Decision-Making Signatures of Methamphetamine and Alcohol Use Disorders.

BACKGROUND: Aberrant decision making is a hallmark of substance use disorders (SUDs), often impeding recovery. While uncertainty, which comprises risk and ambiguity, is central to real-world choices, its distinct effects in SUDs remain underexplored. In this study, we disentangle risk and ambiguity to identify context-specific impairments in methamphetamine use disorder (MUD) and alcohol use disorder (AUD). METHODS: We used a Choice under Risk and Ambiguity task to examine uncertainty decision making (UDM) in 101 individuals with MUD, 56 individuals with AUD, and their respective healthy cont

Genetically Informed Disassortative Brain Morphometric Similarities Revealing Suicide Risk in Bipolar Disorder.

BACKGROUND: Cortical structure alterations in bipolar disorder (BD) have consistently been reported in association with suicide with high heritability. Currently, the multifaceted genetic landscape responsible for replicable neuroanatomical alterations with suicidal effects is poorly explored but could help develop personalized risk assessments in clinics. METHODS: Anatomically informed suicidal effects quantified with morphometric similarity network (MSN) upon structural magnetic resonance imaging were evaluated in 2 independent BD cohorts that consisted of patients with suicide attempt (SA)

Abnormal Association Between Neural Activity and Genetic Expressions of Impulsivity in Attention-Deficit/Hyperactivity Disorder: An Adolescent Brain Cognitive Development Study.

BACKGROUND: Impulsivity in highly heritable attention-deficit/hyperactivity disorder (ADHD) has been studied using neural activity via functional magnetic resonance imaging (fMRI) or genetic data, but rarely with multivariate methods that link both. We investigated coupled neural activity and gene expression signatures, using parallel independent component analysis (pICA) and ABCD (Adolescent Brain Cognitive Development) Study data. METHODS: Children with ADHD (n = 394; 63% male) and healthy control children (n = 1000; 47% male) of European ancestry were included. The participants were random

Dynamic and Static Resting-State Functional Connectivity of Canonical Networks in Military and Civilian Populations With Posttraumatic Stress Disorder and/or Mild Traumatic Brain Injury.

BACKGROUND: Posttraumatic stress disorder (PTSD) and mild traumatic brain injury (mTBI) are associated with alterations in the functional connectome, specifically in canonical resting-state networks including the default mode network (DMN), central executive network (CEN), and salience network (SN). Comorbid PTSD+TBI is linked to worse functional outcomes, but little is known about effects on the functional connectome. METHODS: We investigated brain phenotypes from resting-state functional magnetic resonance imaging associated with PTSD (n = 326), mTBI (n = 448), and comorbid PTSD+mTBI (n = 2

Reduced Gray-White Matter Contrast in Chronic Posttraumatic Stress Disorder in World Trade Center Responders.

BACKGROUND: Following the collapse of the World Trade Center (WTC), many people experienced severe trauma and 23% of WTC responders developed posttraumatic stress disorder (PTSD). We hypothesized that gray-white matter contrast (GWC) would be different in participants with PTSD compared with demographically matched trauma-exposed control participants with no history of PTSD. METHODS: T1-weighted structural images for 99 WTC responders collected on a 3T Siemen's magnetic resonance imaging scanner were retrieved and segmented to measure global, regional, and voxelwise GWC. Group-level analyses

Disrupted Brain Connectivity in Newborns Following Antenatal Opioid Exposure.

BACKGROUND: The neural bases of adverse neurodevelopmental outcomes in antenatal opioid exposure are poorly understood. Very limited in vivo human newborn imaging studies have reported disrupted functional connectivity (FC) in limbic and reward-related brain regions, but these studies used small samples and lacked matched controls. Our objective was to compare brain FC in antenatal opioid-exposed and unexposed newborns to study the impact of opioid exposure on early brain development. METHODS: Resting-state functional magnetic resonance imaging (rs-fMRI) data were collected using 3T MRI at 4

Reward Sensitivity in Patients Receiving Opioid Agonist and Antagonist Treatment for Opioid Use Disorder: An Observational Study.

