BACKGROUND: The Anopheles gambiae and Anopheles funestus mosquito species complexes are the primary vectors of Plasmodium falciparum malaria in sub-Saharan Africa. To better understand the environmental factors influencing these species, the abundance, distribution and transmission data from a south-eastern Kenyan study were retrospectively analysed, and the climate, vegetation and elevation data in key locations compared.
METHODS: Thirty villages in Malindi, Kilifi and Kwale Districts with data on An. gambiae sensu strict, Anopheles arabiensis and An. funestus entomological inoculation rates…

BACKGROUND: Knockdown resistance (kdr) is one of the primary resistance mechanisms present in anopheline species. Although this mutation is largely spread across the Anopheles gambiae s.l. members, its prevalence in other species is still not well documented.
METHODS: The present study investigated the distribution and allelic frequencies of kdr in An. gambiae s.l., An. pharoensis, and An. ziemanni samples collected in 2022 and 2023 in nine sites spread across five ecogeographical settings in Cameroon. Members of the An. gambiae complex were identified molecularly by polymerase chain reaction…

Despite previous decline of malaria in Ethiopia, an outbreak in Dire Dawa in 2022 implicated invasive vector An. stephensi as responsible. The transmission of Plasmodium by invasive An. stephensi raises questions about the molecular basis of compatibility, and the origin of the Plasmodium being transmitted. The Plasmodium P47 gene is a parasite-vector interaction gene in Anopheles , and along with corresponding mosquito P47 receptor ( P47Rec ), can be critical in establishment of Plasmodium infections in anophelines. Here, we analyzed P47 and P47Rec sequences to determine the origin of Plasmod…

BACKGROUND: Susceptibility of principal Anopheles malaria vectors to common insecticides was monitored over a 5-year period across Malawi to inform and guide the national malaria control programme.
METHODS: Adult blood-fed Anopheles spp. and larvae were collected from multiple sites in sixteen districts across the country between 2011 and 2015. First generation (F1) progeny aged 2-5 days old were tested for susceptibility, using standard WHO procedures, against pyrethroids (permethrin and deltamethrin), carbamates (bendiocarb and propoxur), organophosphates (malathion and pirimiphos-methyl) a…

BACKGROUND: The Anopheles gambiae sensu lato (s.l.) and Anopheles funestus s.l. species complexes contain the most important malaria vectors in Africa. Within the An. funestus group of at least 11 African species, the vector status of all but the nominal species An. funestus appears poorly investigated, although evidence exists that Anopheles rivulorum and Anopheles vaneedeni may play minor roles. A new species, An. rivulorum-like, was described from Burkina Faso in 2000 and subsequently also found in Cameroon and Zambia. This is the first paper reporting the presence of this species in South …

Mating is a vital behavior for mosquito reproduction, yet it remains poorly understood under captive conditions. We examined the copulation dynamics of 2 key malaria vectors, Anopheles funestus sensu stricto Giles and Anopheles arabiensis Patton, under laboratory settings in Tanzania. We conducted observations in 24-h cycles and monitored copulation events and insemination of females, initially using flashlights for nighttime visibility, followed by red lights in subsequent experiments. We observed how variations in mosquito age and artificial lighting influenced mating success for these 2 mos…

Using high-depth whole genome sequencing of F0 mating pairs and multiple individual F1 offspring, we estimated the nuclear mutation rate per generation in the malaria vectors Anopheles coluzzii and Anopheles stephensi by detecting de novo genetic mutations. A purpose-built computer program was employed to filter actual mutations from a deep background of superficially similar artifacts resulting from read misalignment. Performance of filtering parameters was determined using software-simulated mutations, and the resulting estimate of false negative rate was used to correct final mutation rate …

Three strains of Japanese encephalitis (JE) virus were recorded from mosquitoes collected in Lombok, Indonesia, during March 1979, from pools of Anopheles vagus, An. annularis and Culex tritaeniorhynchus respectively. This is believed to be the first report of isolation of JE virus from An. vagus. The frequencies of JE viral infection in zoophilic Anopheles species were higher than in Cx tritaeniorhynchus, the principle vector of JE virus in Asia. The low frequency of infection in Cx tritaeniorhynchus and the relatively infrequent raising of pigs may account for the low prevalence of JE neutra…

Anopheles mosquitoes serve as a significant vector for the transmission of malaria and arboviruses holding great medical importance, but their phylogenetic relationships have not yet been clearly elucidated. This study was the first to sequence and analyze the mitochondrial genomic features of An. hyrcanus and An. messeae, and to incorporate them into a phylogenetic analysis with 102 other species within the genus Anopheles. Both genomes contain 37 genes, including 22 tRNAs, 13 protein-coding genes and 2 rRNAs, as well as one control region. The genome characteristics, codon usage, and tRNA se…

Malaria, both Plasmodium falciparum (Welch) and Plasmodium vivax (Grassi & Feletti), has reemerged as a significant public health disease issue in Peru, especially in forested areas in the eastern part of the country. The spread of Anopheles darlingi Root, the principal South American malaria vector, into new areas of Peru is thought to be a factor in this resurgence. However, epidemiological evidence suggests that in malaria endemic areas of eastern Peru where An. darlingi does not occur, other species are involved in malaria transmission. The objective of this study was to analyze Anopheles …

BACKGROUND: Nchelenge District in northern Zambia suffers from holoendemic malaria transmission despite a decade of yearly indoor residual spraying (IRS) and insecticide-treated net (ITN) distributions. One hypothesis for this lack of impact is that some vectors in the area may forage in the early evening or outdoors. Anopheles gibbinsi specimens were identified in early evening mosquito collections performed in this study area, and further insight was gleaned into this taxon, including characterizing its genetic identity, feeding preferences, and potential role as a malaria vector.

Malaria transmission by Anopheles subpictus Grassi, 1899 and Anopheles culicifacies Giles, 1901 was studied from March 2004 to February 2007 in Angul District, Orissa, India, which is highly endemic for malaria. Adult mosquitoes were collected from human dwellings using sucking tubes and a mechanical aspirator. After identification, some An. subpictus and An. culicifacies specimens were subjected to a precipitin test to determine their anthropophilic index and the remaining samples were preserved in isopropyl alcohol for sporozoite detection by nested PCR. An. subpictus was the most prevalent …

BACKGROUND: The emergence of resistance to synthetic (chemical) insecticides along with their harmful effects on human health, non-target organisms and the environment necessitates the development of new complementary bioinsecticides that are effective, environmentally friendly, biodegradable and target-specific. This study was undertaken to evaluate larvicidal activities of 80% methanol and n-hexane extracts of four plants that are traditionally used by communities against mosquitoes.
METHODS: The dried plant parts of Ocimum lamiifolium, Amaranthus hybridus, Premna schimperi, and Lepidium sa

BACKGROUND: The colonization of mosquitoes susceptible to Plasmodium vivax via direct membrane feeding assay (DMFA) has the potential to significantly advance our knowledge of P. vivax biology, vector-parasite interaction and transmission-blocking vaccine research. Anopheles darlingi and Anopheles deaneorum are important vectors of malaria in the Western Brazilian Amazon. Since 2018, well-established colonies of these species have been maintained in order to mass produce mosquitoes destined for P. vivax infection. Plasmodium susceptibility was confirmed when the colonies were established, but …
