Bioorthogonal pro-metabolites for profiling short chain fatty acylation.

Short chain fatty acids (SCFAs) play a central role in health and disease. One function of these signaling molecules is to serve as precursors for short chain fatty acylation, a class of metabolically-derived posttranslational modifications (PTMs) that are established by lysine acetyltransferases (KATs) and lysine deacetylases (KDACs). Via this mechanism, short chain fatty acylation serves as an integrated reporter of metabolism as well as KAT and KDAC activity, and has the potential to illuminate the role of these processes in disease. However, few methods to study short chain fatty acylation…

Ring-opening metathesis polymerization-based recyclable magnetic acylation reagents.

An operationally simple method for the acylation of amines utilizing carbon-coated metal nanoparticles as recyclable supports is reported. Highly magnetic carbon-coated cobalt (Co/C) and iron (Fe/C) nanobeads were functionalized with a norbornene tag (Nb-tag) through a "click" reaction followed by surface activation employing Grubbs-II catalyst and subsequent grafting of acylated N-hydroxysuccinimide ROMPgels (ROMP=ring-opening metathesis polymerization). The high loading (up to 2.6 mmolg(-1) ) hybrid material was applied in the acylation of various primary and secondary amines. The products w…

Fatty acid acylation of dopamine in the carotid body.

In this article, we put forward a hypothesis concerning the assembling and storage of dopamine molecules in the dense-core vesicles of the carotid body chemoreceptor cell. We posit that dopamine molecules are packed and sustained in the vesicular form due to the formation of N-acyldopamine, a condensation product of fatty acid acyl chain and dopamine at the amino group of the latter. N-acyldopamine would then be stored in a micelle-like supramolecular structure formed due to self-association through the hydrophilic dopamine headgroups. This hypothesis may help explain the perennial problem of

Palladium-Catalyzed Intermolecular Acylative Heck Reactions with Imides as Acyl Electrophiles.

We disclose palladium-catalyzed, intermolecular, acylative Heck reactions that use imides as acyl electrophiles. The catalyst generated from [Pd(allyl)Cl]2 and DPEphos promotes the reaction between N-benzoylglutarimides and norbornene in the presence of silver phosphate. The acylative Heck reaction encompasses an array of N-benzoylglutarimide electrophiles that contain electron-donating, halogenated, and electron-withdrawing substituents to generate α,β-unsaturated ketones in moderate to high yields (25-82%). The bicylic α,β-unsaturated ketones are readily transformed into polycyclic architect

S-Acylation of Proteins of Coronavirus and Influenza Virus: Conservation of Acylation Sites in Animal Viruses and DHHC Acyltransferases in Their Animal Reservoirs.

Recent pandemics of zoonotic origin were caused by members of coronavirus (CoV) and influenza A (Flu A) viruses. Their glycoproteins (S in C … rate specificity is disturbed. We speculate that newly emerged CoVs or Flu viruses also depend on S-acylation for replication and will use the human DHHCs for that purpose. This feature makes these DHHCs attractive targets for pan-antiviral drugs.

Dynamic S-acylation of IDH2 regulates mitochondrial redox homeostasis.

Mitochondrial redox homeostasis is fundamental for cellular function, and its dysregulation is associated with various diseases, including cancer. Isocitrate dehydrogenase 2 (IDH2) is a key enzyme that maintains this balance by generating NADPH. However, the mechanisms controlling IDH2 subcellular localization remain incompletely understood. Here, we identify reversible S-acylation as a critical regulator of IDH2 localization. Using chemical reporters, we demonstrate that IDH2 is S-acylated at a conserved cysteine residue, mediated by ZDHHC3 and APT1. Loss of IDH2 S-acylation disrupts its mito…

Photoredox-catalyzed carbonylative acylation of styrenes with Hantzsch esters.

Ketones exist widely in naturally occurring products and are indispensable building blocks in organic synthesis. Carbonylation represents one of the most straightforward methods for ketone preparation and has become an attractive field in modern organic chemistry as well. Among the strategies, photocatalytic carbonylation is also worthy of further exploration. Herein, we developed a three-component carbonylation that provides a new method for the synthesis of ketones from Hantzsch esters, CO and styrenes. The reaction was performed under a blue light environment and yields a series of ketones

Photoinduced cerium-catalyzed C-H acylation of unactivated alkanes.

Ketones are ubiquitous motifs in the realm of pharmaceuticals and natural products. Traditional approaches to accessing these species involve the addition of metal reagents to carboxyl compounds under harsh conditions. Herein, we report a cerium-catalyzed acylation of unactivated C(sp3)-H bonds using bench-stable acyl azolium reagents under mild and operationally-friendly conditions. This reaction exhibits excellent generality, accommodating a wide range of feedstock chemicals such as cycloalkanes and acyclic compounds as well as facilitating the late-stage functionalization of pharmaceuticals…

Regioselective Acylation of Diols and Triols: The Cyanide Effect.

