Nutritional strategies for autophagy activation and health consequences of autophagy impairment.

OBJECTIVES: Currently, the plague of chronic diseases, such as overweight, obesity, diabetes, and cardiovascular diseases, is associated with chronic inflammation as an effect of homeostasis disbalance. One of the processes involved in homeostasis maintenance is autophagy, which is also referred to as self-eating or cellular recycling. Due to the correlation between the epidemic scale of chronic diseases and autophagy impairment, strategies for autophagy activation are urgently needed. METHODS: In this review, we comprehensively summarized the current data on autophagy types, dysfunction, ass

Multiple Facets of Autophagy and the Emerging Role of Alkylphosphocholines as Autophagy Modulators.

Autophagy is a highly conserved multistep process and functions as passage for degrading and recycling protein aggregates and defective organelles in eukaryotic cells. Based on the nature of these materials, their size and degradation rate, four types of autophagy have been described, i.e. chaperone mediated autophagy, microautophagy, macroautophagy, and selective autophagy. One of the major regulators of this process is mTOR, which inhibits the downstream pathway of autophagy following the activation of its complex 1 (mTORC1). Alkylphosphocholine (APC) derivatives represent a novel class of a

Apicomplexan autophagy and modulation of autophagy in parasite-infected host cells.

Apicomplexan parasites are responsible for a number of important human pathologies. Obviously, as Eukaryotes they share a number of cellular features and pathways with their respective host cells. One of them is autophagy, a process involved in the degradation of the cell's own components. These intracellular parasites nonetheless seem to present a number of original features compared to their very evolutionarily distant host cells. In mammals and other metazoans, autophagy has been identified as an important contributor to the defence against microbial pathogens. Thus, host autophagy also lik

Chaperone-mediated autophagy.

Chaperone-mediated autophagy (CMA) is the only type of autophagy in mammalian cells able to selectively degrade cytosolic proteins in lysosomes. CMA is maximally activated in response to stressors such as prolonged starvation, exposure to toxic compounds, or oxidative stress. We have found that CMA activity decreases in aging and in some age-related disorders such as Parkinson's disease. Impaired CMA under these conditions may be responsible for the accumulation of damaged proteins inside cells and for their higher vulnerability to stressors. In contrast to other forms of autophagy, where subs

Selective autophagy: xenophagy.

Xenophagy is an autophagic phenomenon that specifically involves pathogens and other non-host entities. Although the understanding of the relationship between autophagosomes and invading organisms has grown significantly in the past decade, the exact steps to confirm xenophagy has been not been thoroughly defined. Here we describe a methodical approach to confirming autophagy, its interaction with bacterial invasion, as well as the specific type of autophagic formation (i.e. autophagosome, autolysosome, phagolysosome). Further, we argue that xenophagy is not limited to pathogen interaction wit

Function of Atg11 in non-selective autophagy and selective autophagy of Candida albicans.

Candida albicans is an important opportunistic pathogenic fungus in the human body. It is a common microbe inhabiting on the mucosa surfaces of healthy individuals, but may cause infections when the host immune system is weak. Autophagy is a "self-eating" process in eukaryotes, which can recover and utilize damaged organelles and misfolded proteins. Here we investigated the role of the autophagy-related protein Atg11 in C. albicans. Deletion of ATG11 led to the defect in growth under the nitrogen starvation condition. Western blotting and GFP localization further revealed that the transport an

Spatial autophagy failure as a pharmacodynamic endpoint for autophagy-targeted dermatological interventions.

Autophagy-modulating dermatological interventions include topical, intralesional and systemic therapies, defined bioactive molecules and nutraceutical candidates. Their effects are commonly evaluated using tissue-averaged LC3-II, p62/SQSTM1 and canonical pathway markers. Although these measures support assessment of autophagy pathway engagement, they may miss spatially restricted pharmacodynamic non-response and cannot determine whether persistent local dysfunction reflects inadequate exposure or biology-limited non-response. Here we propose spatial autophagy failure (SAF) as a spatial pharmac

ECM-regulation of autophagy: The yin and the yang of autophagy during wound healing.

Wound healing is a complex sequence of tissue protection, replacement, and reorganization leading to regenerated tissue. Disruption of any of these steps results in the process being incomplete as an ulcer or over-exuberant as a hypertrophic scar. Over the past decade, it has become evident that the extracellular matrix and associated components orchestrate this process. However, the cellular events that are induced by the extracellular matrix to accomplish wound healing remain to be defined. Herein we propose that matrix-regulated cellular macro-autophagy is key to both the tissue replacement

Autoimmunity, diet and autophagy.

