The ethics of animal experimentation in the field of shock and trauma research.

This is a personal review of the concepts used in deciding ethical questions as to the use of animals in research in the field of shock and trauma. There are for me ethical considerations since I feel animals can and should be used experimentally to advance the cause of the alleviation of pain and suffering amongst severely injured patients. The framework within which such decisions are to be made has been provided for us in Sweden by laws enacted some years ago. Even with ethical review by specially appointed, regional Ethical Committees there still remain problems for the individual research

Guidelines to Apply for Ethical Approval of Animal Experimentation and Use of Human Samples.

Animal experimentation is a fundamental tool in biomedical research for the study of diseases and the development of interventions such as drugs, vaccines, and probiotics for the treatment of human pathologies. Animal models enable analysis of pathophysiological processes in complex organisms with genetic and functional similarities to humans, which is essential for evaluating the safety and efficacy of new therapeutic interventions and surgical techniques before their clinical application. Among the most commonly used models are mice, rats, and fish, employed in the study of various diseases,…

[Clinical study on uterine cervix ripening with hyaluronidase and animal experimentation on mechanism of action].

The clinical course of labor in 53 cases with cervical injection of hyaluronidase (HD) and 51 cases with intravenous infusion of oxytocin to ripen the cervix was studied. The success rate of HD group (92.5%) was significantly higher than that of control group (62.8%). HD was without adverse side effects, and did not induce uterine contraction and therefore was preferable for high risk pregnancy cases. This method needed less time for cervical priming than others. The drug is easily available, not expensive and the method is simple. The animal experiments showed the mechanism of action of cervi

[The risk of cardiac arrest during laparoscopy. A study of 50,000 laparoscopies and animal experimentation].

In a French national enquiry into 50,000 laparoscopies it emerges that the number of cardiac arrests works out at 1 in 2,000. The percentage of reversible cardiac arrests was 1 in 2500 and of irreversible cardiac arrests 1.2 in 10,000. 25 cases were studied in great detail, both clinically and physiopathologically. This was thanks to a very full questionnaire most kindly filled up by those who had told us that this type of accident had happened in their practice. The physiopathogenesis of circulatory arrests during laparoscopy is complex. It appears however that among the different potentiatin

Assessment of cutaneous and ocular irritancy: a decade of research on alternatives to animal experimentation.

Over the past decade increasing societal and scientific pressure has promoted the development of alternatives to local tolerance testing in laboratory animals. The use of isolated organs, fertilized hen's eggs, and cell culture systems has been proposed as well as the study of a toxicant's biochemical effects and structure-activity relationships. This paper critically reviews the current status of these approaches and discusses the preliminaries for the establishment of alternative methods in the process of safety assessment.

Alternative to animal experimentation in pharmacology teaching: Development and validation of an equivalent digital learning tool.

Regarding animal experiments in pharmacology teaching, ethical considerations led us to examine an alternative approach to the use of living animals. This study aimed to assess whether digital tools could replace live animal experiments in terms of motivation and knowledge acquisition. The study was carried out with students enrolled in the 5th year of the industry/research stream at the Faculty of Pharmacy of the University of Limoges. The participants were randomly assigned to groups of traditional or digital teaching methods, with the common theme of the class being the effect of a diuretic

A simple and rapid assay to evaluate purity of foot-and-mouth disease vaccine before animal experimentation.

Currently, foot-and-mouth disease (FMD) vaccine purity is tested in cattle to detect antibodies against the non-structural protein (NSP) after repeated immunization with the final vaccine product. In case of vaccine failure, the manufacturing company would suffer significant economic loss. To prevent such unfortunate losses with the final vaccine product, in vitro testing is required to quantitate an NSP antigen during the manufacturing process prior to animal experiments. A novel lateral-flow assay device was developed using a monoclonal antibody (MAb) against the 3B NSP. To determine the min

Development and Characterisation of a Human Chronic Skin Wound Cell Line-Towards an Alternative for Animal Experimentation.

Background: Chronic skin wounds are a growing financial burden for healthcare providers, causing discomfort/immobility to patients. Whilst animal chronic wound models have been developed to allow for mechanistic studies and to develop/test potential therapies, such systems are not good representations of the human chronic wound state. As an alternative, human chronic wound fibroblasts (CWFs) have permitted an insight into the dysfunctional cellular mechanisms that are associated with these wounds. However, such cells strains have a limited replicative lifespan and therefore a limited reproduci

[Cortisone-induced osteonecrosis: knowledge acquired from observations in man and comparison with the results of animal experimentation].

