Optimizing IVF lab workflows through data-driven insights: associations between lab management, procedural timings, and workload with blastulation rates.

STUDY QUESTION: Do IVF laboratory workflows influence the mean blastulation rate per cohort of inseminated metaphase II oocytes (m-BR)? SUMMARY ANSWER: Neither the total number of procedures nor the workload per operator affected m-BR; instead, each additional hour in the interval from ovulation trigger to oocyte denudation (range 36-44 h) was associated with a measurable decline, especially beyond the 40-h threshold. WHAT IS KNOWN ALREADY: Control of laboratory conditions and standardized protocols are essential for optimizing m-BR in IVF. While advancements in technology and culture system

Generation Victoria (GenV): protocol for a longitudinal birth cohort of Victorian children and their parents.

BACKGROUND: In a world confronted with new and connected challenges, novel strategies are needed to help children and adults achieve their full potential, to predict, prevent and treat disease, and to achieve equity in services and outcomes. Australia's Generation Victoria (GenV) cohorts are designed for multi-pronged discovery (what could improve outcomes?) and intervention research (what actually works, how much and for whom?). Here, we describe the key features of its protocol. METHODS: GenV is a whole-population longitudinal platform open to all ≈150,000 children born within a two-year wi

Hypoplastic Left Heart Syndrome Practice Variation Across 31 Centres From 20 European Countries. An AEPC Imaging Working Group Study.

UNLABELLED: Despite significant advances in knowledge and the development of guidelines, the management of hypoplastic left heart syndrome (HLHS) remains highly variable. A structured questionnaire was circulated across European Association of Paediatric & Congenital Cardiology (AEPC) affiliated centres. The aims were to evaluate standards in pre-operative assessment, types of surgery, follow-up and medical practices in children with HLHS. Thirty-one centres from 20 countries completed the survey. Delivery of babies with HLHS occurred in co-located maternity hospitals in 74% of centres; 29% we

Dynamic of swine influenza virus infection in weaned piglets in five enzootically infected herds in Germany, a cohort study.

BACKGROUND: Within the last decades industrial swine herds in Europe grown significantly, creating an optimized reservoir for swine influenza A viruses (swIAV) to become enzootic, particularly in piglet producing herds among newborn, partly immunologically naïve piglets. To date, the only specific control measure to protect piglets from swIAV is the vaccination of sows, which provides passive immunity through maternally derived antibodies in colostrum of vaccinated sows. Interruption of infection chains through management practices have had limited success. This study focused on weaned piglets

Predicting of early miscarriage based on machine learning: a retrospective cohort study in China.

BACKGROUND: Early miscarriage (EM) remains a significant reproductive health challenge, with current serum hormone biomarkers (β-hCG, progesterone, estradiol) showing inconsistent predictive value across studies. To address these limitations, artificial intelligence (AI) and machine learning (ML) techniques are increasingly applied to integrate complex biomarker data and improve the early prediction of EM. METHODS: This retrospective cohort study analyzed 17,926 singleton pregnancies (1,918 EM vs. 16,008 live births) from Chongqing (2017-2023), China, to establish optimal hormonal predictors

Identification of Nuclear Genetic Loci Linked to Clinical Features of the m.3243A>G Mitochondrial DNA Variant.

BACKGROUND AND OBJECTIVES: Mitochondrial DNA (mtDNA) disorders exhibit striking clinical variability that is poorly explained by known factors such as variant heteroplasmy, age, or sex. Nuclear genetic modifiers likely play a significant role in this heterogeneity. We aimed to characterize the nature of nuclear genetic involvement for 2 common syndromic presentations of the common pathogenic mtDNA variant, m.3243A>G: mitochondrial encephalomyopathy, lactic acidosis, and stroke-like episodes (MELAS) and maternally inherited diabetes and deafness (MIDD). METHODS: We assembled a multicenter coho

Case Report: Sedation crisis caused by drug-induced sleep endoscopy in a pediatric patient with symptomatic multilevel airway obstruction.

BACKGROUND: Unexplained life-threatening events during pediatric sedation examinations are a major clinical challenge. Drug-induced sleep endoscopy (DISE) enables dynamic assessment of the site, severity, and pattern of airway collapse by simulating sleep through pharmacologically induced sedation. However, limited studies have utilized DISE as a diagnostic approach for elucidating the etiology of sedation-related events. CASE PRESENTATION: A male patient aged 10 years was admitted with a cough for >30 days and a history of cardiopulmonary resuscitation (CPR) 2 days earlier. The patient was d

The value of red blood cell distribution width in perioperative cardiac function assessment in children with congenital heart disease: A retrospective cohort study.

Congenital heart disease (CHD) is the leading cause of birth defects in children, with high morbidity and mortality rates. Despite advances in surgical and interventional treatments, perioperative management of CHD remains challenging, particularly in complex cases and neonates. Effective perioperative assessment of cardiac function is essential for optimizing surgical outcomes and improving postoperative prognosis. This study investigates the association between red blood cell distribution width (RDW) and perioperative cardiac function in children with CHD. Specifically, we aim to determine w

To ignore or not to ignore placental calcifications on prenatal ultrasound: a systematic review and meta-analysis.

OBJECTIVE: The human placenta is known to calcify with advancing gestational age, and, in fact, the presence of significant calcifications is one of the components of grade III placenta, typical of late gestation. As such, the presence of significant placental calcifications often prompts obstetric providers to expedite delivery. This practice has been attributed, in part, to the presumed association between grade III placenta and adverse pregnancy outcomes. Such approach, however, can be the source of major anxiety and may lead to unnecessary induction of labor, with its associated predisposi

Gastroschisis and omphalocele: treatments and long-term outcomes.

Between February 1994 and April 2004, we treated 40 children with gastroschisis and 26 children with omphalocele. We recorded the course of pregnancy, pre- and post-natal complications, delivery, operation, post-operative therapy, and long-term outcomes. Additionally, we conducted follow-up examinations of 37 of these 66 children (56%). We analysed their abdominal musculature, development, cosmetic result and quality of life. The median duration of follow-up was 6.3 years (range 1-10). In 35/40 children (88%) with gastroschisis and in 18/26 children (69%) with omphalocele, there had been prena
