Interactions between ketamine and phencyclidine and dorsal root potentials (DRPs), evoked from the raphe nuclei.

Dorsal root potentials (DRPs) are thought to reflect presynaptic inhibition of primary afferent activity. Electrical stimulation of sites near the nucleus raphe magnus evokes two such potentials (DRP-1 and DRP-2) along dorsal roots of the lumbar and sacral spinal cord in the anaemically decerebrated cat. Dorsal root potential-2 is thought to be serotonergically mediated, while the neurochemical mediator of dorsal root potential-1 is not known. Serotonin antagonists selectively inhibit dorsal root potential-2 while the shorter latency dorsal root potential-1, which can be elicited anywhere in t

Origin and utilization of volatile fatty acids in the rat.

The arteriovenous differences in the caecum of the rat have been compared for volatile fatty acids (VFA) and for electrolytes. Our results suggest the possibility of an exchange between VFA and chloride at the level of the caecal wall, rather than a net exchange between VFA and bicarbonate; however, the role of bicarbonate or Cl- at the cellular level is still unknown. Acetate uptake by the liver was enhanced when acetate in the afferent plasma was increased in fed as in starved rats, showing that acetyl CoA synthetase was still active during starvation. A release of endogenous acetate was onl

Exploring the Role of Parietal Transcranial Magnetic Stimulation on Embodiment-Related Processes During Virtual Prosthesis Control.

Despite advancements in bionic technology, disrupted afferent and efferent pathways in individuals with limb loss may hinder prosthetic embodiment. Transcranial Magnetic Stimulation (TMS) has emerged as a promising tool for investigating and modulating sensorimotor processes, though its application in neurorehabilitation for individuals with limb differences remains limited. This pilot study explores whether TMS over parietal regions associated with the grasping-function and motor planning can enhance the sense of embodiment and influence motor behaviour. In this feasibility protocol, six heal

TGF-β and IL-4 + IL-13 induce neuroplasticity in an in vitro model of hPSC-derived sensory neurons.

Chronic type 2 inflammation is known to drive the neuroplasticity of both afferent and efferent vagal nerves innervating many organs. This results in increased neuronal density and sensitivity, possibly contributing to pathologies such as eczema and asthma. However, the mechanisms driving these neuronal changes, particularly in sensory pathways, remain poorly understood, and appropriate in vitro models for their study are lacking. Here, we describe the differentiation of sensory neurons from human pluripotent stem cells. The generation of sensory neurons was validated by verifying the expressi

Autonomic cardiovascular mechanisms linked to stress in dental practice.

Background Abundant evidence from human and animal studies indicates that the sympathetic response during mental stress preferentially involves the heart. Acute or chronic stress can lead to cardiovascular disease, including hypertension and cardiac arrhythmias. Stressors in dental practice are numerous, including drill noise, sight of a needle and pain. Numerous studies indicate that many adults suffer from dental fear or dental anxiety, and many develop dental phobia. Fear, anxiety or phobia are associated with increases in heart rate, changes in systolic blood pressure, and decreased oxygen

The gut microbiota and gut-brain axis in Alzheimer's disease: From pathogenesis to treatment.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder whose pathological course involves amyloid-β (Aβ) deposition, tau abnormalities, neuroinflammation, and neurovascular dysfunction. Interest in the microbiota-gut-brain axis does not arise because gut dysbiosis has been established as an independent initiating cause of sporadic AD, but because this axis connects modifiable peripheral factors-including diet, medication, ageing, and intestinal physiology-with barrier homeostasis, immunometabolic state, neural afferent signaling, and the brain's response to pathology. Human studi

Gastric varices: a contemporary review of vascular anatomy and therapeutic strategies.

Gastric varices (GV) are found in 20% of patients with portal hypertension and bleed infrequently, but are associated with greater blood loss, higher transfusion requirements and mortality than oesophageal varices. The vascular anatomy of GV is characterised by different afferent and efferent pathways, which vary depending on its anatomical location. Contemporary radiological classification systems complement traditional endoscopic systems and assist in selection of the optimal treatment modality in an individual patient. Endoscopic cyanoacrylate injection remains the standard therapy for acut

Topical Protection of Esophageal Mucosa as a New Treatment of GERD.

Among the various factors implicated in the pathogenesis of gastroesophageal reflux disease (GERD), visceral hypersensitivity and mucosal resistance have been recently re-evaluated in relation to the increasing phenomenon of proton pump inhibitor failure, particularly in patients with nonerosive reflux disease (NERD). Intensive research has allowed us to understand that noxious substances contained in the refluxate are able to interact with esophageal epithelium and to induce the elicitation of symptoms. The frequent evidence of microscopic esophagitis able to increase the permeability of the

Type II Cells in the Human Carotid Body Display P2X7 Receptor and Pannexin-1 Immunoreactivity.

The carotid body is a peripheral chemoreceptor that consists of clusters of chemoreceptive type I cells, glia-like type II cells, afferent and efferent nerves, and sinusoidal capillaries and arterioles. Cells and nerves communicate through reciprocal chemical synapses and electrical coupling that form a "tripartite synapse," which allows for the process of sensory stimuli within the carotid body involving neurotransmission, autocrine, and paracrine pathways. In this network there are a variety of neurotransmitters and neuromodulators including adenosine 5'-triphosphate (ATP). Carotid body cell

Time series classification of multi-channel nerve cuff recordings using deep learning.

Neurostimulation and neural recording are crucial to develop neuroprostheses that can restore function to individuals living with disabilities. While neurostimulation has been successfully translated into clinical use for several applications, it remains challenging to robustly collect and interpret neural recordings, especially for chronic applications. Nerve cuff electrodes offer a viable option for recording nerve signals, with long-term implantation success. However, nerve cuff electrodes' signals have low signal-to-noise ratios, resulting in reduced selectivity between neural pathways. Th

Chronic cough as a disease.

