Experiments on the transmission of Babesia major and Babesia bigemina by Haemaphysalis punctata.

Experiments on the transmission by Haemaphysalis punctata of three large Babesia strains were carried out. Three Babesia-free batches of laboratory reared H. punctata ticks were infected with two strains of Babesia major, B. major (Xingjiang strain), isolated with adult ticks of H punctata and B. major (Henan strain), isolated with H. longicornis) and a strain of Babesia bigemina by feeding them on the calves infected by inoculation of blood stabilates. H. punctata was shown to be capable of transmitting the B. major strains transovarially. The larvae, nymphs and adults developed from female t

Clinical observations on Babesia gibsoni and Babesia canis infections in dogs.

Thirty-five cases of Babesia gibsoni infection and 11 cases of Babesia canis infection were diagnosed and treated in dogs at Kadena Air Base in Okinawa, Japan, between April 1979 and February 1980. Diagnosis was made by demonstrating the organisms in blood smears and by serologic examination, using an indirect fluorescent antibody test. Serologic findings correlated well with the occurrence of infection in mature dogs, but poorly in young (1- to 3-month-old) dogs. Although these 2 intraerythrocytic parasites were readily distinguishable on Giemsa-stained blood smears and by the indirect fluore

Babesia conradae, sp. Nov., a small canine Babesia identified in California.

Small piroplasms as a cause of canine babesiosis have usually been identified as Babesia gibsoni. Recent genetic studies suggested that small piroplasms are more likely comprised of at least three genotypically distinct species. In southern California, canine babesiosis caused by a small piroplasm has been documented since 1990. Morphological characteristics of this parasite include a small (0.3-3.0 microm) intraerythrocytic merozoite stage with predominantly ring, piriform, tetrad, amoeboid, or anaplasmoid forms. Transmission electron microscopic images of merozoites demonstrate the presence

A short term in vitro cultivation of Babesia rodhaini and Babesia microti.

In vitro cultivation of Babesia rodhaini (BR) and Babesia microti (BM) was attempted. When RPMI1640 was supplemented with 30 or 40% of non-treated fetal bovine serum (FBS), the gas mixture of 3% CO2-8% O2 best supported the growth of both parasites. Under this optimized condition, the percent parasitized erythrocytes peaked to approximately 4- and 2-times initial values for Br and BM, respectively. The cultivated parasites retained the infectivity to the host mice. BM showed the characteristic feature of division during cultivation. However, the lots of FBS will have to be taken into considera

Babesia divergens vaccine.

A vaccine strategy against Babesia divergens bovine babesiosis was successfully developed after perfecting of an efficient in vitro culture. Crude supernatants and purified fractions were able to induce a vaccine protection in gerbils against B. divergens infection. More, supernatants induced an effective vaccine protection in cattle. The role of B. divergens exoantigens of 17, 37, 46, 70 and 90 kDa in the development of the immune response was clearly demonstrated in gerbils, cattle, and man.

Transfection of Babesia duncani: A Genetic Toolbox of this Pathogen to Advance Babesia Biology.

Human babesiosis is a tick-borne disease caused by Babesia pathogens. The disease, which presents with malaria-like symptoms, can be life-threatening, especially in individuals with weakened immune systems and the elderly. The worldwide prevalence of human babesiosis has been gradually rising, prompting alarm among public health experts. In other pathogens, genetic techniques have proven to be valuable tools for conducting functional studies to understand the importance of specific genes in development and pathogenesis as well as to validate novel cellular targets for drug discovery. Genetic m

Development of droplet digital PCR for the detection of Babesia microti and Babesia duncani.

Babesia spp. are obligate protozoan parasites of red blood cells. Transmission to humans occurs through bites from infected ticks or blood transfusion. Infections with B. microti account for the majority of the reported cases of human babesiosis in the USA. A lower incidence is caused by the more recently described species B. duncani. The current gold standard for detection of Babesia is microscopic examination of blood smears. Recent PCR-based assays, including real-time PCR, have been developed for B. microti. On the other hand, molecular assays that detect and distinguish between B. microti

Theileria (Babesia) equi and Babesia caballi infections in horses in Galicia, Spain.

