Plant sterol ester-modified naringenin liposomes: A novel strategy for effectively penetrating the mucus barrier.

Liposomes (LPs) have attracted attention for their potential in oral drug delivery, including the delivery of compounds like naringenin (Nar), a flavonoid found in citrus fruits. However, they encounter obstacles when crossing the mucus and epithelial barriers of the gastrointestinal tract. Plant sterol esters (SE), beyond their antioxidant and immunomodulatory properties, demonstrate unique capabilities in modulating phospholipid bilayer characteristics. Herein, we have developed a Nar loaded SE-LP (SE-Nar-LP) utilizing SE as a cholesterol substitute to effectively overcome the obstacles face

Adrenal glands as secondary target organs of inhaled benzalkonium chlorides.

Benzalkonium chloride (BKC) is a quaternary ammonium compound widely used in aerosol disinfectants and sanitizers, raising concerns about it … mulation per organ weight among extrapulmonary secondary organs, while oral exposure showed minimal gastrointestinal absorption. Repeated exposures of BKC at doses of 50 and 100 μg/rat (10 times with three-day intervals) induced no pulmonary inflammation but elicited mild systemic responses, including increased white blood cell counts. However, other systemic toxicity-relate …

Unlocking the potential of microfluidic assisted formulation of exenatide-loaded solid lipid nanoparticles.

Exenatide, a first-in-class GLP-1 receptor agonist, is used to control glycaemic levels in type 2 diabetes. There are two approved injectable formulations: one solution for immediate action and one dispersion for prolonged action. Oral exenatide has low bioavailability due to poor gastrointestinal stability and absorption. To address these obstacles, we designed Solid Lipid Nanoparticles (SLN) including DOTAP in the formulation to yield high exenatide encapsulation by hydrophobic ion pairing and DSPE-PEG2kDa to convey colloidal stability and mucus diffusivity. The microfluidic production of SL

Phytochemical characterization and insight into the in vitro anthelmintic activity of Artemisia brevifolia Wall ex Dc. against the different life stages of the gastrointestinal nematode of sheep.

Gastrointestinal nematodes (GINs) are the major constraint to the production of small ruminants worldwide, causing significant losses in growth, productivity, and overall health. Clinical infections may linked to anaemia, diarrhoea, anorexia, oedema, and even mortality, whereas subclinical infections result in hidden production losses. Synthetic drugs are the primary means of control; however, their indiscriminate use has led to the emergence of parasite strains resistant to multiple classes of anthelmintics, this has created strong need for sustainable alternatives. Ethnomedicinal plants have

Comparative evaluation of two herbal formulas for gastrointestinal function and gut microbiota modulation in rats with loperamide-induced dyspepsia.

Dyspepsia is a prevalent refractory condition that arises from various causes and lacks definitive treatment. There is an urgent need for evidence to support the use of herbal medicines in the treatment of gastroenterological disorders. This study aimed to compare the therapeutic effects of two common herbal formulas-namely, Shihosogan-tang (SST) and Yijung-tang (YJT)-on loperamide (LOP)-induced dyspepsia and to explore their potential mechanisms. A dyspepsia model was established using Sprague-Dawley rats by intraperitoneal LOP injection at a dose of 3 mg/kg/day for one week. During this peri

Evaluating spleen volume in inflammatory bowel disease.

Crohn's disease (CD) and ulcerative colitis (UC), known as inflammatory bowel disease (IBD), are characterized by immune system dysregulation. The spleen holds a primary role in systemic inflammation and immune responses. Splenic involvement or splenomegaly in IBD patients may result from secondary causes, such as portal hypertension, myeloproliferative diseases, amyloidosis, splenic abscesses or granulomas. Current research on the direct association between IBD and spleen volume (SV) has expanded significantly. In CD, SV is predominantly increased, and is associated with worsen clinical outco

The Impact of Bariatric Surgery on the Development and Progression of Dermatologic Diseases: A Narrative Review.

Obesity is a major global health concern characterized by excessive fat accumulation, which significantly increases the risk of numerous comorbidities. While lifestyle modifications and pharmacotherapy are commonly employed, bariatric surgery is recognized as a highly effective treatment option. These procedures alter gastrointestinal anatomy, restricting food intake and modifying nutrient absorption, thereby reducing hunger and increasing satiety. Beyond weight reduction, bariatric surgery can improve or resolve obesity-related conditions, including type 2 diabetes, hypertension, sleep apnea,

Innovative Famciclovir Eye Drop Formulations for Herpes Zoster Infections.

PURPOSE: Herpes zoster ophthalmicus, a manifestation of the varicella-zoster virus in the eye, presents significant clinical challenges. As there is no direct ocular treatment for this infection, new therapeutic options should be studied. Famciclovir, a widely used antiviral drug for herpes zoster, offers higher bioavailability than acyclovir (a common herpes zoster treatment). However, this drug is currently limited to oral dosage forms, which are associated with inherent limitations such as variable absorption or gastrointestinal side effects. This study aimed to formulate new famciclovir ey

HIF-PHD inhibitor desidustat ameliorates iron deficiency anemia.

Iron deficiency anemia is caused by many pathological conditions like chronic kidney disease (CKD), inflammation, malnutrition and gastrointestinal abnormality. Current treatments that are erythropoiesis stimulating agents (ESAs) and iron supplementation are inadequate and often lead to tolerance and/or toxicity. Desidustat, a prolyl hydroxylase (PHD) inhibitor, is clinically used for the treatment of anemia with CKD. In this study, we investigated the effect of desidustat on iron deficiency anemia (IDA). IDA was induced in C57BL6/J mice by iron deficient diet feeding. These mice were then tre

Efficacy of vitamin C with Fe supplementation in patients with iron deficiency anemia: a systematic review and meta-analysis.

Oral iron (Fe) supplementation is one of the mainstays of treatment for iron deficiency anemia (IDA). However, its therapeutic effects are limited when there is poor absorption from the gastrointestinal tract. Vitamin C is hypothesized to improve uptake when combined as an adjunct agent. We aimed to determine the difference in hematologic outcomes in patients with IDA receiving oral iron, with or without vitamin C. MEDLINE, Embase, Web of Science, and Cochrane Central Register of Controlled Trials were searched from database inception to July 2023 for studies investigating the use of oral iron

Amino acid solution mitigates hypothermia response and intestinal damage following exertional heat stroke in male mice.

Increased gut permeability is implicated in the initiation and extent of the cytokine inflammatory response associated with exertional heat stroke (EHS). The primary objective of this study was to determine if a five amino acid oral rehydration solution (5AAS), specifically designed for the protection of the gastrointestinal lining, would prolong time to EHS, maintain gut function and dampen the systemic inflammatory response (SIR) measured during EHS recovery. Male C57/BL6J mice instrumented with radiotelemetry were gavaged with 150 μL of 5AAS or H2 O, and ≈12 h later were either exposed to a