Autophagy is a highly conserved lysosomal recycling pathway that couples nutrient availability to cellular quality control and immune regulation. Accumulating evidence identifies autophagy as a central mechanistic interface through which dietary exposures influence metabolic inflammation and the maintenance or breakdown of immune tolerance in autoimmune diseases. This review synthesizes current molecular and cellular insights into how nutrition modulates autoimmune susceptibility and disease activity by shaping autophagic pathways across immune and metabolic tissues. Dietary signals exert bidi

[Autophagy and Neurological Diseases].

Autophagy is an essential degradation mechanism that maintains intracellular homeostasis. In recent years, an increasing number of cases with mutations in autophagy-related genes, such as ATG7, have been reported. These findings highlight the crucial role of autophagy in human neurodevelopment. However, the severity of clinical symptoms does not always correlate with the degree of autophagy impairment observed in patient-derived cells, and phenotypic manifestations can vary widely. These findings indicate that autophagy dysfunction alone does not fully explain disease mechanisms, even in neuro

Autophagy in lipid metabolism.

Autophagy is a complex and highly regulated cellular process essential for maintaining homeostasis in adipose tissue by modulating adipocyte differentiation, lipogenesis, and lipolysis. Although lipids are primarily known as energy-storage compounds, they also play a role in triglyceride transport, intracellular communication via steroid hormones, organ protection, and thermoregulation. Lipid metabolism relies on the dynamic balance between synthesis, storage and degradation of key lipid classes such as: triglycerides, sterols and phospholipids. Among the pathways controlling lipid metabolism,

Synthetic Activators of Autophagy.

Autophagy is a central process for degradation of intracellular components that do not operate correctly. Molecular mechanisms underlying this process are extremely difficult to study, since they involve a large number of participants. The main task of autophagy is redistribution of cellular resources in response to environmental changes, such as starvation. Recent studies show that autophagy regulation could be the key to achieve healthy longevity, as well as to create therapeutic agents for treatment of neurodegenerative diseases such as Parkinson's and Alzheimer's diseases. Thus, developmen

Autophagy and bone diseases.

Autophagy is a ubiquitous cellular process, allowing the removal and recycling of damaged proteins and organelles. At the basal level, this process plays a role in quality control, thus participating in cellular homeostasis. Autophagy can also be induced by various stresses, such as nutrient deprivation or hypoxia, to allow the cell to survive until conditions improve. In recent years, the role of this process has been widely studied in many pathologies such as neurodegenerative diseases or cancers. In bone tissue, various studies have shown that autophagy is involved in the survival, differen

Autophagy, Hyperlipidemia, and Atherosclerosis.

Autophagy is an evolutionarily conserved process in eukaryotes that processes the turnover of intracellular substances. Atherosclerosis is a disease caused by multiple factors, it mainly occurs on the walls of large and medium blood vessels and atherosclerotic plaques form in the intima of the blood vessels. Hyperlipidemia is considered to be a very dangerous factor leading to cardiovascular and cerebrovascular diseases, especially atherosclerosis. This chapter mainly introduces the key role of autophagy in hyperlipidemia and atherosclerosis, that is, impaired lipophagy affects the degradation

Autophagy and Alzheimer's Disease.

Alzheimer's disease (AD) is the most common type of dementia and is characterized by progressive cognitive decline. Increasing evidence has demonstrated that the autophagic process plays an important role in AD. In this chapter, we will discuss the role of autophagy in the pathogenesis of AD and other types of dementia, including dementia with Lewy bodies (DLB), frontotemporal dementia (FTD), vascular dementia (VD) and prion diseases. In addition, we will discuss autophagy-targeted therapies as future treatments for AD.

Autophagy and mitophagy in ALS.

Amyotrophic lateral sclerosis (ALS) is a debilitating and incurable disease involving the loss of motor neurons and subsequent muscle atrophy. Genetic studies have implicated deficits in autophagy and/or mitophagy in the onset of the disease. Here we review recent progress in our understanding of the pathways for autophagy and mitophagy in neurons, and how these pathways may be affected by mutations in genes including DCTN1, OPTN, TBK1, VCP, and C9ORF72. We also discuss the implications of modulating autophagy in ALS, highlighting both the potential of the approach and the concerns raised by t

Autophagy in apicomplexan parasites.

Autophagy is a highly conserved eukaryotic degradation process that permits the recycling of intracellular components. The molecular machinery and the functions of autophagy have been classically characterized in mammalian cells and yeast, but have long remained unexplored in less-studied eukaryotes. Apicomplexan parasites are early-diverging eukaryotes responsible for a number of important human and veterinary diseases. In light of recent investigations into autophagy function in two of these pathogens, Plasmodium and Toxoplasma, it seems their autophagy-related machinery could be involved in
