Dietary phytosterol alleviates adverse effects caused by high-carbohydrate diets on growth performance, antioxidant capacity, glucolipid metabolism, and metabolomics in largemouth bass (Micropterus salmoides) juveniles.

High-carbohydrate diets (HCD) usually lead to metabolic syndrome in many fish species. However, the role of phytosterol (PS) in alleviating HCD effects on largemouth bass (Micropterus salmoides) is currently unknown. This study investigated the effects of dietary PS on growth performance, antioxidant capacity, histopathology, glucolipid metabolism, and metabolomics in juvenile largemouth bass fed a 5% corn-starch-enriched diet. Fish weighing 11.14 ± 0.48 g were fed a control diet (CON), a 5% corn-starch-enriched diet (HCD), and the same corn-starch-enriched diet supplemented with PS (0.025%, 0

Western diet induces iron-dependent enteric neurodegeneration via ferroptosis.

The Western diets (WD), high in saturated fats such as palmitic acid (PA), promotes enteric neurodegeneration and motility disorders. Using murine models, in vitro systems, and human myenteric ganglia, we investigated whether a WD and PA drive iron-dependent ferroptotic injury in the enteric nervous system (ENS). Mice were fed a control diet (CD) or a WD for 12 weeks, with or without systemic AAV9-MaCPNS2 delivery of Nfe2l2 to enteric neurons. Colonic motility was assessed by a bead expulsion assay. We assessed ferroptosis using convergent readouts including iron dysregulation (transferrin rec

Activation of autophagy through Sirt1/AMPK/mTOR mediates the protective effects of fraxin against insulin resistance and steatosis in rats.

Insulin resistance (IR) and hepatic steatosis are key pathological features of type 2 diabetes mellitus (T2DM). Fraxin, a natural flavonoid with potential metabolic regulatory properties, has been proposed to modulate metabolic disorders. This study aimed to investigate whether fraxin protects against IR and steatosis via Sirt1/AMPK/mTOR-mediated autophagy activation. A T2DM rat model was established using high-fat diet (HFD) feeding combined with streptozotocin (STZ) injection. Rats were treated with fraxin (25 or 50 mg/kg) for 4 weeks, followed by assessment of fasting blood glucose (FBG), i

Metformin induces ferroptosis associated with lipidomic remodeling in AML.

Metabolic reprogramming is a hallmark of cancer and is essential for sustaining leukemogenesis. In acute myeloid leukemia (AML), a high dependency on oxidative phosphorylation (OXPHOS) is often linked to poor outcomes, and its inhibition has shown to be highly effective. However, most OXPHOS inhibitors are not clinically translatable because of significant side effects. Thus, repurposing safe US Food and Drug Administration-approved drugs that can target OXPHOS is of great interest. Here, we evaluated metformin, an antidiabetic drug that inhibits OXPHOS, in a genetically diverse panel of prima

Chronic kidney disease induces distinct alterations of macrophage lipid metabolism in a mouse model of atherosclerosis.

Chronic kidney disease (CKD) is associated with altered lipid metabolism and chronic inflammation, which both contribute to an accelerated risk of atherosclerotic cardiovascular disease. Macrophage polarization towards a pro-inflammatory phenotype plays a key role in atherosclerotic cardiovascular disease development and is mediated by a rewiring of macrophage immunometabolism. While prior studies have investigated associations between the systemic lipidome and CKD-accelerated CVD, the impact of CKD on macrophage lipid metabolism remains unknown. In this study, we profiled the macrophage lipid

Inhibition of TWEAK/Fn14 Ameliorates Sarcopenic Obesity by Restoring Mitochondrial Biogenesis via the AMPK/SIRT1/PGC-1α Axis.

PURPOSE: Sarcopenic obesity (SO) represents a critical geriatric syndrome where muscle atrophy and obesity synergistically exacerbate metabolic dysfunction. While the TWEAK/Fn14 pathway is a known regulator of muscle wasting, its specific role and therapeutic potential in the pathogenesis of SO remain unclear. METHODS: To mimic the lipid-rich microenvironment of SO, we utilized a "PrePA-Diff" in vitro model where C2C12 myoblasts were exposed to palmitic acid prior to differentiation. In vivo, SO was established by feeding aged (18-month-old) male C57BL/6 mice a high-fat diet (HFD) for 12 week

The insulin-like growth factor 2 mRNA-binding protein 2 affects tumor cell metabolism via mitochondrial transporter activity and lipid alterations.

The insulin-like growth factor 2 mRNA-binding protein (IGF2BP) family is overexpressed in cancer and associated with poor prognosis. IGF2BP2 has been linked to single metabolic alterations by acting on its RNA targets. Here, we used a comprehensive approach to elucidate the effects of IGF2BP2 on primary and lipid metabolism. 13C-metabolic flux analysis (MFA) combined with RNA-Seq data revealed that IGF2BP2 affects mitochondrial fluxes by regulating the expression of several mitochondrial transporters, such as mitochondrial pyruvate carrier 1 (MPC1) and uncoupling protein 2 (UCP2). Methyl pyruv

Upregulation of SQSTM1 Regulates Ferroptosis and Oxidative Stress in Müller Cells of the Diabetic Neural Retina by Modulating ACSL4.

Diabetic retinopathy (DR), a leading cause of vision impairment worldwide, is characterized by early neuronal damage in the retina, termed diabetic neuropathy in the retina (DNR). This condition is marked by neuronal apoptosis and glial activation. Müller glia are retinal cells highly susceptible to diabetic metabolic stress that may undergo ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation. However, the role of ferroptosis in DNR pathogenesis remains undefined. In this study, we investigated Müller cell injury under high-glucose and palmitic acid (HGP) c