BACKGROUND: Disrupted reward processing is a core component in neurobiological theories of addictions, including opioid use disorder (OUD). While acute opioid agonist and antagonist administration can modulate reward behavior and experiences, it remains unclear how typical long-term OUD treatment with these medications impacts patients' sensitivity to substance-free rewards. Therefore, we conducted a cross-sectional study of reward sensitivity in opioid agonist- and antagonist-treated patients with OUD and healthy volunteers. METHODS: Ninety-six patients with OUD on extended-release naltrexon

Presence, Severity, and Functional Associations of Incomplete Hippocampal Inversion in 22q11.2 Deletion Syndrome.

BACKGROUND: The hippocampus is smaller and functionally disrupted in individuals with 22q11.2 deletion syndrome (22q11DS), but the cause remains unclear. During gestational weeks 20 to 30, an inversion in the dentate gyrus and cornu ammonis occurs. This process can go awry, resulting in incomplete hippocampal inversion (IHI). In the general population, IHI is more common in the left hemisphere than the right hemisphere; however, its prevalence, severity, and functional impact in 22q11DS remain unexplored. Investigating IHI in 22q11DS could uncover morphological hippocampal abnormalities linked

Polygenic Risks for Depression and Neural Responses to Reward and Punishment in Young Adults.

BACKGROUND: Extensive research has shown aberrant reward and punishment processing in people with depression. Genetic risks contribute to depression, but whether or how these risks of depression may affect behavioral and neural responses to reward and punishment remains unclear. METHODS: We curated the data of 879 young adults performing a gambling task during brain imaging from the Human Connectome Project. Depression severity was assessed with the Achenbach Adult Self Report. Polygenic risk scores (PRSs) for depression were computed for all participants. With published routines and at a cor

Larger Neural Responses to Reward in Gambling Disorder: Relationships With Depression and Gambling Severity.

BACKGROUND: For most people, gambling is a type of entertainment that engages pleasure, risk, and reward drives. However, some individuals develop gambling disorder (GD), a behavioral addiction that involves continued gambling despite negative consequences. Disturbances in reward neurocircuitry have been implicated in GD but are not well characterized, including how neural alterations relate to the clinical symptomatology of GD and commonly co-occurring presentations such as depression. METHODS: Electroencephalography (EEG) was recorded while participants with GD (n = 26) and healthy control

Craving Induction and Treatment Response in Transcranial Magnetic Stimulation for Tobacco Use Disorder.

BACKGROUND: Repetitive transcranial magnetic stimulation (rTMS) is an emerging treatment for several mental health conditions including substance use disorders. Symptom provocation before rTMS is hypothesized to activate disease-related neurocircuitry and enhance treatment effects. For addiction, craving induction is frequently used before treatment, but its clinical utility requires further exploration. To assess whether craving induction enhances treatment outcomes, we analyzed data from the pivotal multisite trial of deep TMS (DTMS) for tobacco use disorder (TUD). METHODS: A total of 262 p

Structural Covariance of Early Visual Cortex Is Negatively Associated With Posttraumatic Stress Disorder Symptoms: A Mega-Analysis From the ENIGMA PTSD Working Group.

BACKGROUND: Identifying robust neural signatures of posttraumatic stress disorder (PTSD) symptoms is important to facilitate precision psychiatry and help in understanding and treatment of the disorder. Emergent research suggests that the structural covariance of early visual regions is associated with later PTSD development. However, large-scale analyses are needed in heterogeneous samples of trauma-exposed and trauma-naïve individuals to determine whether such a neural signature is a robust marker of vulnerability. METHODS: We analyzed data from the ENIGMA (Enhancing Neuro Imaging Genetics

Functional Connectivity Gradients Reveal Altered Hierarchical Cortical Organization in Functional Neurological Disorder.

