Effects of cromakalim on bradykinin-, histamine- and substance P-induced airway microvascular leakage in the guinea-pig.

The effects of cromakalim on the increase in microvascular permeability induced by histamine, substance P or bradykinin in guinea-pig airways were studied in vivo. Extravasation of i.v. injected Evans blue dye was used as an index of permeability. We also studied the effects of cromakalim on the contractile effect of substance P, histamine or bradykinin on the isolated guinea-pig main bronchus and on the contractile response of isolated guinea-pig main bronchi to electrical field stimulation. Cromakalim (30 to 300 micrograms.kg-1) did not inhibit the increase in microvascular permeability indu

Sodium cromoglycate: evidence of tachykinin antagonist activity in the human skin.

The mechanism of action of the antiasthmatic drug sodium cromoglycate (SCG) is unclear. One possibility is that SCG antagonizes the effects of the tachykinin substance P (SP), an agent known to cause airway edema. However, when SP is inhaled by humans, it has no demonstrable effect on airway function; therefore, the possibility that SCG prevents SP-induced changes in microvascular permeability was examined in human skin in vivo where potent edema-producing effects are seen. SCG (5-500 nmol) caused significant (P < 0.05) dose-dependent inhibition of SP-induced edema (wheal) formation when coadm

Effect of substance P administration on vascular permeability in the rat dental pulp and submandibular gland.

The effects of substance P (SP) administration on vascular permeability were studied in the dental pulp (DP) of upper and lower incisors and in the submandibular gland (SMG) of male rats. Vascular permeability was assessed by means of extravasation of Evans blue dye. SP was diluted in 0.5% bovine serum albumin (BSE) and infused into the left common carotid artery. Separate groups of animals receive chloropyramine, an H1 histamine receptor antagonist (10 mg kg-1 i.v.) or indomethacin, a prostaglandin synthesis inhibitor (4 mg kg-1 i.v.) prior to SP infusions. Infusion of SP for 5 min increased

Antihistamine activity of Bryophyllum calycinum.

1. The juice obtained by pressing the leaves of Bryophyllum calycinum (Crassulaceae) exhibited histamine-blocking activity. 2. The juice contains flavonoid compounds, carbohydrates and mineral salts. A flavonoid fraction (fraction B) obtained by partitioning the juice between n-butanol and water contained the substance responsible for the antihistamine activity. 3. When assayed on the isolated guinea pig ileum, 50 mg/ml juice and 0.15 mg/ml fraction B produced parallel and concentration-dependent rightward displacement of the concentration-response curve to histamine (EC50 (FL): 1.30 (0.26-5.2

Active immunization induces lung hyperresponsiveness in the guinea pig. Pharmacologic modulation and triggering role of the booster injection.

In order to investigate whether bronchopulmonary hyperresponsiveness represents a unique property of sensitized lungs, we examined the responses of lungs from either actively sensitized, passively sensitized, or nonsensitized (control) guinea pigs to in vitro bronchoconstriction (BC) and release of thromboxane (TX) B2, 6-keto-PGF1 alpha, and histamine induced by platelet-activating factor (PAF-acether) or leukotriene (LT) D4. Guinea pigs were actively sensitized with 10 micrograms of either ovalbumin or Dermatophagoides farinae extract in AI(OH)3 injected intraperitoneally twice at a 2-wk inte

Anti-inflammatory property of 401 (MCD-peptide), a peptide from the venom of the bee Apis mellifera (L.).

1 Peptide 401, a potent mast cell degranulating factor from bee venom, substantially inhibited the oedema provoked by subplantar injection of carrageenin or intra-articular injection of turpentine in the rat. The ED(50) of 401 was c. 0.1 mg/kg. The anti-inflammatory effect was assessed by measurement of the increased (125)I-albumin content of an injected site in comparison with an uninjected contralateral site.2 Peptide 401 also suppressed the increased vascular permeability due to intradermal injection of various smooth muscle spasmogens (histamine, bradykinin, 5-hydroxytryptamine (5-HT), and

Growth hormone releasing peptide-6 (GHRP-6) ameliorates acute lung injury and its subsequent evolvement to interstitial fibrosis.

Acute lung injury/acute respiratory distress syndrome is a complex, characterized by acute onset, alveolar damage, and progressive hypoxemia … educed neutrophilic alveolitis, attenuated lung compliance failure, contributed to improve alveolar-capillary permeability, and reduced interleukin-1 beta serum levels. The chronic scenario, GHRP-6 preserved lung parenchymal integrity accounted for meager collagen accumulation. This is the first assessment on the potential protective of GHRP-6 in model of lung damages. …

A novel simulation model to assess the impact of Aerobic-Anoxic biostimulation mediated BTEX migration in a coupled Vadose-Phreatic subsurface system.

