Niche-Mediated Neural Priming Enables Robust and Scalable Generation of Human Choroid Plexus Organoids.

The choroid plexus (ChP) secretes cerebrospinal fluid (CSF) and forms the blood-CSF barrier (BCSFB), playing indispensable roles in brain development and homeostasis. However, tractable human ChP models remain limited, with current organoid protocols largely overlooking the stem cell niche as a determinant of lineage commitment. Here, we report that a small number of ChP organoids sporadically emerge during skin organoid induction, and switching human embryonic stem cells (hESCs) culture from E8 to mTeSR1 medium-preconditioning cells toward a neural-primed state-substantially enhances ChP orga

Obesity and Its Role in Fetal Programming-A Narrative Review.

The prevalence of maternal obesity is rapidly increasing, which represents a major public health concern worldwide. Currently more than 50% of all adult women are overweight or obese, and this trend is reflected in women of child-bearing age. Maternal obesity is characterized by metabolic dysfunction and chronic inflammation, and is associated with health problems in both the mother and the offspring. Intrauterine programming occurs during embryonic and fetal development, a critical period not only for the formation of tissues and organs but also for the etiology of diseases later in life. The

Synaptic and Non-Synaptic Functions of PTPRD: A Receptor Tyrosine Phosphatase at the Crossroads of Neural Circuitry and Metabolism.

Protein-tyrosine phosphatase receptor-type D (PTPRD) is an adhesion-coupled phosphatase that translates extracellular binding codes into intracellular phosphotyrosine signaling from embryogenesis through adulthood. Alternative inclusion of the Ig-domain mini-exons meA and meB tailors the ectodomain surface, thereby dictating high-affinity engagement with IL1RAPL1, IL1RAP, Slitrks, LRFN4/5 (SALM3/5), neuroligin-3, and other postsynaptic partners. Intracellularly, the catalytically active D1 domain and scaffold-like D2 module, anchored to liprin-α, coordinate presynaptic vesicle release, postsyn

Transcriptomic profiling of miRNA-mRNA interactions in canine oocytes and cumulus cells during in vitro maturation: a sequencing analysis.

Oocyte maturation is a critical process for successful fertilization and early embryonic development. In this study, we investigated the molecular mechanisms underlying oocyte maturation in dogs by analyzing the transcriptomic profiles of miRNAs and mRNAs in canine cumulus cells (CCs) and oocytes during in vitro maturation (IVM). RNA sequencing identified 285 miRNAs expressed in oocytes and 310 in CCs, with 282 miRNAs shared between the two cell types, highlighting the role of intercellular communication in maintaining miRNA expression equilibrium. Differential expression analysis revealed 222

Intranasally administered whole virion inactivated vaccine against clade 2.3.4.4b H5N1 influenza virus with optimized antigen and increased cross-protection.

The global spread, frequent antigenic changes, and pandemic potential of clade 2.3.4.4b highly pathogenic avian influenza H5N1 underscore the urgent need for robust cross-protective vaccines. Here, we developed a clade 2.3.4.4b H5N1 whole inactivated virus (WIV) vaccine strain with improved structural stability, productivity, and safety. By analyzing the evolutionary trends of clade 2.3.4.4b H5N1 viruses, we identified a key mutation (R90K) that increases heat stability while preserving antigenicity. Additionally, the PB2 gene of PR8 was replaced with a prototypical avian PB2 gene to increase

From Petri Dish to Primitive Heart: How IVF Alters Early Cardiac Gene Networks and Epigenetic Landscapes.

Numerous infants have been conceived by in vitro fertilization (IVF) and other assisted reproductive technologies (ART). Increasing evidence indicates that these approaches induce minor alterations in molecules during the initial phases of embryogenesis. This narrative review examines the molecular pathophysiology of embryonic cardiogenesis in the context of assisted reproductive technology, emphasizing transcriptional and epigenetic regulation. Essential transcription factors for cardiac development, including NKX2-5, GATA4, TBX5, ISL1, MEF2C, and HAND1/2, play a crucial role in mesodermal sp

HIRA, NKX2-5, and GATA4 Alterations versus Cardiac Malformations Related to 22q11.2 Deletion Syndrome.

INTRODUCTION: Congenital heart disease (CHD) comprises a wide spectrum of structural defects. However, the etiology of a large proportion of CHDs remains undefined. Among the genetic causes, 22q11.2 deletion syndrome is the condition which most stands out. This association is related to many cardiac embryonic development genes being in the chromosome 22 region, as well as being a region with a high probability of errors in gene recombination, influencing normal levels of gene expression and affecting a gene's copy number. OBJECTIVE: This study aimed to compare molecular findings using multipl

Development and Optimization of an RNA-Isolating Protocol for Mammalian Spermatozoa.

The RNAs present in spermatozoa play a crucial role in reproduction and embryonic development. They represent a promising diagnostic tool for assessing male infertility. However, their extraction is challenging due to their low concentration and highly condensed chromatin structure, as well as the presence of numerous cellular contaminants. These challenges vary across species and require the development of an optimized and reliable isolation method to obtain high-quality RNAs, which is essential for further molecular analyses regarding the roles played by these RNAs. This study evaluated two

Vascular Patterning affects Intramembranous Ossification through HIF1α-Vegf Signaling.

