Therapeutic potential of spray-based delivery system for the treatment of dermatological disorders: clinical insights and formulation advances.

Dermatological disorders such as psoriasis, acne, fungal infections, and chronic wounds represent a persistent therapeutic challenge owing to their chronicity, recurrence, and the highly organized structure of the stratum corneum, which restricts effective drug penetration. Conventional topical formulations often exhibit several limitations, including non-uniform drug distribution, dose variability, limited residence time, and suboptimal patient compliance. Spray-based drug delivery systems have emerged as a promising alternative by enabling non-contact application, improved drug deposition, e

Bioinspired antimicrobial glass coatings for clear and infection-resistant surfaces.

Optically transparent glass surfaces that simultaneously prevent fogging, resist contamination, and eliminate microbial growth are highly desirable for medical optics, protective equipment, and advanced architectural applications. In applications where sustained visual clarity and resistance to microbial colonization are critical for diagnostic accuracy and patient safety, as well as in self-cleaning glass panels that reduce water consumption and reliance on chemical surfactants, multifunctional surface coatings are highly desirable. Here, we report a mussel-inspired, single-step dip-coating s

Exophiala dermatitidis Eye Infection: Case Report and Literature Review.

Exophiala endophthalmitis of exogenous origin is an exceptionally rare but severe ocular infection, characterized by diagnostic delays, limited therapeutic guidance, and frequently poor outcomes. Herein, we report one new case of an 80-year-old woman who presented with severe fungal keratitis progressing to endophthalmitis two years after an uncomplicated cataract surgery. The condition was initially misdiagnosed and treated with topical antibiotics and corticosteroids. By cultivation, microscopy, histopathological, and PCR analysis of the samples, Exophiala dermatitidis was identified as the

Duplication of superoxide dismutase and a mutation in aquaglyceroporin mediates the sensitivity of Plasmodium falciparum to cryptosporin, a natural product derived from Acaromyces ingoldii.

Cryptosporin, a fungal metabolite, exhibited potent antimalarial activity against both asexual blood stage Plasmodium falciparum and liver-stage Plasmodium berghei with minimal human HepG2 toxicity. Unlike atovaquone, cryptosporin's mechanism is independent of mitochondrial electron transport. Minimum inoculum of resistance showed a low risk of resistance development. RNA-Seq analysis revealed the upregulation of genes associated with sexual development including many canonical markers such as Pfs25, and PfCCp3, suggesting a stress response that is also seen when parasites are treated with art

From overlap to resolution: Cellular solid-state NMR at ultrahigh-field 1.5 GHz demonstrated on fungal cell walls.

Ultrahigh-field solid-state NMR in the GHz frequency range has opened new frontiers for probing complex, heterogeneous biological systems with unprecedented spectral resolution. Here, we demonstrate the advantages of a 1.5 GHz (35.2 T) NMR spectrometer for cellular solid-state NMR by quantitatively assessing 13C linewidth improvements in intact, living fungal cells of Aspergillus fumigatus. High-quality 2D13C-13C correlation spectra were acquired within 2-5 h, showing consistent linewidth narrowing of 0.05-0.15 ppm under idealized conditions and exceeding 0.2 ppm under realistic, power-limited

Isolation and characterization of an entomopathogenic fungus for Nilaparvata lugens control and its effects on host-associated bacterial community.

The brown planthopper, Nilaparvata lugens, is a major rice pest causing significant yield losses across Asia. Concerns over the environmental, health, and resistance-related impacts of current chemical control strategies highlight the need for sustainable alternatives. In this study, we isolated and characterized a novel entomopathogenic fungus, strain NLS-1, from mycosed N. lugens cadavers. Based on morphological observations and internal transcribed spacer (ITS) sequence analysis, strain NLS-1 was assigned to Aspergillus section Flavi and designated as Aspergillus sp. NLS-1. Cinnamic acid-butenedioic lactone derivatives as crop protection agents: synthesis, bioactivity, and molecular docking studies.

BACKGROUND: The rapid spread of pesticide resistance in agricultural pests and plant pathogens necessitates the development of novel lead compounds with multi-target biological activity. To address this challenge, a series of cinnamic acid-butenedioic lactone derivatives were designed and synthesised based on an active-subsystem assembly strategy. RESULTS: All compounds were structurally characterised by 1H NMR, 13C NMR and high-resolution mass spectrometry (HRMS), and their insecticidal and antifungal activities were systematically evaluated. Several derivatives exhibited strong aphicidal ac

Heat shock transcription factor splicing variant TtHSF2β-I regulates lignocellulose and lignin-related compound degradation in Trametes trogii S0301.

White-rot fungi are the primary biological decomposers of lignocellulosic biomass owing to their powerful lignin-modifying enzyme (LMEs) system. Here, we analyzed the regulatory role of TtHSF2β-I, an alternatively spliced variant of heat shock transcription factor (HSF) in Trametes trogii S0301, at three levels of substrate complexity: native poplar wood, synthetic lignocellulosic media, and specific lignin-derived monomers. Overexpression of TtHSF2β-I resulted in extensive disruption of poplar wood lignocellulose, whereas its deletion impaired fungal growth and substrate degradation. Transcri

Design and Evaluation of a Thiourea-Based Sensor for Hg2+ Detection and Antimicrobial Activity.

In this work, a new thiourea-based compound, TH4, was synthesized and its structure was investigated through FTIR, 1H NMR and DFT analyses. The sensing properties of TH4 were systematically assessed against a wide range of metal ions to determine its selectivity and sensitivity. Among these, TH4 exhibited a pronounced fluorescence "turn-on" response selectively in the presence of Hg2+, indicating a strong and specific interaction. The formation of a stable TH4-Hg2+ complex was supported by Job's plot analysis, which revealed a 1:2 binding stoichiometry. The sensor demonstrated excellent sensit

Effects of a sequential application of plant protection products on soil microbes and free-living nematodes in a field experiment.

