INTRODUCTION: 2,4-dinitrophenol (DNP) is a mitochondrial toxin sometimes used as a weight loss agent. Reports of fatalities from DNP have been increasing since 2000, suggesting an increase in use. Our understanding of DNP toxicity in humans comes from reports to Poison Control and postmortem analyses, sources that are biased to more extreme presentations. This leads to a gap in our knowledge about the adverse effects of DNP at nonlethal doses. Here we investigate the doses and effects of DNP as reported online.
METHODS: We analyzed publicly available Internet posts that we collected from 2017…

Preventing the harm caused by nerve degeneration is a major challenge in neurodegenerative diseases and in various forms of trauma to the nervous system. The aim of the current work was to investigate the effects of systemic administration of 2,4-dinitrophenol (DNP), a compound with newly recognized neuroprotective properties, on sciatic-nerve degeneration following a crush injury. Sciatic-nerve injury was induced by unilateral application of an aneurysm clip. Four groups of mice were used: uninjured, injured treated with vehicle (PBS), injured treated with two intraperitoneal doses of DNP (0.…

OBJECTIVE: We evaluated the potential neuro-regenerative effects of the mitochondrial uncoupler 2,4-Dinitrophenol in experimental autoimmune neuritis, an animal model for an acute autoimmune neuropathy.
METHODS: Experimental autoimmune neuritis was induced in Lewis rats. Different concentrations of 2,4-Dinitrophenol (1 mg/kg, 0.1 mg/kg and 0.01 mg/kg) were applied during the recovery phase of the neuritis (at days 18, 22 and 26) and compared to the vehicle. Any effects were assessed through functional, electrophysiological, and morphological analysis via electron microscopy of all groups at d…

Transthyretin (TTR), a homotetrameric thyroxine transport protein found in the plasma and cerebrospinal fluid, circulates normally as a innocuous soluble protein. In some individuals, TTR polymerizes to form insoluble amyloid fibrils. TTR amyloid fibril formation and deposition have been associated with several diseases like familial amyloid polyneuropathy and senile systemic amyloidosis. Inhibition of the fibril formation is considered a potential strategy for the therapeutic intervention. The effect of small water-soluble, hydrophobic ligand 2,4-dinitrophenol (2,4-DNP) on TTR amyloid formati…

2,4-Dinitrophenol (DNP) is a potent mitochondrial uncoupler briefly marketed in the 1930s as a weight-reducing agent before being banned by the FDA after reports of severe toxicity. Since the early 2000s, DNP has reemerged as an illicit "fat-burner", causing characteristic metabolic disturbances with a high risk of fatal outcome. We describe two Swedish cases of DNP poisoning: one fatal after suicidal ingestion and one non-fatal after use for weight reduction. Clinical data, mitochondrial respirometry, and analysis of gas exchange and ventilatory dynamics were used to characterize the metaboli…

The morphofunctional state of the rat liver biomembranes is studied under conditions of the peroral effect of alkyl dinitrophenols: dinitroorthocresole and secondary 2,4-dinitro-6-butylphenol. It is established that free radicals and products of peroxide lipid oxidation (diene conjugates, hydroperoxides, final products reacting with 2-thiobarbituric acid) are very important for realization of the membrane-tropic action of the above compounds. Possible mechanisms of development of the membrane-damaging action of these xenobiotics and their effect on the state of the mitochondrial respiratory ch

Following anoxia, a rapid and marked mitochondrial-linked cell death occurs in the cerebral cortex of newborn rats which leads to insult advancement within a couple of days and causes lifelong neurobehavioral abnormalities. The present study investigated the role of 2,4 dinitrophenol (2,4 DNP) in three doses, i.e.,1, 2.5, and 5 mg/kg on anoxia-induced time-dependent mitochondrial dysfunction and associated neurobehavioral outcome using a well-established global model of anoxia. Briefly, rat pups of 30-h age (P2) were subjected to two episodes of anoxia (10 min each) at 24 h of the time interva

Catalase activity of intact E. coli bacteria had a broad pH-optimum (4.0-7.5). Addition of protonofore 2,4-dinitrophenol (200 microM) caused pH-dependency modification (6.5-7.0). Both the catalase activity and curves mode of native cells in the presence of dinitrophenol and cell-free extracts are almost identical. The loss of catalase activity at acid pH was caused by cells destruction and dinitrophenol addition, that makes it possible to suppose that this activity is connected with some membrane component functioning. The induction of "acid" catalase by oxidative stress was blocked with addit…

2,4-dinitrophenol, initially developed for industrial purposes, is used in the synthesis of dyes and various drugs, including herbicides, fungicides and insecticides. Incidentally, it was discovered that 2,4-dinitrophenol increases human basal metabolic rate. As a consequence, people who want to loose weight can abuse 2,4-dinitrophenol. It is a compound of choice for bodybuilders who abuse it in combination with anabolic steroids and clenbuterol. Numerous deaths have been attributed to 2,4-dinitrophenol. Up to now, 2,4-dinitrophenol was never reported in hair. The authors present an original m…

1. These studies examined the theory that ATP served to regulate muscle sugar transport by a feedback mechanism. Xylose uptake by isolated rat soleus muscle was determined over a 5-min period following preincubation at 37 degrees C for various times in the presence of insulin (0.1 unit/ml), 2,4-dinitrophenol (0.5 or 0.05 mM) or salicylate (5 mM) or under anaerobic conditions. 2. Xylose uptake, measured in freshly isolated soleus muscles, was approximately 3.5--4.0 mumol/g per h. When the muscles were preincubated at 37 degrees C, this rate fell by 50% during the first 30 min and then slowly in…

Studies on the mechanism of chemically induced intestinal epithelial injury were carried out using isolated, rat small intestinal epithelial cells. Compounds such as 2,4-dinitrophenol (DNP) and diethyl maleate (DEM), caused NADH loss, an increase in cytosolic Ca2+ concentration and protein thiol loss. Further, these compounds accelerated cell aggregation and decreased cell viability. Calmodulin antagonists inhibited protein thiol loss induced by either of the compound, inhibited cell aggregation and prolonged cell viability, but did not influence NADH loss. It has been reported that the calmod…

The effect of 2,4-dinitrophenol (2,4-DNP) on the oxidative metabolism of hexobarbital by cytochrome P-450 was investigated in perfused rat liver. In the livers from fed, phenobarbital (PB)-treated rats, 2,4-DNP (50 microM) had no effect on the redox state of cytochrome P-450 or on oxygen uptake during mixed-function oxidation of hexobarbital. In the livers from fasted, PB-treated rats, 2,4-DNP (50 microM) significantly decreased the amount of reduced (oxygenated) cytochrome P-450 and the drug-induced oxygen uptake by about 50%. 2,4-DNP caused a decrease of metabolites of hexobarbital in perfus…
