State-Dependent Effects of Ventromedial Prefrontal Cortex Continuous Thetaburst Stimulation on Cocaine Cue Reactivity in Chronic Cocaine Users.

Cue-induced craving is a significant barrier to obtaining abstinence from cocaine. Neuroimaging research has shown that cocaine cue exposure evokes elevated activity in a network of frontal-striatal brain regions involved in drug craving and drug seeking. Prior research from our laboratory has demonstrated that when targeted at the medial prefrontal cortex (mPFC), continuous theta burst stimulation (cTBS), an inhibitory form of non-invasive brain stimulation, can decrease drug cue-related activity in the striatum in cocaine users and alcohol users. However, it is known that there are individua

rsfMRI effects of KB220Z™ on neural pathways in reward circuitry of abstinent genotyped heroin addicts.

Recently, Willuhn et al. reported that cocaine use and even non-substance-related addictive behavior increases as dopaminergic function is reduced. Chronic cocaine exposure has been associated with decreases in D2/D3 receptors and was also associated with lower activation of cues in occipital cortex and cerebellum, in a recent PET study by Volkow's et al. Therefore, treatment strategies, like dopamine agonist therapy, that might conserve dopamine function may be an interesting approach to relapse prevention in psychoactive drug and behavioral addictions. To this aim, we evaluated the effect of

Protection against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced parkinsonism by the catecholamine uptake inhibitor nomifensine: behavioral analysis in monkeys with partial striatal dopamine depletions.

The neurotoxic effects of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine on dopamine neurons in monkeys were found to be reduced when the catecholamine uptake inhibitor nomifensine was administered during several weeks after 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine. The obtained protection was partial, leading to 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced changes in dopamine levels to 8, 16, 52 and 59% of control values in the caudate nucleus and to 10, 16, 101 and 99% in the putamen of four animals, respectively. At the same doses, 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine alon…

Brain Volume Changes in Patients with Acute Brain Dysfunction Due to Sepsis.

BACKGROUND: Sepsis-induced brain dysfunction (SIBD) is often encountered in sepsis patients and is related to increased morbidity. No specific tests are available for SIBD, and neuroimaging findings are often normal. In this study, our aim was to analyze the diagnostic value of volumetric analysis of the brain structures and to find out its significance as a prognostic measure. METHODS: In this prospective observational study, brain magnetic resonance imaging (MRI) sections of 25 consecutively enrolled SIBD patients (17 with encephalopathy and 8 with coma) and 22 healthy controls underwent vo

Gender effects in alcohol dependence: an fMRI pilot study examining affective processing.

BACKGROUND: Alcohol dependence (AD) has global effects on brain structure and function, including frontolimbic regions regulating affective processing. Preliminary evidence suggests alcohol blunts limbic response to negative affective stimuli and increases activation to positive affective stimuli. Subtle gender differences are also evident during affective processing. METHODS: Fourteen abstinent AD individuals (8 F, 6 M) and 14 healthy controls (9 F, 5 M), ages 23 to 60, were included in this facial affective processing functional magnetic resonance imaging pilot study. Effects of HIV and childhood trauma on brain morphometry and neurocognitive function.

A wide spectrum of neurocognitive deficits characterises HIV infection in adults. HIV infection is additionally associated with morphological brain abnormalities affecting neural substrates that subserve neurocognitive function. Early life stress (ELS) also has a direct influence on brain morphology. However, the combined impact of ELS and HIV on brain structure and neurocognitive function has not been examined in an all-female sample with advanced HIV disease. The present study examined the effects of HIV and childhood trauma on brain morphometry and neurocognitive function. Structural data w

Disrupted expected value and prediction error signaling in youths with disruptive behavior disorders during a passive avoidance task.

OBJECTIVE: Youths with disruptive behavior disorders, including conduct disorder and oppositional defiant disorder, show major impairments in reinforcement-based decision making. However, the neural basis of these difficulties remains poorly understood. This partly reflects previous failures to differentiate responses during decision making and feedback processing and to take advantage of computational model-based functional MRI (fMRI). METHOD: Participants were 38 community youths ages 10-18 (20 had disruptive behavior disorders, and 18 were healthy comparison youths). Model-based fMRI was u

A novel preclinical model of germinal matrix hemorrhage using neonatal rats.

BACKGROUND: Germinal matrix hemorrhage (GMH) is a neurological disorder associated with very low birth weight premature infants. This event can lead to post-hemorrhagic hydrocephalus, cerebral palsy, and mental retardation. This study developed a novel animal model for pre-clinical investigations. METHODS: Neonatal rats underwent infusion of clostridial collagenase into the right germinal matrix (anterior caudate) region using stereotaxic techniques. Developmental milestones were evaluated over 10 days, cognitive function at 3 weeks, and sensorimotor function at 4 weeks after collagenase infu

Alterations of N/OFQ and NOP receptor gene expression in the substantia nigra and caudate putamen of MPP+ and 6-OHDA lesioned rats.

It has been suggested that the opioid-like neuropeptide nociceptin/orphanin FQ(N/OFQ) and its receptor (NOPr) may contribute to Parkinson's disease. Based on this idea, the aim of our study was to investigate the involvement of the N/OFQ-NOPr system in an animal model of Parkinson's disease and to evaluate if this neuropeptidergic system is acting through mechanisms involving glutamate and/or GABA. We injected the neurotoxins MPP+ or 6-OHDA into the cerebral ventricles and 10 days later measured N/OFQ and NOPr gene expression in caudate putamen (CP) and substantia nigra (SN), by RT-PCR. A larg

Striatal morphology correlates with sensory abnormalities in unaffected relatives of cervical dystonia patients.

Structural grey matter abnormalities have been described in adult-onset primary torsion dystonia (AOPTD). Altered spatial discrimination thresholds are found in familial and sporadic AOPTD and in some unaffected relatives who may be non-manifesting gene carriers. Our hypothesis was that a subset of unaffected relatives with abnormal spatial acuity would have associated structural abnormalities. Twenty-eight unaffected relatives of patients with familial cervical dystonia, 24 relatives of patients with sporadic cervical dystonia and 27 control subjects were recruited. Spatial discrimination thr

Altered reward processing in women recovered from anorexia nervosa.

OBJECTIVE: Individuals with anorexia nervosa are known to be ascetic and able to sustain self-denial of food as well as most comforts and pleasures in life. Building on previous findings of altered striatal dopamine binding in anorexia nervosa, the authors sought to assess the response of the anterior ventral striatum to reward and loss in this disorder. METHOD: Striatal responses to a simple monetary reward task were investigated using event-related functional magnetic resonance imaging. To avoid the confounding effects of malnutrition, the authors compared 13 healthy comparison women and 13