Chronic cough is a frequent reason for medical consultation and has significant impact on quality of life. Due to the limited effectiveness of currently available treatments, and delays in accessing care, patients are often inadequately managed. There remains an overreliance by clinicians on outdated management algorithms, addressing chronic cough only as symptom of other medical conditions, and advocating investigation and trials of treatment of diseases which are often not present. This may lead to unnecessary cost, frustration and potential harm. Newer clinical guidelines in essence conside

Vagus nerve stimulation: An update on a novel treatment for treatment-resistant depression.

In this review, we provide an overview of essential clinical trials examining the effect of vagal nerve stimulation (VNS) in treatment-resistant depression (TRD), the applicable neuroanatomy of the vagus nerve, and the proposed mechanism of action (MOA) of VNS in TRD. Vagal nerve stimulation (VNS) is currently the only FDA-approved neurostimulation treatment for severe treatment-resistant depression (TRD). The implanted VNS device sends electrical impulses to the left cervical vagus nerve, resulting in stimulation of afferent vagal brainstem pathways known to be associated with mood regulation

The mechanism of action of pulsed radiofrequency in reducing pain: a narrative review.

Pain from nervous or musculoskeletal disorders is one of the most common complaints in clinical practice. Corticosteroids have a high pain-reducing effect, and their injection is generally used to control various types of pain. However, they have various adverse effects including flushing, hyperglycemia, allergic reactions, menstrual changes, immunosuppression, and adrenal suppression. Pulsed radiofrequency (PRF) is known to have a pain-reducing effect similar to that of corticosteroid injection, with nearly no major side effects. Therefore, it has been widely used to treat various types of pa

Short-latency somatosensory-evoked potentials demonstrate cortical dysfunction in patients with Angelman syndrome.

BACKGROUND: Angelman syndrome (AS) is neurodevelopmental disorder, causal gene of which is maternally expressed UBE3A. A majority of patient … normal latencies of subcortical components indicated cortical dysfunction rather than impairment of afferent pathways in AS Del patients, which might be attributed to GABAergic dysfunction due to loss of UBE3A function and heterozygosity of GABARs.

Neural pathways involved in infection-induced inflammation: recent insights and clinical implications.

Although the immune and nervous systems have long been considered independent biological systems, they turn out to mingle and interact extensively. The present review summarizes recent insights into the neural pathways activated by and involved in infection-induced inflammation and discusses potential clinical applications. The simplest activation concerns a reflex action within C-fibers leading to neurogenic inflammation. Low concentrations of pro-inflammatory cytokines or bacterial fragments may also act on these afferent nerve fibers to signal the central nervous system and bring about earl

Mechanisms of craniofacial pain.

Aim To provide an overview of mechanisms underlying craniofacial pain; to highlight peripheral and central adaptations that may promote chronification of pain in craniofacial pain states such as migraine and temporomandibular disorders (TMD). Background Pain is a common symptom associated with disorders involving craniofacial tissues including the teeth and their supporting structures, the temporomandibular joint and the muscles of the head. Most acute painful craniofacial conditions are easily recognized and well managed, but others, especially those that are chronic (e.g., migraine, TMD and

Central inhibitory effect of intravesically applied botulinum toxin A in chronic spinal cord injury.

AIMS: We evaluated a putative central inhibitory effect of intravesical botulinum toxin A (BoNT-A) on the activity of lumbosacral spinal neurons in a chronic spinal cord injury (SCI) model of bladder overactivity. METHODS: Female Sprague-Dawley rats underwent T8 spinal cord transection. Four weeks later, once overactive neuropathic detrusor pathways had developed, the animals underwent intravesical instillation with either saline (1 ml) or BoNT-A (Botox®, 20 U/1 ml) for 1 hr. Two days later, the rats then completed a cystometric evaluation prior to spinal cord harvest. Sections from the L4-S1

Neural control of the female urethral and anal rhabdosphincters and pelvic floor muscles.

The urethral rhabdosphincter and pelvic floor muscles are important in maintenance of urinary continence and in preventing descent of pelvic organs [i.e., pelvic organ prolapse (POP)]. Despite its clinical importance and complexity, a comprehensive review of neural control of the rhabdosphincter and pelvic floor muscles is lacking. The present review places historical and recent basic science findings on neural control into the context of functional anatomy of the pelvic muscles and their coordination with visceral function and correlates basic science findings with clinical findings when poss

The effect of intravesical oxybutynin on the ice water test and on electrical perception thresholds in patients with neurogenic detrusor overactivity.

AIMS: The C-fiber-mediated bladder-cooling reflex and the determination of the current perception thresholds (CPTs) permit to investigate afferent LUT pathways. They have both been proposed to detect and differentiate neurologic bladder dysfunction. This study evaluates, prospectively, the effect of oxybutynin, an antimuscarinic with direct antispasmodic effect on smooth muscle, on repeated ice water test (IWT) and CPTs in patients with a known incomplete neurogenic bladder. METHODS: Patients with a known incomplete lesion of the bladder innervation, detrusor overactivity during cystometric b

The role of the basal ganglia in data processing.

Complex cerebral activities are likely to be composed of massively repeated simple data processing tasks since the cortical data processing unit, the cortical mini-column, is found throughout the cortex with only minor variations. It has been proposed that one task performed by the cortical mini-column may be to match afferent sensory data to learnt datasets in a process known as automatic association. We hypothesize that basal ganglia circuits, through the relative signal of the nigro-striatal and striato-pallidal pathways, determine the matching threshold for dataset matching within cortical
