[Yersinia pestis. Bacteriology].

The author discusses the evolution in the classification of the bacterium, responsible for plague: first a classification based on phenotypic characteristics, later based on genotypic characteristics, to finally arrive at an evolutionist classification. He treats the seven species of the genus Yersinia that can be distinguished by DNA hybridization. He examines the issue of sequencing and decoding the chromosome and mentions research regarding the phenomenon that the metabolism of the organisms modifies as a reaction to signals of their changing environment. Furthermore the author discusses th

Methods of outbreak investigation in the "era of bacteriology" 1880-1920.

The advent of bacteriological methods in the later 19th century has been seen, on the examples of America and Germany, to have been followed by a new laboratory-based, contact-tracing method of investigating outbreaks of epidemic disease. In Britain, however, this new approach never took firm root, and practising epidemiologists continued to follow an observational and deductive tradition in field investigations, rejecting any primary dependence on bacteriological methods. Alongside this persistent observational practice, there emerged a new statistical approach, based in Pearsonian biometrics

[Bacteriology of the ejaculate--a useful study?].

In a retrospective study we analyzed the results of 318 bacteriologic tests of ejaculates from 175 patients with suspected infections of the prostate, epididymis and urethra. Only 55% of all bacteriologic tests were positive, and the bacteria most frequently found were those normally present in the anterior male urethra, enterococci and coagulase-negative staphylococci. These results indicate that bacteriologic testing of the ejaculate does not yield such reliable results as the segmented urine culture technique and the examination of expressed prostatic secretions.

Use of a bronchoscopic protected catheter technique in the clinical evaluation of a new antibiotic.

We prospectively compared the usefulness of a bronchoscopic protected catheter technique with the results from sputum cultures in the evaluation of moxalactam, a new beta-lactam antibiotic. The significance of a given isolate on protected catheter culture was determined by quantitative bacteriology. 32 patients with community-acquired pneumonia were enrolled in the study and 31 grew common lower respiratory tract pathogens from their protected catheter specimen. The most common single pathogens recovered were Streptococcus pneumoniae (11 patients) and Haemophilus influenzae (2). Mixed flora, p

Detection of Streptococcus dysgalactiae subsp. equisimilis in equine nasopharyngeal swabs by PCR.

Streptococcus (S.) dysgalactiae subsp. equisimilis is responsible for severe diseases in humans, including primary bacteraemia, pneumonia, endocarditis, and toxic shock syndrome. Infection in some animal species can also occur, although a few studies have looked into cross-species infectivity. In horses, S. equisimilis is generally considered infrequent or opportunistic, but has recently been isolated from cases of strangles-like disease. Rapid and sensitive diagnostic techniques could enable epidemiological studies and effective investigation of outbreaks involving these bacteria. In this stu

[Modern laboratory diagnostics for mycobacteria].

Early diagnosis of tuberculosis is one of the most important tools for the interruption of transmission chains and the elimination of the disease. Even today the final diagnosis of tuberculosis has to be confirmed finally by the bacteriological detection of mycobacteria in sputa or other specimens (1 - 4). The suspicion of tuberculosis can be corroborated by chest X-ray and clinical symptoms. Immunologically based assays like the tuberculin skin-test and the new interferon-gamma-release-assays enable the diagnosis of tuberculosis infection, but cannot distinguish between active and latent tube

Comparative Evaluation of Enrichment and Selective Culture Strategies for Detecting Salmonella from Cockroaches.

Background: Cockroaches, including the German cockroach (Blattella germanica) can harbor and disseminate enteric human pathogens, such as Salmonella spp. Detecting Salmonella in field-collected cockroach samples is important for source attribution and monitoring public health risk, but is complicated by the low abundance of the pathogen relative to the rich and complex cockroach microbiota. Studies using different culture methods to detect Salmonella in cockroaches have reported disparate results, emphasizing the need for sensitive, accurate strategies to recover and identify Salmonella from i

[Blood stream infection and blood culture--"progress" and "blind" in blood culture testing].

We have investigated various types of blood culture bottles which are mainly used at present and posed problems present in the blood culture bottles. First, there are differences between resin and ecosorb in the ability to adsorb and inactivate antibiotics in the blood. Second, the delay in placing the bottle (into which blood was inoculated) to the automatic instrument (delay in the start of incubation) greatly affects the automatic detection by BACTEC system and shows false negatives. Third, when the same blood is incubated in plural bottles (aerobic and anaerobic bottles), the differences a

Use of the VITEK 2 system for rapid identification of clinical isolates of Staphylococci from bloodstream infections.

Staphylococci are an increasing cause of bloodstream infections. Rapid reliable identification of these organisms is essential for accurate diagnosis and prompt effective treatment. We evaluated the ability of the VITEK 2 system (bioMérieux, Inc, Hazelwood, Mo.) to identify these organisms rapidly and accurately. A total of 405 clinically relevant nonduplicate staphylococcal isolates (Staphylococcus aureus, n = 130; coagulase-negative staphylococci, n = 275) collected from blood cultures were tested. VITEK 2 results were considered correct when they were identical to those furnished by the com

Classification of the legionellae.

This article reviews the classification and identification of the 23 described species, the 11 unnamed species, and the 49 serogroups presently known in the family Legionellaceae and the genus Legionella. The events leading to the isolation and classification of the first species, Legionella pneumophila, are summarized; these include the outbreak of legionnaires' disease at the 1976 American Legion Convention in Philadelphia, the five outbreaks preceding the Philadelphia outbreak, and the isolation of three Legionella species before 1976. The phenotypic characteristics of legionellae are descr

Comparison of methods for isolation and enumeration of thermophilic actinomycetes from dust.

Thermophilic actinomycetes are the primary sensitizing agents in farmer's lung disease. We compared dilution pour-plate and spread-plate methods for their usefulness in enumerating thermophilic actinomycetes in moldy silage dust and evaluated the ability of a nonquantitative gravity settling technique to recover thermophilic actinomycetes from moldy silage. Spread plates and pour plates yielded similar estimates of total thermophiles. Higher counts were observed on spread plates (P less than 0.05) for Thermoactinomyces candidus, Micropolyspora faeni, and Saccharomonospora viridis. M. faeni and

[The rapid detection and specific identification of bacterial antigens by electro-immunodiffusion in 80 cases of purulent meningitis (author's transl)].

The cerebrospinal fluid of 120 subjects, 80 of whom were suffering from a purulent meningitis, were examined comparatively by classical bacteriological techniques (direct examination and culture) and by electro-immunodiffusion. This procedure makes it possible to detect not only viable bacteria, but also capsular polysaccharide antigens. It revealed the diagnosis in 96.9% of cases of meningococcal, pneumococcal and Haemophilus meningitis, as against 80.3% positive results using classical bacteriological techniques, for the same organisms. Electro-immunodiffusion provides a rapid answer (20 min…

Recent advances in the microbiological diagnosis of bloodstream infections.

Rapid identification (ID) and antimicrobial susceptibility testing (AST) of the causative agent(s) of bloodstream infections (BSIs) are essential for the prompt administration of an effective antimicrobial therapy, which can result in clinical and financial benefits. Immediately after blood sampling, empirical antimicrobial therapy, chosen on clinical and epidemiological data, is administered. When ID and AST results are available, the clinician decides whether to continue or streamline the antimicrobial therapy, based on the results of the in vitro antimicrobial susceptibility profile of the