Organs and tissues develop in close association with vasculature which transports blood and nutrients and helps to remove waste. The vasculature is composed primarily of endothelial cells, which provide structure, form barriers, and are a source of developmental signals. We recently found that the Mediator, a multiprotein complex, which regulates transcription, was essential for proper vascular development. Here, we investigated the specific role of the Mediator tail subunit Med23 in endothelial cells. Endothelial specific knockout of Med23 in mouse embryos using Tek-Cre results in vascular an

Organoids in biliary research: insights into developmental signaling and applications in disease modeling.

The embryonic development of the biliary system is orchestrated by complex signaling pathways, including Notch, TGF-β, Wnt, and GFs, which regulate biliary stem/progenitor cell fate, polarity, and ductal morphogenesis. These pathways not only govern physiological development but are also deeply implicated in pathological processes such as inflammation, fibrosis, and carcinogenesis. However, conventional two-dimensional culture systems and animal models fall short in replicating the spatial structure, developmental dynamics, and human-specific molecular context of the biliary tract. As a result

Etiologies of endometriosis and model systems: is there a risk of a tunnel vision?

Endometriosis is the growth of endometrial-like tissue at non-uterine locations, primarily within the peritoneal cavity. The disease can have diverse presentations with superficial lesions, deep invading lesions and ovarian cysts (endometrioma) as the main subtypes. Immune dysregulation, recurrent inflammatory processes and fibrosis are commonalities of all endometriosis forms. Most theories explaining the etiology of endometriosis take their origin in retrograde menstruation. However, other theories have been proposed, including metaplasia of mesothelial tissue, abnormal proliferation of Müll

Developmental wave of programmed ganglion cell death in human retinal organoids.

The delicate and complex structure of the neural retina that enables proper visual function is achieved during embryonic development through a precise balance of proliferation, differentiation, and cell death. Retinal ganglion cells (RGC), the only output neurons of the retina, show a steady increase in numbers during development except for two waves of cell death that are highly conserved in vertebrates. However, the mechanisms responsible for these phenomena and their conservation in the human retina are incompletely understood. In this work we took advantage of human induced pluripotent ste

Cbfβ: A key regulator in skeletal stem cell differentiation, bone development, and disease.

The skeletal system comprises closely related yet functionally distinct bone and cartilage tissues, regulated by a complex network of transcriptional factors and signaling molecules. Among these, core-binding factor subunit beta (Cbfβ) emerges as a critical co-transcriptional factor that stabilizes Runx proteins, playing indispensable roles in skeletal development and homeostasis. Emerging evidence from genetic mouse models has highlighted the essential role of Cbfβ in directing the lineage commitment of mesenchymal stem cells (MSCs) and their differentiation into osteoblasts and chondrocytes.

Fibulin Family Members: New Players in Liver Fibrosis and Potential Biomarkers in Chronic Liver Diseases.

Chronic liver diseases (CLD) arising from various etiologies including viral hepatitis, excessive alcohol intake, alcoholic associated liver disease (ALD) and metabolic dysfunction-associated steatotic liver disease (MASLD), are associated with persistent hepatic injury ultimately resulting in fibrosis. Liver fibrosis is characterized by the excessive accumulation of extracellular matrix (ECM). This pathological ECM deposition increases tissue stiffness, disrupts normal tissue architecture and impairs liver functions leading to cirrhosis which constitutes a major risk factor for the onset of h

A 3D-printed microdevice encapsulates vascularized islets composed of iPSC-derived β-like cells and microvascular fragments for type 1 diabetes treatment.

Transplantation of insulin-secreting cells provides a promising method for re-establishing the autonomous blood glucose control ability of type 1 diabetes (T1D) patients, but the low survival of the transplanted cells hinder the therapeutic efficacy. In this study, we 3D-printed an encapsulation system containing β-like cells and microvascular fragments (MVF), to create a retrivable microdevice with vascularized islets in vivo for T1D therapy. The functional β-like cells were differentiated from the urine epithelial cell-derived induced pluripotent stem cells (UiPSCs). Single-cell RNA sequenci

Placental Evolution: Innovating How to Feed Babies.

The evolution of the placenta was transformative. It changed how offspring are fed during gestation from depositing all the resources into an egg to continually supplying resources throughout gestation. Placental evolution is infinitely complex, with many moving parts, but at the core it is driven by a conflict over resources between the mother and the baby, which sets up a Red Queen race, fueling rapid diversification of morphological, cellular, and genetic forms. Placentas from even closely related species are highly divergent in form and function, and many cellular processes are distinct. I

[Endothelial-mesenchymal transition, ventricular remodeling after myocardial infarction, and research progress of Chinese medicine intervention].

Ventricular remodeling after myocardial infarction(VRAMI) is a pathological phenomenon triggered by the abrupt occlusion of coronary arteries, leading to myocardial ischemia and hypoxia. This intricate process encompasses alterations in the dimensions, composition, and elasticity of the ventricular tissue and reflects pathophysiological reactions and self-repair after cardiomyocytes are damaged. The characteristic pathological manifestation of VRAMI is the presence of myocardial fibrosis(MF), wherein fibrotic cardiac tissue undergoes a loss of contractile and relaxation capacity, ultimately cu