Groundwater and soil pollution caused by BTEX (Benzene, toluene, ethylbenzene, and xylene) is a threat to subsurface environment and bioremediation offers a promising cleanup strategy. This study presents a novel simulation model that integrates hydrogeochemical and microbial dynamics to assess BTEX migration in a coupled vadose-phreatic (CVP) subsurface for in-situ biostimulation of contaminated aquifers. A 2D Biostimulation-driven Vadose-Phreatic Migration (BioVPM) model is developed. The model considers vertical flow in the vadose zone and 2D flow in phreatic zone, while neglecting capillar

Establishment of a chronic aspiration pneumonia mouse model using oropharyngeal aspiration of food suspension.

BACKGROUND: Aspiration pneumonia (AP) is a prevalent and life-threatening pulmonary disease resulting from repeated aspiration of exogenous materials such as food or gastric contents. However, most existing animal models simulate only acute injury and fail to reproduce the chronic pathological features observed in patients. This study aimed to establish a stable and clinically relevant chronic AP mouse model. METHODS: First, four inhalation techniques including unilateral vagotomy (UV), intranasal inoculation (IN), intratracheal instillation (IT), and oropharyngeal aspiration (OA) were evalua

Sleep loss impairs blood-brain barrier function: Cellular and molecular mechanisms.

Sleep is a physiological process that preserves the integrity of the neuro-immune-endocrine network to maintain homeostasis. Sleep regulates the production and secretion of hormones, neurotransmitters, cytokines and other inflammatory mediators, both at the central nervous system (CNS) and at the periphery. Sleep promotes the removal of potentially toxic metabolites out of the brain through specialized systems such as the glymphatic system, as well as the expression of specific transporters in the blood-brain barrier. The blood-brain barrier maintains CNS homeostasis by selectively transportin

Powering the Future Green Buildings: Multifunctional Ultraviolet-Shielding Transparent Wood.

Indoor UV damage is a serious problem that is often ignored. Common glasses cannot filter UV rays well and have fragility and environmental issues. UV-shielding transparent wood (TW) holds promise, yet striking the right balance between blocking UV rays and allowing sufficient visible-light transmission poses a challenge. The pronounced capillary force, fueled by persistent moisture and extractives in wood, alongside the existence of multiphase interfaces, collectively hinder the uniform penetration of polymers and the effective dispersion of nanomaterials within the wood skeleton. Here, we in

mTOR inhibition as a novel gene therapeutic strategy for diabetic retinopathy.

In addition to laser photocoagulation, therapeutic interventions for diabetic retinopathy (DR) have heretofore consisted of anti-VEGF drugs, which, besides drawbacks inherent to the treatments themselves, are limited in scope and may not fully address the condition's complex pathophysiology. This is because DR is a multifactorial condition, meaning a gene therapy focused on a target with broader effects, such as the mechanistic target of rapamycin (mTOR), may prove to be the solution in overcoming these concerns. Having previously demonstrated the potential of a mTOR-inhibiting shRNA packaged

The Glycocalyx Shedding Influences Hemodynamic and Metabolic Response to Fluid Load in Septic Shock.

OBJECTIVE: Sepsis-associated endothelial dysfunction and degradation result in release of inflammatory mediators, compromise endothelial permeability, and impair alveolar fluid clearance leading to pulmonary edema. Excessive fluid therapy in septic shock damage the endothelial glycocalyx which will increase capillary leakage. The aim of our study was to assess the relationship of endothelial glycocalyx shedding with hemodynamic and metabolic response to fluid load in patients with septic shock. METHODS: Eighteen adult patients were included in prospective observational study. To predict the r

Pro-Resolving Mediator Resolvin E1 Restores Alveolar Fluid Clearance in Acute Respiratory Distress Syndrome.

Acute respiratory distress syndrome (ARDS) is a life-threatening condition characterized by increased permeability of the alveolar-capillary barrier and impaired alveolar fluid clearance. Resolvin E1 (RvE1) is a specialized pro-resolving mediator derived endogenously from omega-3-polyunsaturated fatty acids. RvE1 (10 μg/kg i.v.) was injected to rats 6 h post-lipopolysaccharide (LPS) (14 mg/kg) induction. After another 3 h, alveolar fluid clearance was measured in live rats (n = 8-9). The primary Type II alveolar epithelial cell was isolated and treated by LPS (1 μg/mL) with or without RvE1 (25

Cerebrovascular Pathology in Hypertriglyceridemic APOB-100 Transgenic Mice.

Hypertriglyceridemia is not only a serious risk factor in the development of cardiovascular diseases, but it is linked to neurodegeneration, too. Previously, we generated transgenic mice overexpressing the human APOB-100 protein, a mouse model of human atherosclerosis. In this model we observed high plasma levels of triglycerides, oxidative stress, tau hyperphosphorylation, synaptic dysfunction, cognitive impairment, increased neural apoptosis and neurodegeneration. Neurovascular dysfunction is recognized as a key factor in the development of neurodegenerative diseases, but the cellular and mo