Aseptic osteonecrosis is observed in 25% of cases after renal transplantation. This etiological variety of osteonecrosis is unusual in that it is frequently bilateral and has multiple localisations. Apart from the classical radiological signs, attention in attracted by isolated images of osteocondensation in the metaphyses and/or the diaphyses suggesting massive bony infarction and the appearances of fatigue fractures observed frequently (14%). In 43% of cases, the obvious necrosis was proceeded by early bone pain, around the 7 th day, during massive administration of corticosteroids in the pr

Learning dispositions as a lens for teaching socioscientific issues: Attitudes of university biology students towards animal experimentation.

Attitudes towards socioscientific issues (SSIs) represent crucial learning dispositions, as they can influence the students' willingness to engage with SSIs. Animal experimentation is a particularly contentious SSI, with attitudes ranging from rejection to acceptance. They are influenced by external factors (e.g., research context, test animal) and internal factors (e.g., knowledge, ethical aspects). Understanding these attitudes is essential for fostering reflective engagement with SSIs in higher education. This study (N = 503) examines self-reported attitudes of pre-service teacher biology s…

[Primates as experimental animals].

The term "primates" comprises a varied group of animals, consisting of more than 250 different species. The close evolutionary relationship to man resulted in the use of nonhuman primates as subjects of study for scientists from different research fields. In biomedical research the use of primates is restricted to questions which cannot be answered by animals with less developed physiological senses. Primates play an important role in infectious disease research, as many pathogens relevant to humans can be transferred to selected primate species. In the last few decades this applied especially

[Percutaneous artificial heart valves: from animal experimentation to the first human implantation in a case of calcified aortic stenosis].

The development of artificial cardiac valves capable of being positioned by catheter has become an important subject for research, with the objective of treating valvular patients who are not operable or at very high surgical risk. We tested an artificial valve implantable by the percutaneous route, consisting of three leaflets of bovine pericardium sutured to the inside of a stainless steel stent, deployable by inflating a balloon. Following laboratory evaluation, this valve was implanted with success in animals, then for the first time in man, in a case of calcified aortic stenosis. The pati

The physiology of experimental overfeeding in animals.

BACKGROUND: Body weight is defended by strong homeostatic forces. Several of the key biological mechanisms that counteract weight loss have been unraveled over the last decades. In contrast, the mechanisms that protect body weight and fat mass from becoming too high remain largely unknown. Understanding this aspect of energy balance regulation holds great promise for curbing the obesity epidemic. Decoding the physiological and molecular pathways that defend against weight gain can be achieved by an intervention referred to as 'experimental overfeeding'. SCOPE OF THE REVIEW: In this review, we

Experimental animal models of migraine.

Animal models of migraine have been widely used during the last decades to provide clues for understanding mechanisms underlying pathophysiology of migraine attacks and for developing specific therapeutic agents. They can be grouped into two main types: vascular and neurovascular. Trigemino-vascular system (TVS) is the most relevant efferent component and the mediators of its activity have been thoroughly studied along with some of the receptors involved to characterize anatomical and functional aspects of the system and to test efficacy and mechanisms of therapeutic agents. Neurovascular mode

Developmental asymmetries in experimental animals.

The early embryo orients to the antero-posterior axis and differentiates along this, and the dorso-ventral and lateral axes. From Drosophila melanogaster, detailed knowledge has accrued of how segmentation and dorso-ventral differentiation proceed, and of their genic control, mostly by selector and homeobox (Hox) genes. The study of the control of lateral differentiation, instead, has been largely neglected. Yet handed asymmetry (the "obvious" asymmetries of, for example, heart, lung, anatomical features of the nervous system, etc.) is basic and, possibly, universal. In the mouse, two genes co

Carcinogenicity of ochratoxin A in experimental animals.

The carcinogenicity of ochratoxin A, a naturally occurring mycotoxin of the fungal genera Aspergillus and Penicillium, was evaluated in three strains of mice and in one strain of rats. The kidney, and in particular the tubular epithelial cells, was the major target organ for ochratoxin A-induced lesions. In male ddY and DDD mice, atypical hyperplasia, cystadenomas and carcinomas of the renal tubular cells were induced, as were neoplastic nodules and hepatocyte tumours of the liver. In B6C3F1 mice, tubular-cell adenomas and carcinomas of the kidneys were induced in male mice, and the incidences
