A2 amacrine cell output synapses to alpha ganglion cells in the mouse retina.

A2 amacrine cells (AIIACs) are the most important amacrine cells (ACs) in the mammalian retina, as they are the "hubs" for merging rod and cone signals in the inner retina and segregate them into ON and OFF ganglion cell (GC) pathways. It is unclear, however, whether AIIACs send their signals to all types of GCs in the mammalian retina. A major reason for this knowledge gap is that it has not been possible to selectively stimulate AIIACs without stimulating other retinal neurons. In this study, we will use a newly-developed mouse retinal preparation in which channelrhodopsin (a ChR2 [H134R]-Ve…

The mysterious middlemen making your vision pop: understanding the function of amacrine cells.

In many brain regions, inhibitory interneurons represent a highly diverse class of cells, and the specific roles of most subtypes remain unclear. This diversity is particularly striking in the retina, where amacrine cells, the primary inhibitory interneurons, form the most diverse population, with nearly 67 subtypes in mice. Recent methodological advances have provided unprecedented insight into this complexity. Techniques such as transcriptomics, connectomics and targeted electrophysiological recordings have made it possible to isolate and characterize individual amacrine cell types. Here, we…

A comprehensive study of c-Kit-expressing amacrine cells in the mammalian retina.

Amacrine cells assemble complex circuits in the inner retina, where they integrate, modulate, and relay spatial and temporal information to retinal ganglion cells. Nevertheless, their organization and functional diversity are still not fully understood. In this study, we identify a distinct group of amacrine cells that express the stem cell factor receptor-c-Kit. Using combined techniques of anti-CD117 (c-Kit) immunolabeling, confocal imaging, and computational algorithms, we characterize the morphological features and spatial distribution of the c-Kit-expressing amacrine cells. Our results sh…

Dopaminergic amacrine cells express HCN channels in the developing and adult mouse retina.

PURPOSE: To determine the molecular and functional expression of hyperpolarization-activated cyclic nucleotide-gated (HCN) channels in developing and mature dopaminergic amacrine cells (DACs), the sole source of ocular dopamine that plays a vital role in visual function and eye development. METHODS: HCN channels are encoded by isoforms 1-4. HCN1, HCN2, and HCN4 were immunostained in retinal slices obtained from mice at postnatal day 4 (P4), P8, and P12 as well as in adults. Each HCN channel isoform was also immunostained with tyrosine hydroxylase, a marker for DACs, at P12 and adult retinas. …

Cell firing between ON alpha retinal ganglion cells and coupled amacrine cells in the mouse retina.

Gap junctions are channels that allow for direct transmission of electrical signals between cells. However, the ability of one cell to be impacted or controlled by other cells through gap junctions remains unclear. In this study, heterocellular coupling between ON α retinal ganglion cells (α-RGCs) and displaced amacrine cells (ACs) in the mouse retina was used as a model. The impact of the extent of coupling of interconnected ACs on the synchronized firing between coupled ON α-RGC-AC pair was investigated using the dopamine 1 receptor (D1R) antagonist-SCH23390 and agonist-SKF38393. It was obse…

Asymmetric connections with starburst amacrine cells underlie the upward motion selectivity of J-type retinal ganglion cells.

Motion is an important aspect of visual information. The directions of visual motion are encoded in the retina by direction-selective ganglion cells (DSGCs). ON-OFF DSGCs and ON DSGCs co-stratify with starburst amacrine cells (SACs) in the inner plexiform layer and depend on SACs for their direction selectivity. J-type retinal ganglion cells (J-RGCs), a type of OFF DSGCs in the mouse retina, on the other hand, do not co-stratify with SACs, and how direction selectivity in J-RGCs emerges has not been understood. Here, we report that both the excitatory and inhibitory synaptic inputs to J-RGCs a…

Specific amacrine cell changes in an induced mouse model of glaucoma.

BACKGROUND: To investigate retinal cell population changes under chronic elevated intraocular pressure in an inducible mouse model of glauco … perimental elevation of intraocular pressure alters calcium-binding protein immunohistochemistry in amacrine cells. However, these changes are not indicative of amacrine cell loss but may represent early indicators of cellular distress that precede physiological dysfunction or cell death.

GABAergic TH2 Amacrine Cells Participate in Spontaneous Wave Activity in the Developing Retina.

UNLABELLED: Amacrine cells (ACs) are retinal interneurons that regulate synaptic transmission from bipolar cells to retinal ganglion cells (RGCs) and play essential roles in object motion detection, contrast sensitivity, and light adaptation. A subtype of GABAergic ACs identified using a tyrosine hydroxylase (TH) promoter-driven green fluorescent protein (GFP) mouse line has been termed TH2 amacrine cells (TH2-ACs). Although TH2-ACs contribute to the feature selectivity of object-motion signals in the adult retina, their functional properties during early postnatal development remain unclear. …

Postsynaptic Plasticity Triggered by Ca²⁺-Permeable AMPA Receptor Activation in Retinal Amacrine Cells.

