Gastric intubation of 1.5 g ethanol/kg maternal body weight produced a significant teratogenic response in ferrets (Mustela putorius) treated once daily from gestational days 15 through 35. This response was evident by a rise in the frequency of fetuses with one or more malformations (21%), and by a rise in the frequency of litters with one or more malformed fetuses (50%). Palatoschisis and greater relative intermedial canthi distance observed in this study are diagnostic features of the fetal alcohol syndrome (FAS). Although mean (+/- SEM) peak blood alcohol levels of 207 +/- 9 mg/dl did not

To investigate the teratogenic effect of acute alcohol exposure, pregnant C57BL/6J mice were exposed to 25% ethanol (either two doses of 2.9g/kg or one dose 5.8g/kg) during the organogenic period either by intraperitoneal injections or by intubation. The incidence of malformations varied according to (1) the stage of embryonic development at the time of exposure, (2) the route of administration of the alcohol, and (3) the amount of alcohol given and the time period over which it was administered. Oral doses of alcohol were teratogenic although less so than the same dose given intraperitoneally

A survey was conducted of deaf children in the Glasgow area diagnosed between 1965 and 1979 to assess the relative incidences of the known aetiological factors. This took the form of a review of case records supplemented by the use of questionnaires. The results showed small variations from earlier published figures but were broadly similar. The effectiveness of rubella and anti-Rhesus immunization were also evaluated. It was found that more than half of all cases of profound deafness in childhood are acquired rather than inherited and are therefore potentially preventable.

Significant alcohol ingestion during pregnancy can cause a spectrum of malformation of various degrees of severity in offspring. The full expression of "fetal alcohol syndrome" includes reduced growth, facial anomalies, and mental retardation. Affected infants are usually of near-term gestation, but small in weight and length. They continue to exhibit decreased growth postnatally. Mental retardation appears to be related to the degree of dysmorphic severity of appearance. It is primarily caused by central nervous system pathology rather than social environment. The most prevalent ophthalmologi

A semipurified liquid diet was used to develop a chronic rat model of the fetal alcohol syndrome. Control female rats gained weight normally, reproduced normally, and gave birth to normal litters on this diet. Increased neonatal mortality, decreased neonatal weights, and altered sex ratios were observed in offspring of experimental alcoholic animals. Preliminary histological results include a mild delay in cell lamination patterns in the cerebral cortex at 4 days postnatal in alcoholic offspring as well as decreased formation of dendritic spines at 7 days postnatal.

BACKGROUND: fetal Alcohol Spectrum Disorders is a term used to describe a range of physical, cognitive and behavioural deficits in the offspring of women who drank alcohol during pregnancy. A growing body of evidence suggests alcohol consumption in the presence of poor maternal nutrition may increase the risk of harm to the developing fetus.
OBJECTIVE: to investigate relationships between maternal dietary patterns and alcohol consumption, and explore which factors influence women's decisions about what to eat and drink during pregnancy.
DESIGN: a mixed methods study comprising a questionnair…

BackgroundPrenatal alcohol exposure (PAE) is a major teratogen associated with fetal alcohol spectrum disorders, yet alcohol use during pregnancy remains common in Germany. Previous interventions focusing primarily on education have shown limited effectiveness. The Theoretical Domains Framework (TDF) offers a comprehensive approach to identify psychosocial determinants of behavior, but its application to alcohol use in pregnancy has been limited.ObjectivesTo identify cognitive, emotional, social, and contextual determinants of alcohol consumption during pregnancy using a TDF-based framework, i…

INTRODUCTION: The constellation of birth defects seen in fetuses exposed to alcohol in utero have been described as fetal alcohol spectrum disorders. Evidence suggests that health care providers' communication practices regarding prenatal alcohol use could have beneficial outcomes. There is a paucity of investigations, however, that have examined the health professionals' personal alcohol use and prenatal alcohol recommendations they provide.
METHODS: This study sought to examine and compare midwives' personal alcohol use and communication practices regarding prenatal alcohol consumption. Cer…

BACKGROUND: Prenatal alcohol exposure (PAE) occurs in ~10% of pregnancies in North America and can alter brain development. Prior research on brain morphology in children with PAE shows mixed findings for cortical thickness, and only a few studies have investigated more complex morphological features, showing decreased gyrification with PAE. Here, we use four anatomical measures to provide a comprehensive characterization of cortical morphology in youth with PAE compared to unexposed youth.
METHODS: T1-weighted Magnetic Resonance Imaging (MRI) was used to examine cortical morphology metrics i

