The effect of pH on growth, protein synthesis, and lipid-rich particles of cultured mammalian cells.

A simple procedure has been established for controlling and measuring the pH of media in which the bicarbonate-carbonic acid system is the predominant buffer. The HCO(-) (3) concentration was maintained at 22.5 mM and the H(2)CO(3) concentration was varied by equilibrating the media with 0.5 to 40 per cent CO(2) in air. The curve relating extracellular pH to 3 day cell growth was similar for glass-attached HeLa and Chang liver cells. Maximum growth occurred over a pH range of 7.38 to 7.87. Cell growth declined precipitously on the alkaline side and more gradually on the acid side of the optima

P. aeruginosa Infected 3D Co-Culture of Bronchial Epithelial Cells and Macrophages at Air-Liquid Interface for Preclinical Evaluation of Anti-Infectives.

fDrug research for the treatment of lung infections is progressing towards predictive in vitro models of high complexity. The multifaceted presence of bacteria in lung models can re-adapt epithelial arrangement, while immune cells coordinate an inflammatory response against the bacteria in the microenvironment. While in vivo models have been the choice for testing new anti-infectives in the context of cystic fibrosis, they still do not accurately mimic the in vivo conditions of such diseases in humans and the treatment outcomes. Complex in vitro models of the infected airways based on human ce

The susceptibility of Aire(-/-) mice to experimental myasthenia gravis involves alterations in regulatory T cells.

The autoimmune regulator (Aire) is involved in the prevention of autoimmunity by promoting thymic expression of tissue restricted antigens which leads to elimination of self-reactive T cells. We found that Aire knockout (KO) mice as well as mouse strains that are susceptible to experimental autoimmune myasthenia gravis (EAMG) have lower thymic expression of acetylcholine receptor (AChR- the main autoantigen in MG), compared to wild type (WT) mice and EAMG-resistant mouse strains, respectively. We demonstrated that Aire KO mice have a significant and reproducible lower frequency of CD4+Foxp3+ c

Determinants of the acute-phase protein C-reactive protein in myocardial infarction survivors: the role of comorbidities and environmental factors.

BACKGROUND: C-reactive protein (CRP), a sensitive marker of the acute-phase response, has been associated with future cardiovascular endpoints independently of other risk factors. A joint analysis of the role of risk factors in predicting mean concentrations and variation of high-sensitivity CRP (hsCRP) in serum has not been carried out previously. METHODS: We used data from 1003 myocardial infarction (MI) survivors who had hsCRP measured monthly up to 8 times and multivariate mixed effects statistical models to study the role of time-variant and -invariant factors on the geometric mean of an

Differential expression of tissue-restricted antigens among mTEC is associated with distinct autoreactive T cell fates.

Medullary thymic epithelial cells (mTEC) contribute to the development of T cell tolerance by expressing and presenting tissue-restricted antigens (TRA), so that developing T cells can assess the self-reactivity of their antigen receptors prior to leaving the thymus. mTEC are a heterogeneous population of cells that differentially express TRA. Whether mTEC subsets induce distinct autoreactive T cell fates remains unclear. Here, we establish bacterial artificial chromosome (BAC)-transgenic mouse lines with biased mTEClo or mTEChi expression of model antigens. The transgenic lines support negati

Candidalysin activates innate epithelial immune responses via epidermal growth factor receptor.

Candida albicans is a fungal pathobiont, able to cause epithelial cell damage and immune activation. These functions have been attributed to its secreted toxin, candidalysin, though the molecular mechanisms are poorly understood. Here, we identify epidermal growth factor receptor (EGFR) as a critical component of candidalysin-triggered immune responses. We find that both C. albicans and candidalysin activate human epithelial EGFR receptors and candidalysin-deficient fungal mutants poorly induce EGFR phosphorylation during murine oropharyngeal candidiasis. Furthermore, inhibition of EGFR impair

Occupational effect of sugarcane biomass burning on the conjunctival mucin profile of harvest workers and residents of an adjacent town - A Brazilian panel study.

Pre-harvest burning of sugarcane fields produces large amounts of air pollutants which are known to cause health problems, including ocular surface abnormalities. In this study, we evaluated the effect of biomass burning on mucus quality and mucin gene expression (MUC1, MUC5AC, MUC16) in the conjunctiva of sugarcane workers (SWs) and residents of an adjacent town (RTs). Impression cytology samples of the inferior tarsal and bulbar conjunctiva of 78 SWs and 32 RTs were collected before (T1) and immediately after (T2) a 6-month harvest period. The neutral, acid and total mucus content of goblet

The compatible solute ectoine protects against nanoparticle-induced neutrophilic lung inflammation.

