Postmenopausal hyperandrogenism is a relatively rare diagnosis resulting from excess androgen production from the adrenals or ovaries. The exclusion of malignant causes is a priority. Insulin receptor substrate (IRS) proteins are key mediators of insulin and insulin-like growth factor (IGF) signalling. In mice, deletion of Irs1 is associated with profound growth retardation and increased longevity whereas Irs2-deficiency causes diabetes and female infertility. In the present study, endocrine-pharmacological profiles of new aldosterone antagonists, 15 beta,16 beta-methylenemexrenone (ZK 91587), delta 1-15 beta,16 beta-methylene-spironolactone (mespirenone, ZK 94679) and 7 alpha-thiomethyl analogue of mespirenone (ZK 97894) were characterized in comparing them with those of spironolactone.
