The Disrupted Connectivity Hypothesis of Autism Spectrum Disorders: Time for the Next Phase in Research.

During the past decade, the disrupted connectivity theory has generated considerable interest as a pathophysiological model for autism spectrum disorders (ASD). This theory postulates that deficiencies in the way the brain coordinates and synchronizes activity amongst different regions may account for the clinical symptoms of ASD. This review critically examines the current structural and functional connectivity data in ASD and evaluates unresolved assumptions and gaps in knowledge that limit the interpretation of these data. Collectively, studies very often show group alterations in what are

Blunted neural responses to reward in remitted major depression: A high-density event-related potential study.

BACKGROUND: Major depressive disorder (MDD) is a highly recurrent condition, and improving our understanding of the abnormalities that persist in remitted MDD (rMDD) may provide insight into mechanisms that contribute to relapse. MDD has been characterized by reward learning deficits linked to dysfunction in frontostriatal regions. Although initial behavioral evidence of reward learning deficits in rMDD has recently emerged, it is unclear whether these reflect impairments in neural reward processing that persist into remission. METHODS: We examined behavioral reward learning and 128-channel e

Rapid Changes in Cannabinoid 1 Receptor Availability in Cannabis-Dependent Male Subjects After Abstinence From Cannabis.

BACKGROUND: The widespread use of cannabis, the increasing legalization of "medical" cannabis, the increasing potency of cannabis, and the growing recreational use of synthetic cannabinoid 1 receptor (CB1R) full agonists all underscore the importance of elucidating the effects of cannabinoids on the CB1R system. Exposure to cannabinoids is known to result in CB1R downregulation. However, the precise time course of changes in CB1R availability in cannabis-dependent (CD) subjects after short-term and intermediate-term abstinence has not been determined. METHODS: Using high-resolution research t

Validating gene-brain-behavior associations in autism: A replication study.

BACKGROUND: Autism is characterized by heterogeneity in clinical characteristics, neuroimaging and genetic profiles. To elucidate the multi-level heterogeneity, Buch et al. (2023) identified biologically meaningful subgroups in Autism Brain Imaging Data Exchange (ABIDE). However, ABIDE contains non-biological sources of variability due to retrospectively aligned site protocols. To separate biological from site-related variability and test the robustness of these subgroups, we assessed how well their findings replicate in a deeply phenotyped, prospectively collected autism dataset (EU-AIMS LEAP

Implications of hierarchical Markov models of behavior: on irreversibility, predictability, and dimensionality.

The maturation of quantitative tools for studying the high-level structure of animal behavior, and especially tools which represent spontaneous behavior as a sequence of stereotyped and neurally well-defined 'syllables', demands that the field revisit a fundamental theoretical question: if the coarse structure of behavior can be accurately described by Markov models, what do these models really tell us about behavior? In this work, we explore the theoretical implications of these models and discuss how they allow us to quantitatively formulate questions about the sequence-like nature and effec

Genomic structural equation modeling of impulsivity and risk-taking traits reveals three latent factors associated with brain structure and development.

BACKGROUND: Impulsivity is a multifaceted transdiagnostic trait that emerges in childhood. Research has identified genetic loci and brain systems associated with different facets of impulsivity and risk-taking. However, how these genetic underpinnings overlap across different facets, and how they are associated with brain development during childhood remain unknown. METHODS: Using genomic structural equation modeling on 17 impulsivity and risk-taking traits, we identified latent factors capturing underlying genetic architecture. We then calculated polygenic scores for these factors using Adol

EHR-based machine learning for psychiatric outcomes in youth and young adults: Challenges and opportunities.

Reducing mental health struggles in adolescents and young adults is a clinical and public health priority, given the prevalence of neuropsychiatric symptoms and their potential to disrupt key life transitions and the foundation for later independence. Yet, effective early identification and support of vulnerable youth require improved stratification of risk for psychopathology and associated psychosocial difficulties. In adults, the application of machine learning (ML) methods to large-scale electronic health record (EHR) data has shown some promise for improving prediction of psychiatric outc

Integrated Investigation of Sex Differences in the Human Nociceptin/Orphanin FQ Receptor System: Postmortem Analyses and In Vivo [11C]NOP-1A Positron Emission Tomography Imaging.

BACKGROUND: The nociceptin/orphanin FQ (NOP) system modulates pain, stress, and reward, with emerging evidence for sex-dependent biology. Despite NOP's hypothesized role in psychopathologies with pronounced sex differences, little is known about sex-associated distribution of NOP receptors. METHODS: To fill this gap, we combined postmortem molecular analyses with in vivo [11C]NOP-1A PET imaging. Western blot analyses examined four NOP receptor isoforms in two postmortem cohorts, and OPRL1 mRNA expression was quantified by qPCR. Guided by these findings, [11C]NOP-1A PET data from 68 healthy ad

Longitudinal White Matter Atrophy Predicts Cognitive Decline in Veterans, With Amplified Effects in Severe Posttraumatic Stress Disorder.

BACKGROUND: The integrity of cerebral white matter (WM) is critical for cognitive function and may decline in volume with age and trauma exposure, including posttraumatic stress disorder (PTSD). This study tested whether PTSD severity modifies the longitudinal relationships between age-related WM degeneration and cognitive change in veterans. METHODS: A total of 297 post-9/11 veterans enrolled in the Translational Research Center for Traumatic Brain Injury and Stress Disorders (28 female, 269 male; ages 35 ± 10.1 years) were categorized using Clinician-Administered PTSD Scale scores, with sco

The Roles of Delayed Cortical Maturation and Lower Anticipatory Reward Activation in Predicting Addictive Screen Use in Youth.