BACKGROUND: Neuroimaging studies of functional neurological disorder (FND), a core neuropsychiatric condition, often rely on discrete connections or parcellations that may obscure the brain's functional network architecture. In this study, we applied a gradient-based approach to examine macroscale cortical organization in FND. METHODS: We analyzed resting-state functional magnetic resonance imaging data from 64 patients with mixed FND (FND-mixed), 61 age- and sex-matched healthy control participants (HCs), and 62 psychiatric control participants (PCs) matched on age, sex, depression, anxiety,

Pretreatment Dorsal Anterior Cingulate Cortex and Dorsolateral Prefrontal Cortex Activation Moderates Outcomes of Exposure-Focused Cognitive Behavioral Therapy in Pediatric Anxiety.

BACKGROUND: Cognitive behavioral therapy (CBT) for pediatric anxiety targets heightened emotional sensitivity and impaired cognitive control over emotion, core neurocognitive features of anxiety. Exposure is considered the most active component of CBT, but its efficacy varies. Pretreatment function of substrates for emotion processing and cognitive control may predict response to exposure relative to other CBT subcomponents, such as relaxation. METHODS: Youth with clinical anxiety (N = 118, 7-17 years) completed a functional magnetic resonance imaging task probing emotion processing and cogni

Modulation of Cerebellar-Cortical Connectivity Induced by Modafinil and Its Relationship With Receptor and Transporter Expression.

BACKGROUND: Modafinil is primarily used to treat narcolepsy but is also used as an off-label cognitive enhancer. Functional magnetic resonance imaging studies indicate that modafinil modulates the connectivity of neocortical networks primarily involved in attention and executive functions. However, much less is known about the drug's effects on subcortical structures. Following preliminary findings, we evaluated modafinil's activity on the connectivity of distinct cerebellar regions with the neocortex. We assessed the spatial relationship of these effects with the expression of neurotransmitte

Hypoactivation of the Ventromedial Frontal Cortex in Major Depressive Disorder: A Magnetoencephalography Study of the Reward Positivity.

BACKGROUND: The reward positivity (RewP) is a sensitive and specific electrophysiological marker of reward receipt. These characteristics make it a compelling candidate marker of dysfunctional reward processing in major depressive disorder. We previously proposed that the RewP is a temporal nexus for multiple dimensions of reward value and that a diminished RewP in depression might only reflect a deficit in some of these features. Specifically, we predicted a diminished ventromedial contribution in depression in the context of maintained reward learning. METHODS: We collected magnetoencephalo

Cortical Hypoactivation of Frontal Areas Modulates Resting Electroencephalography Microstates in Children With Attention-Deficit/Hyperactivity Disorder.

BACKGROUND: In the current study, we examined electroencephalography (EEG) microstate alterations and their neural generators during resting state in children with attention-deficit/hyperactivity disorder (ADHD) to explore a potential state biomarker. METHODS: A total of 76 participants, 38 with combined-type ADHD and 38 neurotypical children, took part in the study. Five-minute resting (eyes-open) 128 channel EEG data were acquired, and 2 minutes of clean EEG data were analyzed for microstates, its sources, and connectivity in both groups. Between-groups comparisons were done for microstate

Effects of GABA, Sex, and Stress on Reward Learning in Current and Remitted Major Depression.

BACKGROUND: Neurocognitive factors including aberrant reward learning, blunted GABA (gamma-aminobutyric acid), and potentiated stress sensitivity have been linked to anhedonia, a hallmark depressive symptom, possibly in a sex-dependent manner. However, previous research has not investigated the putative associations among these factors or the extent to which they represent trait- or state-based vulnerabilities for depression. METHODS: Young adults with current major depressive disorder (MDD) (n = 44), remitted MDD (n = 42), and healthy control participants (HCs) (n = 44), stratified by sex as

Examining the Most Important Risk Factors for Predicting Youth Persistent and Distressing Psychotic-Like Experiences.

BACKGROUND: Persistence and distress distinguish more clinically significant psychotic-like experiences (PLEs) from those that are less likely to be associated with impairment and/or need for care. Identifying risk factors that identify clinically relevant PLEs early in development is important for improving our understanding of the etiopathogenesis of these experiences. Machine learning analyses were used to examine the most important baseline factors distinguishing persistent distressing PLEs. METHODS: Using Adolescent Brain Cognitive Development (ABCD) Study data on PLEs from 3 time points