Amacrine cells are thought to be a major locus for mechanisms of light adaptation and contrast enhancement in the retina. However, the potential for plasticity in their AMPA receptor currents remains largely unknown. Using paired patch-clamp recordings between bipolar cell terminals and amacrine cells, we have simultaneously measured presynaptic membrane capacitance changes and EPSCs. Repetitive bipolar cell depolarizations, designed to maintain the same amount of exocytosis, nevertheless significantly potentiated evoked EPSCs in a subpopulation of amacrine cells. Likewise, repetitive iontopho…

Subtype specification of GABAergic amacrine cells by the orphan nuclear receptor Nr4a2/Nurr1.

In the mammalian retina, amacrine cells (ACs) contain numerous subtypes with extremely diverse morphologies and physiological functions. To date, how these subtypes arise during retinogenesis remains largely unknown at the molecular level. The orphan nuclear receptor Nr4a2 plays an essential role in specifying ventral midbrain dopaminergic neurons, and its mutations are associated with familial Parkinson's disease. Here we show that Nr4a2 is also critically involved in the specification of AC subtype identity. During mouse retinogenesis, Nr4a2 is expressed in a subset of postmitotic GABAergic …

Mechanism of generation of spontaneous miniature outward currents (SMOCs) in retinal amacrine cells.

A subtype of retinal amacrine cells displayed a distinctive array of K(+) currents. Spontaneous miniature outward currents (SMOCs) were observed in the narrow voltage range of -60 to -40 mV. Depolarizations above approximately -40 mV were associated with the disappearance of SMOCs and the appearance of transient (I(to)) and sustained (I(so)) outward K(+) currents. I(to) appeared at about -40 mV and its apparent magnitude was biphasic with voltage, whereas I(so) appeared near -30 mV and increased linearly. SMOCs, I(to), and a component of I(so) were Ca(2+) dependent. SMOCs were spike shaped, oc…

Developmental switch of GABAergic signaling in starburst amacrine cells driven by chloride transporter dynamics.

During early postnatal development, γ-aminobutyric acid (GABA) signaling undergoes a functional switch from excitation to inhibition, driven by age-dependent shifts in intracellular chloride concentrations ([Cl⁻]i). In the retina, starburst amacrine cells (SACs) are pivotal for establishing direction-selective circuitry. However, the molecular mechanisms and the precise timing of the reversal potential of GABA-induced currents (EGABA) shift in mouse SACs remain to be fully elucidated. We investigated the maturation of GABA responsiveness and chloride homeostasis in mouse SACs during the neonat…

Optogenetic targeting of AII amacrine cells restores retinal computations performed by the inner retina.

Most inherited retinal dystrophies display progressive photoreceptor cell degeneration leading to severe visual impairment. Optogenetic reac … t targets gene expression to a specific interneuron of the retina called the AII amacrine cell. AII amacrine cells simultaneously activate the ON pathway and inhibit the OFF pathway. We show that the optogenetic stimulation of AII amacrine cells allows restoration of both ON and OFF responses in the retina, but also mediates other types of retinal processing suc …

PTEN regulates starburst amacrine cell dendrite morphology during development.

Neurons are subject to extensive developmental regulation to ensure precise subtype-specific morphologies that are intimately tied to their function. Starburst amacrine cells (SACs) in the mammalian retina have a highly stereotyped, radially symmetric dendritic arbor that is essential for their role in direction-selective circuits in the retina. We show that PTEN, the primary negative regulator of the PI3K-AKT-mTOR pathway that is highly implicated in neurodevelopmental disorders, regulates SAC morphology in a cell-autonomous manner. Pten-deficient mouse SACs show a nearly twofold increase in …

Reduced Neuronal Self-Avoidance in Mouse Starburst Amacrine Cells With Only One Pcdhg Isoform.

The clustered protocadherins (cPcdhs) are a family of ∼60 homophilic cell adhesion molecules expressed across three gene clusters (Pcdha, Pcdhb, and Pcdhg) with a variety of essential roles in the developing nervous system. Some of these roles rely on specific isoforms, whereas others are more consistent with a model of isoform redundancy or a requirement for diversity. The γ-Pcdhs (expressed from the Pcdhg gene cluster) are particularly important for neuronal self-avoidance in starburst amacrine cells in the mouse retina. Here, we used mouse mutants to test two of the C-type isoforms-γC4 and …

Molecular identification of wide-field amacrine cells in mouse retina that encode stimulus orientation.

Visual information processing is sculpted by a diverse group of inhibitory interneurons in the retina called amacrine cells. Yet, for most of the >60 amacrine cell types, molecular identities and specialized functional attributes remain elusive. Here, we developed an intersectional genetic strategy to target a group of wide-field amacrine cells (WACs) in mouse retina that co-express the transcription factor Bhlhe22 and the Kappa Opioid Receptor (KOR; B/K WACs). B/K WACs feature straight, unbranched dendrites spanning over 0.5 mm (∼15° visual angle) and produce non-spiking responses to either l…

We conclude that the acetylcholine-synthesizing neurons of the ganglion cell layer are displaced amacrine cells. When DAPI was injected intraocularly instead of being applied to the optic tract, a regular mosaic of neurons in the ganglion cell layer was selectively stained, and two bands of fluorescence were observed in the inner plexiform layer, at the level …