BACKGROUND: Addictive screen use (ASU), above and beyond screen time, has been linked to significant mental health risks. Yet little is known about the neural risk factors that may associate with ASU. We examined 2 neurodevelopmental factors-cognitive control and reward-highlighted in substance use research and their links to ASU. METHODS: We utilized resting-state and monetary incentive delay (MID) task functional magnetic resonance imaging data from the Adolescent Brain Cognitive Development (ABCD) Study at baseline (Y0; ages 9-10 years) to predict addictive videogaming, addictive social me

Family-Level Brain Pattern Similarities and Their Clinical Significance in Young Offspring of Individuals With Psychotic Disorders.

BACKGROUND: Familial high-risk offspring inherently provide an unparalleled opportunity for individual-level risk prediction in psychotic disorders: a personalized forecast of their potential brain pathology should they develop psychosis, based on their own parents' brain pathology. However, whether brain patterns in offspring resembling those of their affected parents have clinical significance remains unclear. The current study aims to quantify these parent-offspring brain similarities in psychotic disorders and explore their clinical significance. METHODS: Using 3T imaging data from the Bi

Microstructural, Morphological, and Metabolic Changes Following Magnetic Resonance-Guided Focused Ultrasound Capsulotomy.

BACKGROUND: Magnetic resonance-guided focused ultrasound anterior capsulotomy (MRgFUS-AC) is an experimental, incisionless intervention for refractory obsessive-compulsive disorder (OCD) and major depressive disorder (MDD). Although its clinical efficacy has been demonstrated, the longitudinal structural and microstructural brain changes that it induces remain incompletely characterized. METHODS: A total of 33 patients with treatment-resistant OCD (n = 14) or MDD (n = 19) underwent MRgFUS-AC targeting the ventral anterior limb of the internal capsule. Altered Sensorimotor-Association Axis Patterning of Global Functional Connectivity in an Autism Subtype With Low Levels of Language, Intellectual, and Adaptive Functioning.

BACKGROUND: During early development, autism can be stratified into subtypes differentiated by non-core language, intellectual, motor, and adaptive functioning features. During toddlerhood, these subtypes show different genomic patterning effects on brain structure and function that follow early primary axes of neurodevelopmental organization. This leads to the hypothesis that early cortical patterning differences may continue to be evident between subtypes in later development within hierarchical organization along the sensorimotor-association (S-A) axis. METHODS: Standardized phenotypic mea

Divergent Pathways Taken in Adolescence Predict Embracing or Resisting Moderate-to-Heavy Drinking in Young Adulthood.

BACKGROUND: Heavy alcohol drinking during adolescence is a major public health concern and a primary risk factor for developing alcohol use disorder (AUD) in adulthood. Identifying robust, modifiable predictors that distinguish adolescents who initiate heavy drinking from those who maintain low consumption can guide targeted prevention strategies. METHODS: We analyzed longitudinal data from 285 participants (141 female and 144 male) of the NCANDA (National Consortium on Alcohol and Neurodevelopment in Adolescence) study. All participants were no-to-low drinkers at age 15 years, remained in th

Cannabis and Tobacco Co-Use Is Associated With Impaired Neurocognitive Performance in Individuals at Clinical High Risk for Psychosis.

BACKGROUND: Cannabis use is highly prevalent among people with psychotic disorders. While there is some indication that cannabis use exacerbates psychosis symptoms, its neurocognitive effects remain unclear. Co-use of cannabis and tobacco is increasing in the general population, but little is known about its effects in people with psychosis or those at risk for developing a psychotic disorder, including its effects on cognitive performance. METHODS: We analyzed data from NAPLS2 (North American Prodrome Longitudinal Study 2), a multisite prospective study of individuals at clinical high risk f

Hierarchical Brain Dynamics Associated With Remission From Major Depression Across Diverse Therapeutic Modalities.

BACKGROUND: Major depressive disorder (MDD) is a highly prevalent psychiatric disorder marked by disrupted brain dynamics. However, the neural mechanisms underlying remission remain poorly understood, particularly regarding common neural markers across diverse therapeutic interventions. Emerging evidence suggests that temporal brain dynamics and their hierarchical organization, referred to as metastates, serve as sensitive markers of individual variability across cognitive functions. In this study, we evaluated whether metastate dynamics derived from resting-state functional magnetic resonance

Threading the Needle: Practical Considerations for Merging Theory-Driven Computational Psychiatry With Data-Driven Analytics to Enhance Precision Health at Scale.

The rapidly evolving field of computational psychiatry enables quantification of specific cognitive processes and their underlying mechanisms in a translational and potentially scalable manner, using a combination of data collection via mechanistically informed behavioral tasks and theory-driven mathematical modeling. In parallel, transdiagnostic, dimensional approaches to psychiatric diagnostics, such as the Research Domain Criteria and Hierarchical Taxonomy of Psychopathology, seek to facilitate links between clinical research and real-world clinical reality, which rarely respects traditiona

Circadian-Related Hypothalamic Structure Differs by Chronotype in Bipolar Disorder.

BACKGROUND: Circadian rhythm disturbances represent a core feature of bipolar disorder (BD), with evening chronotype as a marker for poorer outcomes. We hypothesized that BD psychopathology combined with evening chronotype would be associated with structural alterations in circadian-related hypothalamic regions, particularly the suprachiasmatic nucleus (SCN), specific to BD relative to other psychiatric diagnoses. METHODS: We investigated structural neuroimaging data from the UK Biobank (113 BD, 205 major depressive disorder, 91 psychotic disorders, 199 healthy control). The SCN-containing an
