Use of urine samples from healthy humans, nephritis patients or other animals as an alternative to foetal calf serum in the culture of Leishmania (L.) donovani in vitro.

The effect of supplementing in-vitro cultures of Leishmania donovani with urine was investigated. The parasites were isolated from Bangladeshi patients with visceral leishmaniasis. The urine samples used were collected from healthy human donors, patients with nephrotic syndrome, diabetic nephritis (DN) or diabetes mellitus, a dog and a cow. Promastigotes from blood-agar cultures were inoculated into RPMI-1640 basal medium with 10% heat-inactivated foetal calf serum (FCS) and/or 1%-20% urine. The parasites were then counted in a haemocytometer, on days 2, 4, 5, 6, 7, 8, 10, 12 and 14 post-inocu

Replacing immunisation-dependent antibody reagent production: Challenges under the UK Replacing Animals in Science 2025 strategy.

The UK's 2025 Replacing Animals in Science strategy introduces a commitment to end polyclonal antibody reagent production by 2030. While this represents a significant milestone, focusing exclusively on polyclonal reagents raises scientific, ethical and implementation challenges, given that monoclonal and other recombinant antibody formats frequently remain dependent on animal immunisation. This study examines whether current UK licensing practice and reporting frameworks are positioned to deliver genuine replacement. A structured, Three Rs-aligned replacement framework is articulated, reflecti

Scandinavia and the replacement of in vivo toxicity tests: Some personal reflections. The 2015 Bjorn Ekwall Memorial Award Lecture.

A personal, and therefore unavoidably biased, review is given, of the significance of the contributions made by selected Scandinavian individuals, organisations and events, to the development of in vitro toxicology procedures as potential replacements for toxicity tests in laboratory animals. In addition to their wider significance, these contributions had a profound effect on whatever contributions I have been able to make, myself. Nevertheless, while there has been much progress in the last 35 years or so, and many lessons have been learned, there is still much to be done, especially as anim

Aligning with the 3Rs: alternative models for research into muscle development and inherited myopathies.

Inherited and acquired muscle diseases are an important cause of morbidity and mortality in human medical and veterinary patients. Researchers use models to study skeletal muscle development and pathology, improve our understanding of disease pathogenesis and explore new treatment options. Experiments on laboratory animals, including murine and canine models, have led to huge advances in congenital myopathy and muscular dystrophy research that have translated into clinical treatment trials in human patients with these debilitating and often fatal conditions. Whilst animal experimentation has e

Alternatives to antibiotics for sustainable livestock production in the context of the One Health approach: tackling a common foe.

The discovery of the growth promoting effects of antibiotics in the 1940s contributed to the economic efficacy of the livestock industry. In response to increased animal protein demand from the 1950s, antimicrobial use at sub-therapeutic levels for growth promotion and disease prevention (antimicrobial growth promoter, AGP) improved feed-to-weight ratio, meat quality and overall health of livestock. These benefits encouraged the heavy use of AGPs such that about 70% of global use of antimicrobials was for food animals. Despite the numerous benefits of AGPs, the emergence of antimicrobial resis

Comparative costs of the Mouse Inoculation Test (MIT) and Virus Isolation in Cell Culture (VICC) for use in rabies diagnosis in Brazil.

The decision to use laboratory animals rather than in vitro methods is frequently based on the financial costs involved, so the objective of our study was to compare the costs of performing the Mouse Inoculation Test (MIT) and Virus Isolation in Cell Culture (VICC) for use in rabies diagnosis in Brazil. Based on observations of laboratory routines at the Pasteur Institute, São Paulo, we listed the fixed cost (FC) and variable cost (VC) items necessary to perform both tests. Considering that 200 MITs are equivalent to 350 VICC assays, in terms of facilities and staff-hours needed per month, we

Joint positive control testing in guinea pig skin sensitization tests. A harmonized approach.

A scheme for the performance of positive control studies within a coordinated group of laboratories was proposed (joint positive control testing). The procedure has been described, as well as the first results of the validation phase of this joint positive control testing project. Adoption of this proposal within the participating six laboratories would lead to a reduction in the number of guinea pigs required for reliability and sensitivity checks from current approximate 12 studies per year down to 2 studies, i.e., 150-300 fewer animals per year. Another benefit would be the use of a harmoni

Environmental impacts of alternatives to animal-source foods: a scoping review.

Drawing on a comprehensive systematic search of the literature, this scoping Review examined 125 studies on the environmental impacts of alternatives to animal-source foods (Alt-ASFs) compared with conventional animal-source foods. Alt-ASFs generally required fewer natural resources and produced lower greenhouse gas emissions than animal-source foods, with the most consistent advantages observed for plant-based foods. Environmental impacts of insect-based, fungi-based, algae-based, and cell-based alternatives were more variable than plant-based alternatives, reflecting differences in productio

Innovative drugs, chemicals, and enzymes within the animal production chain.

The alarming number of recently reported human illnesses with bacterial infections resistant to multiple antibacterial agents has become a serious concern in recent years. This phenomenon is a core challenge for both the medical and animal health communities, since the use of antibiotics has formed the cornerstone of modern medicine for treating bacterial infections. The empirical benefits of using antibiotics to address animal health issues in animal agriculture (using therapeutic doses) and increasing the overall productivity of animals (using sub-therapeutic doses) are well established. The

Alternatives to antibiotics in poultry feed: molecular perspectives.