RATIONALE: Inflammatory reactions of the airways induced by nanoparticles of occupational and environmental origin contribute to organ-specific and systemic human diseases. Because this kind of exposure in modern societies is often unavoidable, a strategy of molecular prevention on an individual level could help to prevent inflammation-derived secondary diseases. OBJECTIVES: To test whether the compatible solute ectoine [(S)-2-methyl-1,4,5,6-tetrahydropyrimidine-4-carboxylic acid], which is known to reduce cell stress effects on a molecular level, prevents nanoparticle-induced lung inflammati

Beyond FLAIR: PRISM (polarity-preserving real inversion solute mapping)-a new Frontier in real inversion recovery MRI for intracranial and sensory organ fluid assessment.

Assessing subtle compositional changes in cranial and sensory organ fluids-including cerebrospinal fluid (CSF), ocular humors, and inner ear lymph fluid-is vital for neuroimaging. Heavily T2-weighted Fluid-attenuated Inversion Recovery (FLAIR) is sensitive to T1 changes induced by solutes, but conventional magnitude reconstruction (e.g., HYDROPS subtraction) suffers from artifacts like signal cancellation, paradoxical gadolinium-based contrast agent (GBCA) effects, and motion misregistration. This review summarizes the technical characteristics and diverse applications of PRISM (Polarity-prese

Psoriatic arthritis digital phenotyping and inflammation drivers (PDPID) study: protocol for an international multicentre prospective cohort.

INTRODUCTION: Changes in psoriatic arthritis (PsA) disease activity, particularly flares, are unpredictable and can impact patients' quality of life. Digital biomarkers derived from data collected via smart devices provide an opportunity for unobtrusive continuous monitoring of symptoms reflecting disease activity. Besides, our understanding of the mechanisms and triggers behind flares is limited. The primary objectives of the Psoriatic Arthritis Digital Phenotyping and Inflammation Drivers (PDPID) study are: (1) to develop digital biomarker capable of detecting PsA flare using data collected

Residential green space, walkability, and cardiometabolic biomarkers in midlife women: a longitudinal cohort study.

Background. Cardiovascular disease remains the leading cause of death among U.S. women. Green spaces and neighborhood features may influence cardiometabolic health by modulating environmental stressors and physical activity. Few longitudinal studies have examined green space, walkability, and cardio-inflammatory biomarkers, particularly in midlife women during the menopausal transition-a period of elevated cardiovascular risk. Methods. We analyzed longitudinal data from the multi-ethnic, multi-site U.S. Study of Women's Health Across the Nation (1999-2017) to examine associations between resid

PP4 deficiency drives airway epithelial senescence via the PERK-eIF2α-ATF4-p21 axis in severe asthma.

BACKGROUND: Cellular senescence, defined by irreversible cell cycle arrest and the senescence-associated secretory phenotype (SASP), has emerged as a critical driver of airway inflammation and hyperresponsiveness in asthma. Despite well-established associations between senescence, aging, and chronic disease, its precise mechanistic role in asthma pathogenesis remains poorly understood. Protein phosphatase 4 (PP4), a serine/threonine phosphatase with broad physiological functions, is significantly downregulated in airway epithelial cells derived from patients with severe asthma, suggesting a po

Microbial exposure and diversity in Norwegian shrimp processing plants.

Seafood processing workers have a high prevalence of respiratory symptoms and occupational asthma, primarily attributed to allergenic protein exposure. However, exposure to airborne microorganisms from raw materials can also contribute to allergic sensitization and other respiratory ailments. This study aimed to assess microbial exposure in shrimp processing plants and identify susceptible work tasks. Full-shift personal air samples were collected from two Norwegian shrimp processing plants across five distinct work processes: thawing, truck driving, cooking-peeling (technician), packing, and

Negative air ions alleviate nicotine-induced renal damage of spontaneously hypertensive rats via inhibiting oxidative stress and TGF-β/Smad pathway.

BACKGROUND: Nicotine can lead to renal damage in hypertension patients. Negative air ions (NAIs) is a natural antioxidant. However, rare research focus on the effect of NAIs on nicotine aggravated oxidative damage and hypertensive kidney damage. METHODS: We used a spontaneously hypertensive rat (SHR) model to investigate the molecular mechanisms for nicotine-exacerbated renal damage and the intervention effect of NAIs. 8-week-old male rats were injected with nicotine or nicotine+ 6 h/d 4.5 × 104 to 5 × 104 NAIs/cm3 for 1 month or 3 months, and continuous observation of systolic blood pressure
