Exposure to alcohol during gestation can have profound consequences, but not all cells within the embryo are affected equally. Recent advances in molecular embryology have allowed an exploration of this variation. Much of this research has focused on the embryo's vulnerability to the facial malformations characteristic of fetal alcohol syndrome. Studies using mice and chicks show that alcohol exposure at specific stages of early embryo development results in significant death among the cells destined to give rise to facial structures (i.e., cranial neural crest cells). This type of cell death

Controversy exists whether carbohydrate-deficient transferrin (CDT) is valuable as a screening tool for fetal alcohol syndrome. We evaluated serum CDT in 60 non-alcohol-abusing women at different stages of normal pregnancy. CDT was weakly related to week of pregnancy and to human placental lactogen. CDT did not correlate with iron oestradiol or progesterone. By contrast, good correlations were found between transferrin and week of pregnancy or either sex hormone. Using multiple linear regression analysis, only transferrin and week of pregnancy were important predictors of CDT. The diagnostic a

Magnetic resonance imaging was conducted on six children and adolescents with fetal alcohol syndrome and seven matched normal controls. Detailed volumetric analyses demonstrated significant reductions in the cerebral vault, basal ganglia, and diencephalon in the children with fetal alcohol syndrome, compared with control children. In addition, the volume of the cerebellar vault was smaller than controls in 4 of the 6 children with fetal alcohol syndrome, although the group difference did not reach significance. When the basal ganglia were divided into the caudate and lenticular nuclei, both of

Prenatal ethanol exposure has been associated with alterations in a variety of sexually dimorphic behaviors in rats. This study examined the effects of neonatal ethanol exposure on saccharin consumption, a sexually dimorphic behavior in rats. Subjects were Sprague-Dawley rats that were artificially reared (AR) from postnatal day (PN) 4-PN12 through gastrostomy tubes with ethanol exposure limited to PN4-PN10. The AR groups included two ethanol doses (6 g/kg/day and 4 g/kg/day) and an isocaloric maltose-dextrin control. A sham surgery control group was also included. The AR subjects were returne

Capture-recapture methods were used to estimate the prevalence of fetal alcohol syndrome among Alaska Natives born during the period 1982-1989. Potential cases were identified through an Indian Health Service (IHS) patient case file, a pediatric practice case file, and Medicaid claims from private physicians. A total of 74 Alaska Native children aged 3-10 years were identified with a notation of fetal alcohol syndrome by a physician in a medical record. Because not all of these cases had supporting documentation regarding the syndrome, they were classified as possible cases. Of these possible

Alcohol has been established as a teratogenic agent that in prenatal exposure can cause a variety of adverse pregnancy outcomes depending on dose, timing, and conditions of exposure. Although alcohol, like many other teratogenes, can cause alterations in morphology, growth, and neurobehavioral outcomes, this report focuses primarily on the latter effects. As with other teratogens causing neurobehavioral aberrations, the neurobehavioral effects of prenatal alcohol are produced at lower exposure levels than the morphologic or growth effects, and except in extreme cases, they are more devastating

Previous studies in our laboratory have shown that the ontogenic development of gamma-glutamyltranspeptidase (gamma-GTP) activity is delayed by the in utero administration of alcohol. gamma-GTP is responsible for the degradation and recycling of glutathione (GSH) via the gamma-glutamyl cycle. In this study, we examined the effects of the in utero administration of alcohol on GSH levels in gestational age 21-day-old (g21) rats. Pregnant rats were placed on a liquid diet containing either 35% ethanol-derived calories (35% EDC) or a pair-fed (PF) diet or a lab chow (LC) diet starting on day 1 of

The effect of prenatal exposure to alcohol on the development of basal heart rate and on the elicitation and habituation of the heart rate orienting response was examined in three experiments with rats. In all experiments, Etoh dams consumed large, daily amounts of alcohol and their weight gain during pregnancy was less than that of ad lib or pairfed dams. In addition, Etoh-exposed pups weighed less and grew more slowly than their ad lib or pairfed counterparts. Although prenatal exposure to alcohol had a significant effect on the ontogeny of basal heart rate, there was no effect on the magnit

Children with fetal alcohol syndrome (FAS) display great variability in their IQs during follow-up. The course of cognitive performance among twelve school girls aged 10 to 15 years with FAS who lived in foster homes of a chilean public organization for socially handicapped infants, was assessed by comparison of the results of Wechsler's psychometric test for children (WISC) which were done once and repeated 4 years later in the same patients. IQs were 7 to 13 points lower after 4 years in 7/12 girls, slightly better (4 and 6 points) in 2/12 and practically the same in 3/12 patients 1 to 3 poi

A case is described having malformation of the anterior chamber with bilateral high IOP combined with multiple ocular anomalies in a 13-year-old girl. Here, embryotoxon posterius, synechiae out to Schwalbe's line, and hypoplasia of the iris stroma in the form of Rieger's anomaly are associated with: myelinated corneal nerves concurrent with appearance of vessels without scleralization, unilateral orbital hypoplasia without microphthalmia and without enophthalmos within the scope of a unilateral facial hypoplasia, bilateral epicanthus with asymmetry of the lid-openings without hypertelorism, bi

Attitudes and knowledge about Fetal Alcohol Syndrome (FAS) were examined among American Indian communities of Michigan's Upper Peninsula. Indian health workers and community women were interviewed. Education about FAS was provided in each community. The results indicate that information on FAS is reaching many women in these communities and that traditional cultural patterns can support the development of a strong Indian women's health program. At the same time, more must be done in the near term to help those women who are at greatest risk.