The discovery of the growth promoting property of antibiotics led to their use as antibiotic feed additives (AFAs) in animal feed at sub-therapeutic doses. Although this has been beneficial for animal health and productivity, it has been, essentially, a double-edged sword. The continued and non-judicious use of AFAs has led to the selection and dissemination of antibiotic-resistant strains of poultry pathogens such as Salmonella, Campylobacter and Escherichia coli. The rapid spread of drug-resistant pathogens as well as emergence of antibiotic-related environmental pollutants is of global conc

Human organoids for Risk Group 4 virus research: a new frontier in investigating Nipah virus infection of the central nervous system.

UNLABELLED: New approach methodologies, such as high-complexity in vitro systems, are increasingly prioritized in biomedical research as potential alternatives to animal experimentation. We show that cerebral organoids derived from human induced pluripotent stem cells can be leveraged to (i) investigate isolate-specific replication dynamics of Nipah virus and (ii) model key histopathological lesions found in the brain tissue of infected human patients. Furthermore, we discuss the importance of organoid models for the study of Risk Group 4 viruses. IMPORTANCE: Advanced development of medical c

Potency evaluation of rabies vaccine for human use: The impact of reducing the number of animals per dilution - Part 2.

The most critical parameter for the quality control of the rabies vaccine is potency, which is evaluated by challenge test in mice while using a large animal number. Because the 3Rs concept is applied worldwide, it becomes necessary to develop alternative methods to demonstrate the production consistency of these vaccines and reduce the number of animals used for performing assays. Hence, the present study evaluated the impacts of reducing the number of mice used in the NIH test for such vaccines. A retrospective data analysis compared vaccines tested in the standard test with the results of t

Indications for the use of highest priority critically important antimicrobials in the veterinary sector.

BACKGROUND: Among the measures taken to preserve the clinical efficacy of highest priority critically important antimicrobials (HP-CIAs), the WHO has recommended avoiding their use in food-producing animals. Little is known regarding the indications for which different antimicrobial classes are used in animals, even in countries where data on antimicrobial use are available. OBJECTIVES: To outline, in a narrative review, the diseases for which HP-CIAs are used in veterinary medicine, highlighting incongruences with international guidelines and disease conditions where effective alternatives t

Do we still need animals? Surveying the role of animal-free models in Alzheimer's and Parkinson's disease research.

The use of animals in neuroscience and biomedical research remains controversial. Policy is built around the "3R" principle of "Refining, Reducing and Replacing" animal experiments, and across the globe, different initiatives stimulate the use of animal-free methods. Based on an extensive literature screen to map the development and adoption of animal-free methods in Alzheimer's and Parkinson's disease research, we find that at least two in three examined studies rely on animals or on animal-derived models. Among the animal-free studies, the relative contribution of innovative models that may

Alternatives for lung research: stuck between a rat and a hard place.

The respiratory system acts as a portal into the human body for airborne materials, which may gain access via the administration of medicines or inadvertently during inhalation of ambient air (e.g. air pollution). The burden of lung disease has been continuously increasing, to the point where it now represents a major cause of human morbidity and mortality worldwide. In the UK, more people die from respiratory disease than from coronary heart disease or non-respiratory cancer. For this reason alone, gaining an understanding of mechanisms of human lung biology, especially in injury and repair e

3Rs principles for biomedical research in Africa: Evidence, challenges, early career researchers' perspectives, and pathways for One Health research governance.

BACKGROUND: The 3Rs principles (Replacement, Reduction, and Refinement) are globally recognized as a cornerstone of ethical animal research and an essential component of responsible biomedical research systems. Nonetheless, evidence on the awareness, reporting, and implementation in African research settings remain limited. This study therefore examined integration of 3Rs principles in African biomedical research by assessing existing evidence, key challenges, and pathways forward. It also explored awareness of early career researchers concerning ethical animal use and the 3Rs principles, with

Use of genetically modified mouse models for evaluation of carcinogenic risk: considerations for the laboratory animal scientist.

There has been increasing interest in the use of selected genetically modified (GM) mouse models for the testing of chemicals to determine their carcinogenic potential. GM mouse models are believed to be useful tools that offer mechanistically relevant insights for understanding and predicting the human response to chemical exposure. They have been proposed as alternatives to the traditional 2-year mouse oncogenicity bioassay. In this overview we will review the GM mouse models that have been proposed as bioassay alternatives and present some of the key laboratory animal science challenges tha

Unveiling the Impact of Antibiotics and Alternative Methods for Animal Husbandry: A Review.

Since the 1950s, antibiotics have been used in the field of animal husbandry for growth promotion, therapy and disease prophylaxis. It is estimated that up to 80% of the antibiotics produced by the pharmaceutical industries are used in food production. Most of the antibiotics are used as feed additives at sub-therapeutic levels to promote growth. However, studies show the indiscriminate use of antibiotics has led to the emergence of multidrug-resistant pathogens that threaten both animal health and human health, including vancomycin-resistant Enterococcus (VRE), Methicillin-resistant Staphyloc

The classification of motor neuron defects in the zebrafish embryo toxicity test (ZFET) as an animal alternative approach to assess developmental neurotoxicity.

Rodents are widely used to test the developmental neurotoxicity potential of chemical substances. The regulatory test procedures are elaborate and the requirement of numerous animals is ethically disputable. Therefore, non-animal alternatives are highly desirable, but appropriate test systems that meet regulatory demands are not yet available. Hence, we have developed a new developmental neurotoxicity assay based on specific whole-mount immunostainings of primary and secondary motor neurons (using the monoclonal antibodies znp1 and zn8) in zebrafish embryos. By classifying the motor neuron def