During crop growth cycle, several different plant protection products (PPPs) are often applied in combination or sequentially. Such sequential applications result in unintentional mixtures of residues that may affect ecosystem services supported by non-target organisms such as soil microbes and nematodes. This scenario of sequential PPP application is frequent in agricultural practice but rarely addressed experimentally at field scale with regard to environmental impacts. The objective of this study was to evaluate the effect of individual and sequential application of three PPPs (the herbicid

Investigation of Pine Wilt Disease in Chongqing: From Field Occurrence and Genetic Diversity to Endophytic Microbial Composition and Functional Analysis.

Pine wilt disease (PWD), caused by Bursaphelenchus xylophilus, is a destructive forest disease leading to rapid mortality. Although Chongqing is a major epidemic region in China, the population genetic structure of B. xylophilus and the ecological interactions among nematode occurrence, blue stain formation, and microbial community dynamics remain insufficiently clear. This study systematically surveyed nematode incidence and performed morphological and molecular identification, revealing strong correlations between nematode presence, blue stain, and insect infestation (p < 0.0001). Artificial intelligence-based phenotypic screening using Aspergillus oryzae identified a novel antifungal RK-1205-I.

When treated with various compounds, fungi exhibit characteristic morphological changes depending on their mode of action. Previously, we constructed morphology-based databases of the rice blast fungus Pyricularia oryzae and human pathogenic fungus Candida albicans and used them for antifungal screening. As these databases are manually created by human experts, objectivity and throughput may be a concern. To overcome these limitations, we developed a new artificial intelligence (AI)-based automated classification system for morphological changes in the filamentous fungus Aspergillus oryzae. Us

Synergistic Effects of Cepharanthine and Amphotericin B Against Candida albicans by Targeting PMP3-Mediated Sphingolipid Biosynthesis and Membrane Disruption.

Candida albicans is a major opportunistic fungal pathogen causing various mucosal infections, including oral candidiasis, poses challenges in current antifungal therapy because of limited number of drugs, increased pathogen resistance, and notable toxicity associated with existing medications. Combination therapy is crucial for optimizing treatment outcomes. In this study, cepharanthine (CEP) was explored as an adjunct to amphotericin B (AmB) to elucidate the potential mechanism of action of drug combinations against Candida albicans. The minimum inhibitory concentration (MIC) was determined u

Phytochemicals as Novel Antifungal Agents Against Candida species.

Infections caused by Candida, including vulvovaginal candidiasis (VVC) and invasive candidiasis (IC), are a growing public health problem, exacerbated by multidrug resistance, biofilm persistence, and the limited development of antifungal drugs. In this review, we discuss plant-derived natural products with potent anti-Candida activity, specifically terpenoids, alkaloids, flavonoids, phenolics, and their nanoformulations. Many compounds, including berberine, artemisinin, thymol, eugenol, carvacrol, quercetin, catechins, lawsone, and caffeic acid, have shown the ability to modulate the fundamen

Iatrogenic Cushing's Syndrome and Fatal Adrenal Crisis Following Systemic Corticosteroid Therapy: A Case Series of Five Patients.

Corticosteroids are indispensable in the management of inflammatory, autoimmune, and respiratory disease, yet prolonged or unsupervised systemic use can produce iatrogenic Cushing's syndrome, and abrupt withdrawal risks life-threatening adrenal crisis. We report five male patients, aged 12 to 65 years, who developed Cushingoid features following systemic corticosteroid exposure for type two diabetes-associated dermatosis, bronchial asthma, epileptic encephalopathy, rheumatoid arthritis, and osteoarthritis. All five presented with moon facies; additional findings included purple striae, cutaneo

Immune Modulation and Evasion by Cryptococcus spp.: An Integrative Review of Molecular Mechanisms and Experimental Evidence.

The Cryptococcus species complex are major human pathogenic fungi capable of causing cryptococcosis, a severe pneumonia and meningitidis in both immunocompromised and, in some cases, immunocompetent patients. Their pathogenic success depends not only on classical virulence factors, but also on a coordinated capacity to evade, modulate, and exploit host immune responses across multiple stages of infection. In this review, we synthesize experimental evidence on the principal mechanisms that support cryptococcal immune evasion, integrating findings from molecular, cellular, and host-pathogen inte

Natural cyclic depsipeptides and their analogs as potential antimicrobial candidates: mechanistic insight.

Microorganisms such as bacteria, viruses, fungi, and parasites cause serious infections in humans. The misuse of antimicrobial treatments in humans, animals, and agriculture has led to the development of resistant pathogens that can survive and multiply even in the presence of antibiotics. The rise of antibiotic-resistant pathogens has caused significant loss of life and impacted social and economic conditions. Antimicrobial peptides exert antibacterial activity through multiple modes of action. Cyclic peptides are known for greater stability and bioavailability than linear peptides. Among the

Targeting the Candida albicans agglutinin-like sequence 3 adhesin to disrupt oral polymicrobial bacterial-fungal biofilms: mechanisms and therapeutic strategies.

Oral polymicrobial biofilms formed between Candida albicans and bacterial pathogens underlie early childhood caries, denture stomatitis, periodontal disease, and oropharyngeal candidiasis, infections that conventional antimicrobials consistently fail to eradicate. The agglutinin-like sequence 3 (Als3) protein is a GPI-anchored hypha-specific adhesin that serves as the principal molecular scaffold for interkingdom co-aggregation in these biofilms. This review examines the molecular architecture and domain organization of Als3, including the N-terminal peptide-binding cavity (PBC) and the amyloi