The control of equine piroplasmosis is becoming increasingly important to maintain the international market open to the horse industry. The purpose of this study was to demonstrate the occurrence of equine piroplasmosis (Theileria equi and Babesia caballi) in Galicia, north-west Spain, and to compare haematological and serum biochemistry parameters between non-parasitaemic horses and horses parasitaemic with T. equi and B. caballi. Sixty serum samples (control group) were taken from healthy horses pastured on two farms, and examined for evidence of equine T. equi and B. caballi infection by in

Babesia divergens and Babesia microti species-specific genes that may drive pathogenesis.

Human babesiosis is an emerging disease as more cases are being reported worldwide. Most cases in Europe are caused by Babesia divergens, whereas most cases in North America are due to Babesia microti. While B. microti is also found throughout Europe, it appears to be less pathogenic. We generated high-quality nuclear and organellar genome assemblies of two B. divergens and two B. microti isolates from Germany by long-read sequencing and compared them with each other and with the reference RI B. microti from the US. Variants of VESA1 (variant erythrocyte surface antigen 1) and secreted antigen

Genetic characterization and molecular survey of Babesia bovis, Babesia bigemina and Babesia ovata in cattle, dairy cattle and yaks in China.

BACKGROUND: Babesiosis is an important haemoparasitic disease, caused by the infection and subsequent intra-erythrocytic multiplication of protozoa of the genus Babesia that impacts the livestock industry and animal health. The distribution, epidemiology and genetic characterization of B. bigemina, B. bovis, and B. ovata in cattle in China as well as the prevalence of these protozoan agents were assessed. METHODS: A total of 646 blood specimens from cattle, dairy cattle and yaks from 14 provinces were collected and tested for the presence of the three Babesia species via a specific nested PCR

A new PCR assay for the detection and differentiation of Babesia canis and Babesia vogeli.

Babesia spp. are globally distributed tick-borne protozoan parasites that infect the red blood cells of a wide range of vertebrate hosts, including humans. Diagnosis of babesiosis is often impeded by the transient presence of the parasites in peripheral blood, as well as by their pleomorphic nature. Given the reports of an expanding and, in some cases, sympatric geographical distribution of Babesia canis and Babesia vogeli in dogs and associated vectors, in Europe, the development of time-efficient and cost-effective diagnostic tools to detect and differentiate these two species is warranted.

Epidemiological survey of Babesia bovis and Babesia bigemina infections of cattle in Philippines.

A total of 250 blood samples were collected from clinically healthy cattle in five provinces of Philippines. DNA was extracted from the samples and analyzed by nested PCR assays for an epidemiological survey of Babesia bovis and Babesia bigemina infections. Out of the 250 samples, 27 (10.8%) and 16 (6.4%) were positive for B. bovis infection and B. bigemina infection, respectively. Mixed infections were detected in a total of 4 samples (1.6%). Our data provide baseline information regarding the epidemiology of B. bovis and B. bigemina infections in cattle in Philippines, which can be utilized

Babesia canis and Babesia rossi co-infection in an untraveled Nigerian dog.

A sexually intact 6-month-old female Alsatian dog was presented to the Veterinary Clinic of the National Veterinary Research Institute, Vom, Plateau State, Nigeria, for the following complaints: anorexia, hemoglobinuria, fever, tick infestation and general malaise. Microscopy revealed piroplasms with a wide range of sizes (1-5 μm in length) in red blood cells, raising a suspicion of a co-infection with two or more Babesia species. Specific PCR assays for canine Babesia spp. and DNA sequencing revealed the presence of Babesia canis and Babesia rossi co-infection. This study constitutes the firs

Detection of Babesia caballi and Babesia equi in Dermacentor nuttalli adult ticks.

Ticks play an important role in human and veterinary medicine particularly due to their ability to transmit protozoan pathogens. In this study we have demonstrated that polymerase chain reaction (PCR) and nested PCR methods enabled detection of Babesia caballi and Babesia equi in field isolates of Dermacentor nuttalli adult ticks from Mongolia. Primers specific for 218 bp fragment merozoite antigen 1 (EMA-1) gene of B. equi successfully amplified products from all samples of D. nuttalli adult ticks while primers for the 430 bp fragment product from BC48 gene of B. caballi amplified products fr