Central topics of carbohydrate chemistry embrace structural modifications of carbohydrates and oligosaccharide synthesis. Both require regioselectively protected building blocks that are mainly available via indirect multistep procedures. Hence, direct protection methods targeting a specific hydroxy group are demanded. Dual hydrogen bonding will eventually differentiate between differently positioned hydroxy groups. As cyanide is capable of various kinds of hydrogen bonding and as it is a quite strong sterically nondemanding base, regioselective O-acylations should be possible at low temperatu…

Acyl Histidines: New N-Acyl Amides from Legionella pneumophila.

Legionella pneumophila, the causative agent of Legionnaires' disease, is a Gram-negative gammaproteobacterial pathogen that infects and intracellularly replicates in human macrophages and a variety of protozoa. L. pneumophila encodes an orphan biosynthetic gene cluster (BGC) that contains isocyanide-associated biosynthetic genes and is upregulated during infection. Because isocyanide-functionalized metabolites are known to harbor invertebrate innate immunosuppressive activities in bacterial pathogen-insect interactions, we used pathway-targeted molecular networking and tetrazine-based chemosel

Rat intestinal acyl coenzyme A: cholesterol acyl transferase properties and location.

Microsomal acyl coenzyme A:cholesterol acyltransferase activity of rat intestinal mucosa was measured as the incorporation of [1-14C]oleic acid into cholesterol esters. Exogenous cholesterol and sterol carrier protein singly and together extended the time over which the reaction rate was linear. Cholesterol esterification was suppressed by 2-monooleoyl glyceryl ether a 2-monoglyceride analog and potential substrate, but was unaltered by alpha-glycerophosphate or lysolecithin. Esterifying activity was lowest in microsomes from the intestinal segment 0-15 cm distal to the pylorus and highest in

Preparative enzymatic synthesis and hydrophobic chromatography of acyl-acyl carrier protein.

We have used purified preparations of acyl-acyl carrier protein synthetase to prepare pure, native acyl-acyl carrier proteins (acyl-ACP) ranging in chain lengths from C10:0 to C delta 9 18:1. Factors affecting yield are explored and reaction conditions are presented that yield 0.8 to 0.9 mg of C16:0-ACP/ml of reaction mix. Ohter acyl groups, such as C10:0 and C delta 9 18:1 are poorer substrates and gave correspondingly lower yields. Acyl-Acp synthetase may be recovered from the reaction mixture using blue-Sepharose CL-6B and recycled. ACP and acyl-ACP are separated by hydrophobic chromatograp

Multiple acyl-CoA dehydrogenation deficiency as decreased acyl-carnitine profile in serum.

We report a case with late onset riboflavin-responsive multiple acyl-CoA dehydrogenation deficiency (MADD) characterized by decreased acyl-carnitine profile in serum which is consistent with primary systemic carnitine deficiency (CDSP) while just the contrary to a typical MADD. This patient complained with muscle weakness, muscle pain and intermittent vomiting, and was diagnosed as polymyositis, received prednisone therapy before consulted with us. Muscle biopsy revealed mild lipid storage. The findings of serum acyl-carnitines were consistent with CDSP manifesting as decreased free and total

HDAC11 Regulates RNA Splicing via De-Fatty Acylation of SF3B2.

Histone deacetylase 11 (HDAC11) is a lysine de-fatty acylase whose cellular substrates and mechanisms remain incompletely defined. Here, using metabolic labeling, mass spectrometry, click chemistry, and standard molecular biology, we show that SF3B2 is modified by lysine myristoylation at K10 and that HDAC11 efficiently removes this modification in cells, establishing SF3B2 as a direct enzymatic substrate. A de-myristoylation mimetic mutant (SF3B2 K10R) exhibits altered pre-mRNA binding activity in a context-dependent manner. In HCC cells, loss of SF3B2 lysine myristoylation enhances SF3B2 ass

Targeting Ferroptosis-associated Histone Acylation for Amelioration of Neurological Disease.

Ferroptosis is an iron-dependent, lipid peroxidation-driven form of programmed cell death. There is substantial evidence supporting the critical role of ferroptosis in multiple neurological diseases, including stroke, Alzheimer's disease, Parkinson's disease, epilepsy, and traumatic brain injury. Histone acylation, an important epigenetic mechanism, effectively regulates ferroptosis. To date, the regulation of ferroptosis by histone acylation in neurological diseases has rarely been summarized. Therefore, this review discusses the key mechanisms by which histone acylation regulates ferroptosis…

Photobiocatalytic enantioselective C(sp³)-H acylation using thiamine-dependent enzymes.

Thiamine diphosphate (ThDP)-dependent enzymes are a versatile class of enzymes traditionally applied in biocatalysis for carbon-carbon bond forming or cleaving reactions via two electron chemistry. Recent advances have revealed their potential to catalyze radical-mediated transformations when combined with chemical hydrogen atom transfer (HAT) strategies, enabling the enantioselective functionalization of C(sp³)-H bonds. This approach allows for the direct acylation of benzylic C(sp³)-H sites across a broad range of organic substrates, affording enantioenriched ketones with good to high levels…
