficient number of viable LP to the colon still face challenges due to its vulnerability to the highly complex intestinal flora ecosystem. Herein, we present a centrifuge-driven micronozzle system designed for double-layered core-shell alginate microcapsules (DAM), which can serve as an effective carrier for dual delivery of resistant starch nanoparticles (RSNP, prebiotic) and LP (probiotics) for the treatment of colitis. This system enables

Colon-targeted self-assembled nanoparticles loaded with berberine double salt ameliorate ulcerative colitis by improving intestinal mucosal barrier and gut microbiota.

Ulcerative colitis (UC) is a chronic, recurrent inflammatory bowel disease marked by disturbances in intestinal mucosal barriers, persistent inflammation, oxidative stress, and dysbiosis of the intestinal microbiota. Traditional treatments often fail to adequately address these issues, primarily targeting inflammation. To address these limitations, this study developed an innovative approach using self-assembled nanoparticles for oral administration that target colonic inflammation. Berberine hydrochloride and ursodeoxycholic acid were combined to form a double salt (BeU), enhancing solubility

Poison Turned Panacea: Arsenic Trioxide Loaded Hydrogel for Inhibiting Scar Formation in Wound Healing.

Without intervention, the natural wound healing process can often result in scarring, which can have detrimental effects on both the physical and mental well-being of patients. Therefore, it is crucial to develop biomaterials that can promote healing without scarring. Regulating the Yes-associated protein-1/PDZ-binding motif (YAP/TAZ) signaling pathway is possible to reduce excessive fibrosis of fibroblasts and proliferation of vascular endothelial cells, ultimately impacting scar formation. Arsenic trioxide (ATO), an ancient drug with medicinal and toxic properties, has shown promise in regul

Chlorella-enriched hydrogels protect against myocardial damage and reactive oxygen species production in anin vitroischemia/reperfusion model using cardiac spheroids.

Microalgae have emerged as promising photosynthetic microorganisms for biofabricating advanced tissue constructs, with improved oxygenation and reduced reactive oxygen species (ROS) production. However, their use in the engineering of human tissues has been limited due to their intrinsic growth requirements, which are not compatible with human cells. In this study, we first formulated alginate-gelatin (AlgGel) hydrogels with increasing densities ofChlorella vulgaris. Then, we characterised their mechanical properties and pore size. Finally, we evaluated their effects on cardiac spheroid (CS) p

Polymer-based nanoparticles for oral insulin delivery: Revisited approaches.

Diabetes mellitus is a high prevalence and one of the most severe and lethal diseases in the world. Insulin is commonly used to treat diabetes in order to give patients a better life condition. However, due to bioavailability problems, the most common route of insulin administration is the subcutaneous route, which may present patients compliance problems to treatment. The oral administration is thus considered the most convenient alternative to deliver insulin, but it faces important challenges. The low stability of insulin in the gastrointestinal tract and low intestinal permeation, are prob

Zenker's diverticulum complicating achalasia: a 'cup-and-spill' oesophagus.

A 72-year-old woman presented with long-standing gastro-oesophageal reflux, regurgitation of swallowed food and worsening cervical dysphagia. Fluoroscopic barium oesophagography revealed a posterolateral pharyngeal pouch (Zenker's diverticulum (ZD)) complicating a 'cup and spill' oesophageal deformity with a smoothly tapered segment at the gastro-oesophageal junction. CT and high-resolution manometry confirmed that the underlying abnormality was a massively dilated oesophagus with aperistalsis and pan-oesophageal pressurisation, consistent with a diagnosis of oesophageal achalasia (type II). Managing gastro-oesophageal reflux disease in the older patient.

Gastro-oesophageal reflux disease (GORD) in older patients presents particular problems for the clinician. Older patients may present with complications rather than with symptoms, which may be less marked than in younger patients. Extraoesophageal symptoms are also more common in this group, and this may lead to confusion over the exact diagnosis. The increased likelihood of co-pathologies and concomitant medication complicate diagnosis and management further. GORD tends to be more severe for any level of symptom severity in the older patient. Erosive oesophagitis is more common among older pe