BACKGROUND: An overwhelming body of evidence suggests neuroimmune dysfunction as a key underlying mechanism of fetal alcohol spectrum disorder (FASD)-associated adverse central nervous system (CNS) outcomes. While few studies have highlighted the lingering effects of prenatal alcohol exposure (PAE) on producing specific immune factors, others suggest a primed neuroimmune state in adulthood, in which a proinflammatory bias is unmasked following subsequent immune activation in later life. However, the PAE-induced neuroimmune landscape in adulthood remains poorly defined. We hypothesized that PAE

Teratology investigates the origins of congenital disabilities, often linked to environmental factors such as ethanol (EtOH) exposure. Ethanol at 150 μM has been associated with teratogenic effects, oxidative stress, immunological responses, and endocrine disruptions. Fetal alcohol spectrum disorder (FASD) arises from maternal alcohol consumption during pregnancy, leading to developmental delays and cognitive impairment. Due to their diverse therapeutic applications, amino thiazole derivatives are crucial in drug development. This study aimed to determine whether the 2-amino thiazole derivativ

INTRODUCTION: There is growing evidence suggesting that children with prenatal alcohol exposure (PAE) struggle with cognitively demanding tasks, such as learning, attention, and language. Complex structural network analyses can provide insight into the neurobiological underpinnings of these functions, as they may be sensitive for characterizing the effects of PAE on the brain. However, investigations on how PAE affects brain networks are limited. We aim to compare diffusion magnetic resonance imaging (MRI) tractography-based structural networks between children with low-to-moderate PAE in trim

BACKGROUND: Prenatal alcohol exposure during gastrulation (embryonic day [E] 7 in mice, ~3rd week of human pregnancy) impairs eye, facial, and cortical development, recapitulating birth defects characteristic of Fetal Alcohol Syndrome (FAS). However, it is not known whether the prevalence or severity of craniofacial features associated with FAS is affected by biological sex.
METHODS: The current study administered either alcohol (2.9 g/kg, two i.p. doses, 4 hr apart) or vehicle to pregnant C57BL/6J females on E7, prior to gonadal sex differentiation, and assessed fetal morphology at E17.

INTRODUCTION: Children with fetal alcohol spectrum disorders (FASD) exhibit behavioral and affective dysregulation, including hyperactivity and depression. The mechanisms are not known, but they could conceivably be due to postnatal social or environmental factors. However, we postulate that, more likely, the affective dysregulation is associated with the effects of EtOH exposure on the development of fetal serotonergic (5-HT) and/or dopaminergic (DA) pathways, i.e., pathways that in postnatal life are believed to regulate mood. Many women who use alcohol (ethanol, EtOH) during pregnancy suffe…

BACKGROUND: During early development, alcohol exposure causes apoptotic cell death in discrete regions of the embryo which are associated with distinctive patterns of later-life abnormalities. In gastrulation, which occurs during the third week of human pregnancy, alcohol targets the ectoderm, the precursor of the eyes, face, and brain. This midline tissue loss leads to the craniofacial dysmorphologies, such as microphthalmia and a smooth philtrum, which define fetal alcohol syndrome (FAS). An important regulator of alcohol-induced cell death is the pro-apoptotic protein Bax. The current study

BACKGROUND: Attention deficits caused by prenatal ethanol (EtOH) exposure (PE) are a prevalent condition in fetal alcohol spectrum disorders (FASDs). Importantly, the deficits are observed in individuals with FASD who have normal IQs and show no dysmorphic facial features caused by heavy PE. These observations suggest that even moderate PE could lead to attention deficits. This possibility was investigated in the present study using a rat model.
METHODS: Pregnant Sprague Dawley rats were administered EtOH (3 g/kg/day) or vehicle via intragastric gavage on gestational days 8 to 20. The blood E…

While it is known that dilation of cerebral arterioles to NOS-dependent agonists is impaired in rats exposed to prenatal alcohol, no studies have examined the influence of prenatal alcohol on constrictor response of cerebral arterioles. Our goal was to determine whether constrictor responses of cerebral resistance arterioles are altered by prenatal exposure to alcohol and if any changes differed as a function of age or sex. We fed Sprague-Dawley rat dams a liquid diet with or without alcohol (3% ethanol) for the duration of their pregnancy. We then examined reactivity of cerebral arterioles to

Children with prenatal alcohol exposure (PAE) often suffer from cognitive and neurobehavioral dysfunction throughout their lives, which may rise to a level of concern such that children receive a diagnosis under the fetal alcohol spectrum disorders (FASD) umbrella. Magnetoencephalography (MEG) contributes direct insight into neural processing and functional connectivity measures with temporal precision to understand cortical processing disorders that manifest during development. The impairment of perception may become more consequential among school-aged children with an FASD in the process of…