Behavior of mature rats, who had been exposed antenatally to intrauterine alcoholic intoxication, was studied in the open field settings, unavoidable swimming and Simonov's emotional resonance situations; their conditioned reflexes of passive and active avoidance were trained, arterial blood pressure was measured, and sleeping patterns examined. It was demonstrated that animals' orientation/exploration activity was reduced, while passive behavior and fear-associated emotions became more prominent, conditioned reflexes were more difficult to form, vegetative autoregulation was disturbed, and in

Lateral cephalometric radiographs were quantitatively assessed in a series of 15 American black children with fetal alcohol syndrome (FAS). Although none was profoundly affected, FAS had been diagnosed in all the children at birth. Comparisons with age-, sex-, and race-matched controls disclosed a triad of facial profile differences: (1) frontal bossing, (2) palatal plane tipped up in the front with proclined upper incisors and a sharp nasolabial angle acquired from digit habits, and (3) above-average length of the mandibular corpus. Collectively these generate the perception of midface hypopl

Pregnant CD-1 mice were intubated from Days 5-17 of gestation with an isocaloric solution of ethanol and/or maltose dextrins in 10% Bio-Serv liquid diet, at doses of 0, 6, and 9 g/kg/day. The control females were food-restricted so that their food consumption matched that of the high-dose females. At delivery, the pups were assigned surrogate mothers and litters were culled to a uniform litter size, when possible. Immediately following birth some pups were examined using a battery of behavioral/developmental tests to monitor postnatal development. At 6-8 weeks of age, other pups were evaluated

Pregnant C57BL/6cr mice were fed a liquid diet containing 20% of the total calories from either ethanol (E) or sucrose (S) for gestation days 5-17. Adult male and female (six of each from both prenatal-treatment groups) offspring were tested under various schedules of food reinforcement. The first phase was a test of fixed-ratio (FR) acquisition in which the required number of responses per unit of reinforcement was increased from 1 to 20 to 100. Prenatal ethanol exposure interacted with other factors to produce an acquisition deficit. The second phase involved responding under extinction (Ext

Nineteen 5 to 12-weeks-old embryos from women abusing alcohol during pregnancy (alcoholism, habitual drinking) were studied. In 15 (79%) the brain development disorders were detected. Most frequently concerned were the brain hemispheres. Defective development signs were also noted in the midbrain, choroidal plexus and diencephalon. Microdysplasias were found in different brain regions. In one case embryonal signs of the capsula interna, nucleus caudatus, putamen, epiphysis cerebri, habenular nuclei, chiasma, optical tracts were absent. Six groups presenting most typical deviations were identif

Addition of ethanol (ET) to the drinking fluid of pregnant rats has been questioned as an experimental model for the fetal alcohol syndrome (FAS). This model, however, closely simulates human alcohol intake, and in this study we used a modified version of previous protocols to overcome their major defects. A group of female rats was given 10% ET in drinking fluid for one week, 15% for the second week, 20% for the third, and 25% for the fourth, at the end of which they were mated with non-treated males and given 25% ET throughout gestation. Three groups of non-ET treated sex and age-matched rat

Pregnant Wistar rats were exposed to either a liquid diet containing ethanol, pair-fed an identical diet with sucrose substituted for ethanol, or received ad lib chow and water, during days 6-19 of gestation. The activity of their offspring was tested at 10, 16, 22 or 28 days of age. Pups exposed to alcohol prenatally were more active than controls at 16 and 22 days of age. Thirty min after being placed in the activity chamber, the pups were injected with either 0, 0.5, 1.0 or 4.0 mg/kg of d-amphetamine sulphate, and returned to the chamber for a further 120 min. Amphetamine brought about a do

The offspring of rats fed a diet containing 12.5% (v/v) ethanol prior to, and during, gestation (LTE) were compared to the offspring of rats fed the same diet during gestation only (STE) in three studies. In Study I the neonates were tested on a variety of neuroreflex tasks, in Study II the offspring were tested in an activity wheel and open field, and in Study III the offspring were trained in a runway under partial or consistent reward conditions. Results indicated that STE offspring were delayed in developing fur and in opening their eyes, they righted more rapidly, and were slower in devel
